    Leadership in rural congregations and communities: an exploration in the North-Eastern Free State

    Rural communities and congregations cannot simply be overlooked. Communities in the north-eastern Free State are typical rural towns with a strong emphasis on agricultural activities and less emphasis in terms of processing products and other activities. Over the past few decades, rural communities have experienced many changes in terms of depopulation, the decline of the rural economy, poverty, and unemployment. Congregations in rural communities and their leadership cannot escape these changes that affect their functioning and existence. The key research question is thus: What is the congregational leadership’s opinion and argument about leadership in both the congregation and the community? The research indicated that a shift is taking place as far as the minister’s role in the leadership of the congregation is concerned. The congregational leadership needs to develop a missional perspective that brings the wider community into the ministry of the congregation. Although there is an awareness in respect of the missional calling of the church and the congregation, the change is still limited and there is room for a greater and more deliberate involvement of the congregation and its leadership within the community

    Anaerobes and bacterial vaginosis in pregnancy: virulence factors contributing to vaginal colonisation

    The aetiology and pathogenesis of bacterial vaginosis (BV) is unclear but it appears to be associated with factors that disrupt the normal acidity of the vagina thus altering the equilibrium between the normal vaginal microbiota. BV has serious implications for female morbidity, including reports of pelvic inflammatory disease, adverse pregnancy outcomes, increased susceptibility to sexually transmitted infections and infertility. This paper reviewed new available information regarding possible factors contributing to the establishment of the BV vaginal biofilm, examined the proposed role of anaerobic microbial species recently detected by new culture-independent methods and discusses developments related to the effects of BV on human pregnancy. The literature search included Pubmed (NLM), LISTA (EBSCO), and Web of Science. Because of the complexity and diversity of population groups, diagnosis and methodology used, no meta-analysis was performed. Several anaerobic microbial species previously missed in the laboratory diagnosis of BV have been revealed while taking cognisance of newly proposed theories of infection, thereby improving our understanding and knowledge of the complex aetiology and pathogenesis of BV and its perceived role in adverse pregnancy outcomes.National Research Foundation of South Africa (NRF)Web of Scienc

    Sensory Evaluation of Pinked Sauvignon Blanc Wines

    Sensorial studies on pinked white wines have never been carried out. The aim of this study was to establishthe probability of an aroma and taste difference in pinked Sauvignon blanc wines using a triangular test.The issue of at which point a wine consumer can perceived the wine as pink was analysed using a rankingexperiment. The probability that a wine taster can detect oxidised and pink wines as a wine fault was alsoanalysed. It was found that the panellists could not detect anomaly samples by taste and aroma alone,although some noticed an oxidised aroma on the nose. The pink detection point was established at 0.03 AU.This point can be used to refine the detection point and the assay used. It was established that more than50% of the panellists could detect oxidised and pink wines as a wine fault. The study data can be used toeither train panellists to detect pinking as a wine fault, or to establish a potential new category for pinkSauvignon blanc wines

    Rediscovering ‘disciplemaking’ and the role of faith-sharing

    According to the Gospel of Matthew, disciplemaking seems to be the signature mark of faithful disciples of Jesus the Christ (cf. Mt 28:18–20). Van Aarde refers to this, with reference to Von Harnack and Lohmeyer as the manifesto of the church, being on the same level of meaning as Deuteronomy 6 in the Old Testament. It may be fair to say that this ‘natural’ way of being and doing was in more than one way exchanged for evangelism practices that did nothing to show forth that following the Christ is a better or best way of living life, here already, to its fullest. These practices even reflected negatively on disciplemaking as such. A personal conviction is that ‘discipling’ may even be one of the missing links in the so-called missional conversation. This article will reflect on current theory on discipling and the natural necessity thereof. It will also draw upon findings in empirical research conducted by the National Church Life Surveys during 2014 in South Africa that may give an indication whether discipling is a common praxis among selected South African denominations and congregations. The focus will be on ‘Faith-Sharing’ as a core quality among adult attenders and includes several measures of the involvement of attenders in the outreach of the congregation (e.g. evangelistic activities, readiness to share their faith with others and whether outreach is a highly valued part of the ministry of the congregation)

    Nanoparticles for Applications in Cellular Imaging

    In the following review we discuss several types of nanoparticles (such as TiO2, quantum dots, and gold nanoparticles) and their impact on the ability to image biological components in fixed cells. The review also discusses factors influencing nanoparticle imaging and uptake in live cells in vitro. Due to their unique size-dependent properties nanoparticles offer numerous advantages over traditional dyes and proteins. For example, the photostability, narrow emission peak, and ability to rationally modify both the size and surface chemistry of Quantum Dots allow for simultaneous analyses of multiple targets within the same cell. On the other hand, the surface characteristics of nanometer sized TiO2allow efficient conjugation to nucleic acids which enables their retention in specific subcellular compartments. We discuss cellular uptake mechanisms for the internalization of nanoparticles and studies showing the influence of nanoparticle size and charge and the cell type targeted on nanoparticle uptake. The predominant nanoparticle uptake mechanisms include clathrin-dependent mechanisms, macropinocytosis, and phagocytosis

    FEES protocol derived estimates of sensitivity: aspiration in dysphagia patients

    Aspiration is a common phenomenon in patients with oropharyngeal dysphagia. It can be studied using fiberoptic endoscopic evaluation of swallowing (FEES). FEES is well known and widely used in the diagnosis and treatment of swallowing disorders. However, various protocols exist, and there is no consensus on the examination protocol. The objective of this prospective study was to determine the FEES protocol derived estimates of sensitivity (Se') to detection of aspiration in dysphagic patients. The study estimated the probability of aspiration as a function of the number of swallow trials in dysphagic patients using FEES. The derived sensitivity was calculated based on presence or absence of aspiration in a ten-swallow trial protocol as arbitrary 'gold standard'. Eighty-four persons were included, comprising two patient populations with oropharyngeal dysphagia. Dysphagia in one group was due to head and neck cancer and possible oncological treatment effects on swallowing; in the other it was a result of neurological disease. All patients underwent a standardized FEES examination using ten swallows of thin liquid followed by ten swallows of thick liquid, all in boluses of 10 cc each. FEES recordings were rated for aspiration by an expert panel blinded to patients' identity and clinical history. Descriptive statistics, Kaplan–Meier survival analysis techniques, and Log Rank/Mantel–Cox tests were used. In both patient populations the aspiration risk was underestimated when using a limited number (three or four) of swallow trials. The oncology and neurology patients differed significantly in the number of swallow trials required to determine aspiration for thin liquids (median values 2 and 7 respectively, P = 0.006). FEES protocols using a limited number of swallow trials can underestimate the aspiration risk in both oncological and neurological patients suffering from oropharyngeal dysphagia, especially when using boluses with a thin liquid consistency
