    Ethnopharmacological study of medicinal plants used against skin ailments on Mount Pelion, central Greece

    Throughout centuries, traditional herbal medicine and the employment of medicinal plants have constituted an important tool for the treatment and prevention of numerous diseases. The present study focuses on the collection of ethnopharmacological data regarding the uses of medicinal plants for the treatment of dermatological ailments in various villages of Mount Pelion, Greece. More specifically, the study area is represented by the city of Volos and villages located in Central West Pelion and has not been investigated up to now. The information on the medicinal uses of the various species was obtained through extensive semi-structured interviews or the completion of specific questionnaires by the informants. Although the Covid-19 pandemic caused difficulties and obstacles in carrying out this research procedure, 60 informants were recruited and interviewed (36 women and 24 men). Their age range was between 31 and 97 years and their educational level was characterized by great diversity (primary, secondary, and higher education). The elaboration of the gathered information included the calculation of some quantitative indices, such as Fidelity Level (FL), and Informant Consensus Factor (FIC). Moreover, the relative importance of each reported species was identified by calculating the Use Value (UV). The interviews revealed 38 plant taxa belonging to 27 plant families reported to be used in the study area exclusively against skin diseases. The plant family mostly mentioned by the informants was Hypericaceae, followed by Plantaginaceae and Amaryllidaceae, while among the most popular methods of application are cataplasms, compresses, and topical application of decoction or raw plant material. Some of the most cited species are Hypericum perforatum L., Quercus coccifera L., and Plantago sp., traditionally used to treat skin problems such as eczema, wounds, and insect stings. The present ethnopharmacological study is the first documentation of ethnobotanical knowledge of this area that points out the traditional uses of medicinal plants against skin ailments

    Kainari, a Unique Greek Traditional Herbal Tea, from the Island of Lesvos: Chemical Analysis and Antioxidant and Antimicrobial Properties

    The chemical composition, as well as the total phenolic content (TPC) and the potential antioxidant and antimicrobial activity, of three Kainari-herbal tea samples from different areas of Lesvos Island (Greece) was evaluated. The rich aroma of the mixtures was studied through GC-MS, as well as through Headspace Solid-Phase Microextraction (HS-SPME)/GC-MS analyses. Cinnamon, clove, nutmeg, pepper, and ginger were identified as main ingredients, while, throughout the chemical analysis of the volatiles of one selected sample, several secondary metabolites have been isolated and identified on the basis of GC-MS as well as spectral evidence as eugenol, cinnamic aldehyde and myristicin, cinnamyl alcohol, alpha-terpinyl acetate, and β-caryophyllene. Furthermore, two food dyes, azorubine and amaranth, were also isolated and identified from the infusions. The total phenolic content was estimated and the free radical scavenging activity was determined by DPPH and ABTS assays and the antimicrobial activity of the extracts was tested showing a very interesting profile against all the assayed microorganisms. Due to its very pleasant aroma and taste properties as well as to its bioactivities, Kainari-herbal tea could be further proposed as functional beverage

    Propolis Extracts Inhibit UV-Induced Photodamage in Human Experimental In Vitro Skin Models

    The aim of this study was to assess the antioxidant, photoprotective, and antiaging effects of Greek propolis. Propolis was subjected to n-heptane or methanol extraction. Total phenolic/flavonoid content and antioxidant potential were determined in the extracts. Promising extracts were evaluated for their cytoprotective properties using human immortalized keratinocyte (HaCaT) or reconstituted human skin tissue following exposure to UVB. Assessment of cytotoxicity, DNA damage, oxidative status, and gene/protein expression levels of various matrix metalloproteinases (MMPs) were performed. The propolis methanolic fractions exhibited higher total phenolic and flavonoid contents and significant in vitro antioxidant activity. Incubation of HaCaT cells with certain methanolic extracts significantly decreased the formation of DNA strand breaks following exposure to UVB and attenuated UVB-induced decrease in cell viability. The extracts had no remarkable effect on the total antioxidant status, but significantly lowered total protein carbonyl content used as a marker for protein oxidation in HaCaT cells. MMP-1, -3, -7, and -9, monitored as endpoints of antiaging efficacy, were significantly reduced by propolis following UVB exposure in a model of reconstituted skin tissue. In conclusion, propolis protects against the oxidative and photodamaging effects of UVB and could be further explored as a promising agent for developing natural antiaging strategies

    Chemical analysis of Greek pollen - Antioxidant, antimicrobial and proteasome activation properties

    <p>Abstract</p> <p>Background</p> <p>Pollen is a bee-product known for its medical properties from ancient times. In our days is increasingly used as health food supplement and especially as a tonic primarily with appeal to the elderly to ameliorate the effects of ageing. In order to evaluate the chemical composition and the biological activity of Greek pollen which has never been studied before, one sample with identified botanical origin from sixteen different common plant taxa of Greece has been evaluated.</p> <p>Results</p> <p>Three different extracts of the studied sample of Greek pollen, have been tested, in whether could induce proteasome activities in human fibroblasts. The water extract was found to induce a highly proteasome activity, showing interesting antioxidant properties. Due to this activity the aqueous extract was further subjected to chemical analysis and seven flavonoids have been isolated and identified by modern spectral means. From the methanolic extract, sugars, lipid acids, phenolic acids and their esters have been also identified, which mainly participate to the biosynthetic pathway of pollen phenolics. The total phenolics were estimated with the Folin-Ciocalteau reagent and the total antioxidant activity was determined by the DPPH method while the extracts and the isolated compounds were also tested for their antimicrobial activity by the dilution technique.</p> <p>Conclusions</p> <p>The Greek pollen is rich in flavonoids and phenolic acids which indicate the observed free radical scavenging activity, the effects of pollen on human fibroblasts and the interesting antimicrobial profile.</p

    Qualitative Analysis Related to Palynological Characterization and Biological Evaluation of Propolis from Prespa National Park (Greece)

    Propolis samples from a geographical part of northwest Greece (Prespa National Park, PNP), which is characterized as a plant endemism center and biodiversity hotspot, were characterized through pollen analysis, chemically analyzed, and biologically evaluated. The majority of the studied propolis showed typical chemical constituents (phenolic acids, flavonoids, and chalcones) of European type, while a sample of Mediterranean-type propolis (rich in diterpenes) was also identified. The palynological characterization was implemented to determine the botanical origin and to explain the chemical composition. The total phenolic content and the DPPH assay showed that the European-type propolis samples possessed strong antioxidant activity (86&ndash;91% inhibition at 200 &mu;g/mL). Moreover, promising antibacterial activity of the extracts (MIC values 0.56&ndash;1.95 mg/mL) and moderate antifungal activity (MIC values 1.13&ndash;2.40 mg/mL) were noticed, while the sample with the highest activity had a significant content in terpenes (Mediterranean type). Propolis samples from the PNP area represent a rich source of antibacterial and antioxidant compounds and confirm the fact that propolis is a significant natural product with potential use for improving human health and stimulating the body&rsquo;s defense. Finally, it is noteworthy that a significant chemical diversity was demonstrated, even in samples from a limited geographical area as this of PNP

    Production of bioactive metabolites with pharmaceutical and nutraceutical interest by submerged fermentation of Pleurotus ostreatus in a batch stirred tank bioreactor

    Mushrooms comprise a vast source of new pharmaceutical and nutraceutical products. Submerged fermentation of the mycelial form of mushroom-producing fungi is promising for efficient production of their biomass and active metabolites. The aim of this study was the isolation and identification of bioactive metabolites derived from the edible mushroom Pleurotus ostreatus biomass, produced by submerged fermentation in a batch stirred tank bioreactor. The dichloromethane extract was fractioned by medium pressure liquid chromatography (MPLC) and Sephadex LH-20 column chromatography. For extracting efficiently the phenolic compounds of the methanolic extract, an adsorption-desorption process, using XAD-4 type resin, was performed. The pure compounds were elucidated with 1D/2D NMR-spectroscopic analyses, NMR data comparisons, and chemical correlations combined with GC/MS-LC/MS experiments. The compounds afforded by the dichloromethane extract were identified as linoleic acid (1), oleic acid (2), stearic acid (3), palmitic acid (4) and their corresponding methyl esters (5-8, respectively), benzoic acid (9), trans 3, 4-dihydro-3, 4, 8-trihydroxynapthalen-1(2H)-one (10), 4-hydroxybenzaldehyde (11), indolo-3-carboxylic acid (12) and uracil (13). The investigation of the methanolic extract led to the isolation of 3-formyl-pyrrole (14), 4-hydroxy-benzoic acid (15), uridine (16), nicotinic acid (17) and nicotinamide (18). Based on existing literature data, all these compounds exhibit valuable biological properties. The information obtained is considered fundamental for further investigation of the P. ostreatus fermentation process on an industrial scale for enhanced bioactive metabolite production.
