    Vascular function in the aging human brain during muscle exertion

    : To determine how brain oxygenation is stably maintained during advancing age, cerebral oxygenation and hemoglobin were measured real-time at 10 Hz using near-infrared spectroscopy (NIRS) at rest (30 seconds) and during a 10-repeated handgrip strength test (30 seconds) for 834 adults (M/F = 45/55%) aged 20-88 y. The amplitude of cerebral hemodynamic fluctuation was reflected by converting 300 values of % oxygen saturation and hemoglobin of each 30-second phase to standard deviation as indicatives of brain oxygenation variability (BOV) and brain hemodynamic variability (BHV) for each participant. Both BOV (+21-72%) and BHV (+94-158%) increased during the maximal voluntary muscle exertions for all age levels (α < 0.05), suggesting an increased vascular recruitment to maintain oxygen homeostasis in the brain. Intriguingly, BHV was >100 folds for both resting and challenged conditions (α < 0.001) in >80% of adults aged above 50 y despite similar BOV compared with young age counterparts, indicating a huge cost of amplifying hemodynamic oscillation to maintain a stable oxygenation in the aging brain. Since vascular endothelial cells are short-lived, our results implicate a hemodynamic compensation to emergence of daily deficits in replacing senescent endothelial cells after age 50 y

    Characteristics of inorganic phosphorus fractions and their correlations with soil properties in three non-acidic soils

    peer reviewedUnderstanding the characteristics and influences of various factors on phosphorus (P) fractions is of significance for promoting the efficiency of soil P. Based on long-term experiments on black soil, fluvo-aquic soil, and loess soil, which belong to Phaeozems, Cambisols, and Anthrosols in the World Reference Base for Soil Resources (WRB), respectively, five fertilization practices were selected and divided into three groups: no P fertilizer (CK/NK), balanced fertilizer (NPK/NPKS), and manure plus mineral fertilizer (NPKM). Soil inorganic P (Pi) fractions and soil properties were analyzed to investigate the characteristics of the Pi fractions and the relationships between Pi fractions and various soil properties. The results showed that the proportion of Ca10-P in the sum of total Pi fractions was the highest in the three soils, accounting for 33.5% in black soil, 48.8% in fluvo-aquic soil, and 44.8% in loess soil. Long-term fertilization practices resulted in periodic changes in soil Pi accumulation or depletion. For black soil and fluvo-aquic soil, the Pi accumulation was higher in the late period (10–20 years) of fertilization than in the early period (0–10 years) under NPK/NPKS and NPKM, whereas the opposite result was found in loess soil. The Pi accumulation occurred in all Pi fractions in black soil; mainly in Ca8-P, Fe-P, and Ca10-P in fluvo-aquic soil; and in Ca2-P, Ca8-P, and O-P in loess soil. Under CK/NK, the soil Pi was depleted mainly in the early period in each of the three soils. In addition to the labile Pi (Ca2-P) and moderately labile Pi (Ca8-P, Fe-P, Al-P), the Ca10-P in black soil and fluvo-aquic soil and O-P in loess soil could also be used by crops. Redundancy analysis showed that soil properties explained more than 90% of the variation in the Pi fractions in each soil, and the explanatory percentages of soil organic matter (SOM) were 43.6% in black soil, 74.6% in fluvo-aquic, and 38.2% in loess soil. Consequently, decisions regarding the application of P fertilizer should consider the accumulation rate and the variations in Pi fractions driven by soil properties in non-acidic soils

    Dynamics of Oxygen-Independent Photocleavage of Blebbistatin as a One-Photon Blue or Two-Photon Near-Infrared Light-Gated Hydroxyl Radical Photocage

    Development of versatile, chemically tunable photocages for photoactivated chemotherapy (PACT) represents an excellent opportunity to address the technical drawbacks of conventional photodynamic therapy (PDT) whose oxygen-dependent nature renders it inadequate in certain therapy contexts such as hypoxic tumors. As an alternative to PDT, oxygen free mechanisms to generate cytotoxic reactive oxygen species (ROS) by visible light cleavable photocages are in demand. Here, we report the detailed mechanisms by which the small molecule blebbistatin acts as a one-photon blue light-gated or two-photon near-infrared light-gated photocage to directly release a hydroxyl radical (•OH) in the absence of oxygen. By using femtosecond transient absorption spectroscopy and chemoselective ROS fluorescent probes, we analyze the dynamics and fate of blebbistatin during photolysis under blue light. Water-dependent photochemistry reveals a critical process of water-assisted protonation and excited state intramolecular proton transfer (ESIPT) that drives the formation of short-lived intermediates, which surprisingly culminates in the release of •OH but not superoxide or singlet oxygen from blebbistatin. CASPT2//CASSCF calculations confirm that hydrogen bonding between water and blebbistatin underpins this process. We further determine that blue light enables blebbistatin to induce mitochondria-dependent apoptosis, an attribute conducive to PACT development. Our work demonstrates blebbistatin as a controllable photocage for •OH generation and provides insight into the potential development of novel PACT agents

    An Updated Search of Steady TeV γ−\gamma-Ray Point Sources in Northern Hemisphere Using the Tibet Air Shower Array

    Using the data taken from Tibet II High Density (HD) Array (1997 February-1999 September) and Tibet-III array (1999 November-2005 November), our previous northern sky survey for TeV γ−\gamma-ray point sources has now been updated by a factor of 2.8 improved statistics. From 0.0∘0.0^{\circ} to 60.0∘60.0^{\circ} in declination (Dec) range, no new TeV γ−\gamma-ray point sources with sufficiently high significance were identified while the well-known Crab Nebula and Mrk421 remain to be the brightest TeV γ−\gamma-ray sources within the field of view of the Tibet air shower array. Based on the currently available data and at the 90% confidence level (C.L.), the flux upper limits for different power law index assumption are re-derived, which are approximately improved by 1.7 times as compared with our previous reported limits.Comment: This paper has been accepted by hepn
