    NOD1-Mediated Mucosal Host Defense against Helicobacter pylori

    Infection of the stomach with Helicobacter pylori is an important risk factor for gastritis, peptic ulcer, and gastric carcinoma. Although it has been well established that persistent colonization by H. pylori is associated with adaptive Th1 responses, the innate immune responses leading to these Th1 responses are poorly defined. Recent studies have shown that the activation of nucleotide-binding oligomerization domain 1 (NOD1) in gastric epithelial cells plays an important role in innate immune responses against H. pylori. The detection of H. pylori-derived ligands by cytosolic NOD1 induces several host defense factors, including antimicrobial peptides, cytokines, and chemokines.     The Nutritional Status of Japanese Black Breeding Cows Grazed in a Larger Abandoned Paddy Field

    Because rice consumption has decreased in Japan, the number of abandoned paddy fields has increased. Some of the abandoned paddy fields are being used for livestock grazing, effectively using the abandoned fields and reducing the production costs on livestock farms. Such grazing may use an integrated grazing system of scattered small pastures where each of the small abandoned paddy fields scattered in a given agriculture area is defined as one grazing yard, and a herd consisting of two or three head of cattle is moved from one grazing yard to another. However, because the number of abandoned paddy fields has increased, it may be possible to have cattle grazing in larger fields made by combining multiple abandoned paddy fields (Shindo and Tejima 2006).     A Case of Severe Esophageal Intramural Pseudodiverticulosis Whose Symptoms Were Ameliorated by Oral Administration of Anti-Fungal Medicine

    Esophageal intramural pseudodiverticulosis (EIPD) is a rare disease of unknown etiology that displays multiple pseudodiverticula radiologically, leading to benign esophageal stricture. Dysphagia, which sometimes slowly progresses, is the main symptom in the majority of cases. We here report a 59-year-old male EIPD patient who suffered from severe dysphagia. Radiography and endoscopy of this patient disclosed a severe constriction in the upper thoracic esophagus. Although we tried several endoscopic procedures including frequent endoscopic balloon dilatation (EBD), the effect was very limited and his dysphagia relapsed shortly after the treatments. During the procedures, we noticed some white, thick, creamy liquid emerging from the orifices of EIPD, and PAS staining of biopsy specimens revealed infection with Candida albicans. Hence, the patient was given anti-fungal medicine in addition to EBD. The additional treatment with anti-fungal medicine dramatically improved his symptoms and the esophageal constriction. This case suggests that anti-fungal treatment is an effective first-line therapy even against a severe form of esophageal constriction in EIPD

    Potential demand for eco-museum excursions in Higashihiroshima City

    エコミュージアムにおいて,地域に散在する遺産をどのように結びつけるかは1つの課題である。1つの解決策として,域内をめぐる見学ツアーを提供することが考えられる。本稿の目的は,東広島市で想定できるエコミュージアム・ツアーにどの程度の需要が見込めるのかを示すことである。そのために,広島県民へのウェブアンケート調査と,広島大学学生の意見調査を行った。ウェブアンケートからは,エコミュージアムに関心をもつ層が3分の1おり,ツアーへの参加希望は,「水と酒」をテーマとするものが56.0%,「生物と農村生活」で44.9%,「バイオマス関連」で40.3%,「伝説と歴史」で30.3%であることが示された。また,学生の意見としては,大学のある地域について知る機会を大事にしたい,日常的に自然に接する機会が少ないので提示された機会は貴重だと考える者がいる一方,参加費用面でそもそも無理だと判断する者がおり,両者はほぼ半々となった。見学ツアーに対して,広島大学総合博物館単独での実施能力を大きく上回る潜在的な需要があることが確認できた。Finding ways to connect scattered heritages in an eco-museum constitutes an issue. Solutions include providing excursions around the region. This article aims to demonstrate how much demand can be expected for the excursions of an eco-museum in Higashihiroshima City. To this end, an online questionnaire survey was administered to citizens of Hiroshima Prefecture, and an opinion survey of Hiroshima University students was conducted. According to the online survey, 56.0% of respondents were interested in participating in the excursion on the theme of “water and liquor”, and 44.9% were interested in the excursion regarding “wild living and rural life”. Further, 40.3% were interested in the “biomass-related” excursion and 30.3% were interested in the excursion on “local history and legends”. The opinions of university students were as follows. Almost half the students stated that the presented opportunities are valuable as they want to learn about the area surrounding the university and there are few opportunities to interact with nature in everyday life. Conversely, the other half of participants deemed the participation fee excessively high.     Human Oncoprotein 5MP Suppresses General and Repeat-Associated Non-AUG Translation via eIF3 by a Common Mechanism

    eIF5-mimic protein (5MP) is a translational regulatory protein that binds the small ribosomal subunit and modulates its activity. 5MP is proposed to reprogram non-AUG translation rates for oncogenes in cancer, but its role in controlling non-AUG initiated synthesis of deleterious repeat-peptide products, such as FMRpolyG observed in fragile-X-associated tremor ataxia syndrome (FXTAS), is unknown. Here, we show that 5MP can suppress both general and repeat-associated non-AUG (RAN) translation by a common mechanism in a manner dependent on its interaction with eIF3. Essentially, 5MP displaces eIF5 through the eIF3c subunit within the preinitiation complex (PIC), thereby increasing the accuracy of initiation. In Drosophila, 5MP/Kra represses neuronal toxicity and enhances the lifespan in an FXTAS disease model. These results implicate 5MP in protecting cells from unwanted byproducts of non-AUG translation in neurodegeneration

    Pancreatic insulin release in vitamin C-deficient senescence marker protein-30/gluconolactonase knockout mice

    We recently identified senescence marker protein-30 as the lactone-hydrolyzing enzyme gluconolactonase, which is involved in vitamin C biosynthesis. In this study, we investigated the effects of vitamin C on insulin secretion from pancreatic β-cells using senescence marker protein-30/gluconolactonase knockout mice. In intraperitoneal glucose tolerance tests, vitamin C-deficient senescence marker protein-30/gluconolactonase knockout mice demonstrated impaired glucose tolerance with significantly lower blood insulin levels at 30 and 120 min post-challenge than in wild type mice (p<0.01–0.05). In contrast, vitamin C-sufficient senescence marker protein-30/gluconolactonase knockout mice demonstrated significantly higher blood glucose and lower insulin only at the 30 min post-challenge time point (p<0.05). Senescence marker protein-30/gluconolactonase knockout mice showed enhanced insulin sensitivity regardless of vitamin C status. Static incubation of islets revealed that 20 mM glucose-stimulated insulin secretion and islet ATP production were significantly decreased at 60 min only in vitamin C-deficient SMP30/GNL knockout mice relative to wild type mice (p<0.05). These results indicate that the site of vitamin C action lies between glycolysis and mitochondrial oxidative phosphorylation, while SMP30 deficiency itself impairs the distal portion of insulin secretion pathway

    Gene up-regulation in response to predator kairomones in the water flea, Daphnia pulex

    <p>Abstract</p> <p>Background</p> <p>Numerous cases of predator-induced polyphenisms, in which alternate phenotypes are produced in response to extrinsic stimuli, have been reported in aquatic taxa to date. The genus <it>Daphnia </it>(Branchiopoda, Cladocera) provides a model experimental system for the study of the developmental mechanisms and evolutionary processes associated with predator-induced polyphenisms. In <it>D. pulex</it>, juveniles form neckteeth in response to predatory kairomones released by <it>Chaoborus </it>larvae (Insecta, Diptera).</p> <p>Results</p> <p>Previous studies suggest that the timing of the sensitivity to kairomones in <it>D. pulex </it>can generally be divided into the embryonic and postembryonic developmental periods. We therefore examined which of the genes in the embryonic and first-instar juvenile stages exhibit different expression levels in the presence or absence of predator kairomones. Employing a candidate gene approach and identifying differentially-expressed genes revealed that the morphogenetic factors, <it>Hox3</it>, <it>extradenticle </it>and <it>escargot</it>, were up-regulated by kairomones in the postembryonic stage and may potentially be responsible for defense morph formation. In addition, the juvenile hormone pathway genes, <it>JHAMT </it>and <it>Met</it>, and the insulin signaling pathway genes, <it>InR </it>and <it>IRS-1</it>, were up-regulated in the first-instar stage. It is well known that these hormonal pathways are involved in physiological regulation following morphogenesis in many insect species. During the embryonic stage when morphotypes were determined, one of the novel genes identified by differential display was up-regulated, suggesting that this gene may be related to morphotype determination. Biological functions of the up-regulated genes are discussed in the context of defense morph formation.</p> <p>Conclusions</p> <p>It is suggested that, following the reception of kairomone signals, the identified genes are involved in a series of defensive phenotypic alterations and the production of a defensive phenotype.</p

    Detection of the Far-infrared [O III] and Dust Emission in a Galaxy at Redshift 8.312: Early Metal Enrichment in the Heart of the Reionization Era

    We present the Atacama Large Millimeter/submillimeter Array (ALMA) detection of the [O III] 88 μ\mum line and rest-frame 90 μ\mum dust continuum emission in a Y-dropout Lyman break galaxy (LBG), MACS0416_Y1, lying behind the Frontier Field cluster MACS J0416.1-2403. This [O III] detection confirms the LBG with a spectroscopic redshift of z=8.3118±0.0003z = 8.3118 \pm 0.0003, making this object one of the furthest galaxies ever identified spectroscopically. The observed 850 μ\mum flux density of 137±26137 \pm 26 μ\muJy corresponds to a de-lensed total infrared (IR) luminosity of LIR=(1.7±0.3)×1011LL_{\rm IR} = (1.7 \pm 0.3) \times 10^{11} L_{\odot} if assuming a dust temperature of Tdust=50T_{\rm dust} = 50 K and an emissivity index of β=1.5\beta = 1.5, yielding a large dust mass of 4×106M4 \times 10^6 M_{\odot}. The ultraviolet-to-far IR spectral energy distribution modeling where the [O III] emissivity model is incorporated suggests the presence of a young (τage4\tau_{\rm age} \approx 4 Myr), star-forming (SFR 60M\approx 60 M_{\odot} yr1^{-1}), moderately metal-polluted (Z0.2ZZ \approx 0.2 Z_{\odot}) stellar component with a mass of Mstar=3×108MM_{\rm star} = 3 \times 10^8 M_{\odot}. An analytic dust mass evolution model with a single episode of star-formation does not reproduce the metallicity and dust mass in τage4\tau_{\rm age} \approx 4 Myr, suggesting a pre-existing evolved stellar component with Mstar3×109MM_{\rm star} \sim 3 \times 10^9 M_{\odot} and τage0.3\tau_{\rm age} \sim 0.3 Gyr as the origin of the dust mass.Comment: Accepted by ApJ. 18 pages, 10 figures, 5 table
