    Age Scale for Assessing Activities of Daily Living

    The purpose of this study was to develop an age scale for assessing activities of daily living (ADL) among community-dwelling adults aged 75 years or older. Participants were 1006 older Japanese: 312 men (79.6 ± 4.3 years) and 694 women, (79.9 ± 5.5 years). Participants completed a battery of 8 performance tests related to ADL and the Barthel index (BI) questionnaire. Spearman rank-order correlation analysis was applied to obtain the correlation of the 8 ADL performance tests with the total BI score. Three variables were high rank-order correlated with BI, secondly those items were subjected to the principal component analysis. The weighted combination of the principal component scores was summed. Resulting in an ADL score (ADLS), women = 0.075 X1 - 0.082 X2 - 0.063 X3 + 0.124, men = 0.051 X1 - 0.105 X2 - 0.099 X3 + 0.249, where X1 = hand-grip strength, X2 = timed up and go, X3 = five-chair sit to stand. Individual ADLS was transformed to an ADL age scale (ADLA). The estimation was - 5.493 ADLS + 79.90 for women, and - 4.272 ADLS + 79.57 for men. Due to the distortion at the regression edges, the equation was corrected as suggested by Dubina et al. ADLA women after correction was = 0.447 (chronological age: CA) - 5.49ADLS + 44.17, men = 0.519CA - 4.27ADLS + 38.26. ADLA can be used to identify or monitor the characteristics of the ADL levels of physical abilities in older Japanese aged 75 years or older

    Impact of deposition pressure and two-step growth technique on the photoresponsivity enhancement of polycrystalline BaSi2 films formed by sputtering

    We investigate the influence of deposition pressure in the range 0.25–1.0 Pa on the photoresponsivity of 200 nm thick BaSi2 films grown by sputtering at 600 °C. BaSi2 films formed at 0.8 Pa exhibit a high photoresponsivity. The deposited Ba-to-Si atomic ratio depends significantly on the sputtering pressure. That\u27s why the pressure influences the photoresponsivity. BaSi2 films grown by a two-step growth technique show much higher photoresponsivity almost equivalent to those grown by molecular beam epitaxy. The photoresponsivity reaches 0.75 A W−1 at 2.0 eV at a bias voltage of 0.5 V applied between the top and bottom electrode

    ユニバーサルデザインの理念を地域に根付かせてそれが具体的な実践につながるようにするにはどうすればいいだろうか。これまで何年かにわたって米国などでユニバーサルデザインを推進してきた専門家を招へいして講演会などを企画してきたが、平成20年度にはセンターフォアユニバーサルデザインでユニバーサルデザインの父と呼ばれたロナルド・メイス教授を支えてきたレスリー・ヤング女史にお話をお願いし、関係組織からの情報提供と合わせて有用な情報を得た。なお、本報告は平成20年度の文化・芸術研究センター長特 研究報告として 置づけられる。What can be done to implement the concept of universal design in the community, not just in words but in action? The authors have tried for the past several years to organize lecture sessions with invited speakers from the States and UK, and in 2008 the invited speaker was Ms Leslie Young, who has worked with late Professor Ronald Mace at the Center for Universal Design from its establishment.     Overexpression of CRKII increases migration and invasive potential in oral squamous cell carcinoma.

    CT10 regulator of kinase (CRK) was originally identified as an oncogene product of v-CRK in a CT10 chicken retrovirus system. Overexpression of CRKII has been reported in several human cancers. CRKII regulates cell migration, morphogenesis, invasion, phagocytosis, and survival; however, the underlying mechanisms are not well understood. In the present study, we evaluated the possibility of CRKII as an appropriate molecular target for cancer gene therapy. The expression of CRKII in 71 primary oral squamous cell carcinomas and 10 normal oral mucosal specimens was determined immunohistochemically, and the correlation of CRKII overexpression with clinicopathological factors was evaluated. Overexpression of CRKII was detected in 41 of 70 oral squamous cell carcinomas, the frequency being more significant than in normal oral mucosa. In addition, CRKII overexpression was more frequent in higher-grade cancers according to the T classification, N classification, and invasive pattern. Moreover, RNAi-mediated suppression of CRKII expression reduced the migration and invasion potential of an oral squamous cell carcinoma cell line, OSC20. Downregulation of CRKII expression also reduced the expression of Dock180, p130Cas, and Rac1, and the actin-associated scaffolding protein cortactin. These results indicate that the overexpression of CRKII is tightly associated with an aggressive phenotype of oral squamous cell carcinoma. Therefore, we propose that CRKII could be a potential molecular target of gene therapy by RNAi-targeting in oral squamous cell carcinoma

    Induction of Tertiary Lymphoid Structures With Antitumor Function by a Lymph Node-Derived Stromal Cell Line

    Tertiary lymphoid structures (TLSs) associate with better prognosis in certain cancer types, but their underlying formation and immunological benefit remain to be determined. We established a mouse model of TLSs to study their contribution to antitumor immunity. Because the stroma in lymph nodes (sLN) participates in architectural support, lymphogenesis, and lymphocyte recruitment, we hypothesized that TLSs can be created by sLN. We selected a sLN line with fibroblast morphology that expressed sLN surface markers and lymphoid chemokines. The subcutaneous injection of the sLN line successfully induced TLSs that attracted infiltration of host immune cell subsets. Injection of MC38 tumor lysate-pulsed dendritic cells activated TLS-residing lymphocytes to demonstrate specific cytotoxicity. The presence of TLSs suppressed MC38 tumor growth in vivo by improving antitumor activity of tumor-infiltrating lymphocytes with downregulated immune checkpoint proteins (PD-1 and Tim-3). Future engineering of sLN lines may allow for further enhancements of TLS functions and immune cell compositions
