Flavonol glycosides from the aerial parts of Aceriphyllum rossii and their antioxidant activities
The methanol extract obtained from the aerial parts ofAceriphyllum rossii (Saxifragaceae) was fractionated into ethyl acetate (EtOAc),n-BuOH and H2O layers through solvent fractionation. Repeated silica gel column chromatography of EtOAc andn-BuOH layers afforded six flavonol glycosides. They were identified as kaempferol 3-O-β-D-glucopyranoside (astragalin,1), quercetin 3-O-β-D-glucopyranoside (isoquercitrin,2), kaempferol 3-O-α-L-rhamnopyranosyl (1→6)-β-D-glucopyranoside (3), quercetin 3-O-α-L-rhamnopyranosyl (1→6)-β-D-glucopyrano-side (rutin,4), kaempferol 3-O-[α-L-rhamnopyranosyl (1→4)-α-L-rhamnopyranosyl (1→6)-β-D-glucopyranoside] (5) and quercetin 3-O-[α-L-rhamnopyranosyl (1→4)-α-L-rhamnopyranosyl (1→6)-β-D-glucopyranoside] (6) on the basis of several spectral data. The antioxidant activity of the six compounds was investigated using two free radicals such as the ABTS free radical and superoxide anion radical. Compound1 exhibited the highest antioxidant activity in the ABTS2,2-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) radical scavenging method. 100 mg/L of compound1 was equivalent to 72.1±1.4 mg/L of vitamin C, and those of compounds3 and5 were equivalent to 62.7±0.5 mg/L and 54.3±1.3 mg/L of vitamin C, respectively. Gaussian Quantum Illumination via Monotone Metrics
Quantum illumination is to discern the presence or absence of a low
reflectivity target, where the error probability decays exponentially in the
number of copies used. When the target reflectivity is small so that it is hard
to distinguish target presence or absence, the exponential decay constant falls
into a class of objects called monotone metrics. We evaluate monotone metrics
restricted to Gaussian states in terms of first-order moments and covariance
matrix. Under the assumption of a low reflectivity target, we explicitly derive
analytic formulae for decay constant of an arbitrary Gaussian input state.
Especially, in the limit of large background noise and low reflectivity, there
is no need of symplectic diagonalization which usually complicates the
computation of decay constants. First, we show that two-mode squeezed vacuum
(TMSV) states are the optimal probe among pure Gaussian states with fixed
signal mean photon number. Second, as an alternative to preparing TMSV states
with high mean photon number, we show that preparing a TMSV state with low mean
photon number and displacing the signal mode is a more experimentally feasible
setup without degrading the performance that much. Third, we show that it is of
utmost importance to prepare an efficient idler memory to beat coherent states
and provide analytic bounds on the idler memory transmittivity in terms of
signal power, background noise, and idler memory noise. Finally, we identify
the region of physically possible correlations between the signal and idler

Connexin32 inhibits gastric carcinogenesis through cell cycle arrest and altered expression of p21Cip1 and p27Kip1
Gap junctions and their structural proteins, connexins (Cxs), havebeen implicated in carcinogenesis. To explore the involvement ofCx32 in gastric carcinogenesis, immunochemical analysis of Cx32and proliferation marker Ki67 using tissue-microarrayed humangastric cancer and normal tissues was performed. In addition, afterCx32 overexpression in the human gastric cancer cell line AGS,cell proliferation, cell cycle analyses, and p21Cip1 and p27Kip1expression levels were examined by bromodeoxyuridine assay,flow cytometry, real-time RT-PCR, and western blotting.Immunohistochemical study noted a strong inverse correlationbetween Cx32 and Ki67 expression pattern as well as theirlocation. In vitro, overexpression of Cx32 in AGS cells inhibitedcell proliferation significantly. G1 arrest, up-regulation of cellcycle-regulatory proteins p21Cip1 and p27Kip1 was also found atboth mRNA and protein levels. Taken together, Cx32 plays someroles in gastric cancer development by inhibiting gastric cancercell proliferation through cell cycle arrest and cell cycle regulatoryproteins. Role of histamine-mediated macrophage differentiation in clearance of metastatic bacterial infection
Macrophages are highly heterogeneous immune cells with a role in maintaining tissue homeostasis, especially in activating the defense response to bacterial infection. Using flow cytometric and single-cell RNA-sequencing analyses of peritoneal cells, we here show that small peritoneal macrophage and immature macrophage populations are enriched in histamine-deficient (Hdc-/-) mice, characterized by a CD11bmiF4/80loCCR2+MHCIIhi and CD11bloF4/80miTHBS1+IL-1α+ phenotype, respectively. Molecular characterization revealed that immature macrophages represent an abnormally differentiated form of large peritoneal macrophages with strong inflammatory properties. Furthermore, deficiency in histamine signaling resulted in significant impairment of the phagocytic activity of peritoneal macrophage populations, conferring high susceptibility to bacterial infection. Collectively, this study reveals the importance of histamine signaling in macrophage differentiation at the molecular level to maintain tissue homeostasis, offering a potential therapeutic target for bacterial infection-mediated diseases
Successful mobilization using a combination of plerixafor and G-CSF in pediatric patients who failed previous chemomobilization with G-CSF alone and possible complications of the treatment
Peripheral blood stem cell (PBSC) mobilization, which uses plerixafor (AMD 3100), a newly developed specific inhibitor of the CXCR4 receptor, in combination with granulocyte-colony stimulating factor(G-CSF), has been shown to enhance the stem cell mobilization in adult patients, but pediatric data are scarce. We documented our experience with this drug in 6 Korean pediatric patients who had failed in chemomobilization, using G-CSF, alone. All patients were mobilized CD34+ cells (median, 11.08 × 106/kg: range, 6.34-28.97 × 106/kg) successfully within 2 to 3 cycles of apheresis, without complications. A total of 7 autologous transplantations were performed, including 1 tandem transplantation. However, 2 patients with brain tumors showed severe pulmonary complications, including spontaneous pneumomediastinum. This is the first study of PBSC mobilization with plerixafor in Asian pediatric patients. Furthermore our study suggests that mobilization with plerixafor may be effective in Korean pediatric patients, who have previously been heavily treated and have failed PBSC mobilization with classical chemomobilization, using G-CSF. However, further studies are needed to examine the possible complications of autologous transplantation, using a mobilized plerixafor product in children
Antidiabetic Effect of Morinda citrifolia
Antidiabetic effects of Morinda citrifolia (aka Noni) fermented by Cheonggukjang (fast-fermented soybean paste) were evaluated using a T2DM (type 2 diabetes mellitus) murine model. Six-week-old KK-Ay/TaJcl mice were randomly divided into four groups: (1) the diabetic control (DC) group, provided with a normal mouse diet; (2) the positive control (PC) group, provided with a functional health food diet; (3) the M. citrifolia (MC) group, provided with an MC-based diet; (4) the fermented M. citrifolia (FMC) group, provided with an FMC-based diet. Over a testing period of 90 days, food and water intake decreased significantly in the FMC and PC groups compared with the DC group. Blood glucose levels in the FMC group were 211.60–252.20 mg/dL after 90 days, while those in the control group were over 400 mg/dL after 20 days. In addition, FMC supplementation reduced glycosylated hemoglobin (HbA1c) levels, enhanced insulin sensitivity, and significantly decreased serum triglycerides and low-density lipoprotein (LDL) cholesterol. Furthermore, a fermented M. citrifolia 70% ethanolic extract (FMCE) activated peroxisome proliferator-activated receptor-(PPAR-) γ and stimulated glucose uptake via stimulation of AMP-activated protein kinase (AMPK) in cultured C2C12 cells. These results suggest that FMC can be employed as a functional health food for T2DM management
