Tumor Necrosis Factor-Ī± Signaling in Macrophages
Tumor necrosis factor-Ī± (TNFĪ±) was cloned over 2 decades ago and its identification in part led to the discovery of a super family of tumor necrosis factors (TNFs) and their receptors. TNFĪ± signals through two transmembrane receptors, TNFR1 and TNFR2, and regulates a number of critical cell functions including cell proliferation, survival, differentiation, and apoptosis. Macrophages are the major producers of TNFĪ± and interestingly are also highly responsive to TNFĪ±. Aberrant TNFĪ± production and TNF receptor signaling have been associated with the pathogenesis of several diseases, including rheumatoid arthritis, Crohnās disease, atherosclerosis, psoriasis, sepsis, diabetes, and obesity. TNFĪ± has been shown to play a pivotal role in orchestrating the cytokine cascade in many inflammatory diseases and because of this role as a āmaster-regulatorā of inflammatory cytokine production, it has been proposed as a therapeutic target for a number of diseases. Indeed anti-TNFĪ± drugs are now licensed for treating certain inflammatory diseases including rheumatoid arthritis and inflammatory bowel disease. In this review we discuss the discovery of TNFĪ± and its actions especially in regulating macrophage biology. Given its importance in several human diseases, we also briefly discuss the role of anti-TNFĪ± therapeutics in the treatment of inflammatory diseases
Does mothers' perception of wheezing affect the nutritional status of wheezers?
Abstract This study was conducted to find out mothers' perception of factors associated with wheezing and its effect on the nutritional status of children with recurrent wheezing. 'Cold food' which includes all refrigerated food, curd and certain types of green leafy vegetables were perceived to be associated with wheezing by 70% of mothers. These items were not included in the diet of children with wheezing. However there was no significant difference in the nutritional intake or nutritional status of children with and without wheezing perhaps due to infrequent inclusion of the same even in the normal children's diet
