    Effect of different grain structures on centerline macrosegregation during direct-chill casting

Copyright @ 2008 Elsevier B.V.Duplex grain structure consisting of coarse-cell and fine-cell dendritic grains is frequently found in the central portion of direct-chill cast billets and ingots. Coarse-cell grains are usually considered as free-floating crystals settled to the bottom of the billet sump. These grains are assumed to be solute-lean and contribute to the negative centerline segregation. In this paper the contribution of coarse-cell and fine-cell grains to macrosegregation is for the first time studied experimentally by direct measurements of their composition. It is shown that the coarse-cell, floating grains are depleted of solute and the areas of their accumulation contribute to the negative macrosegregation. The areas of fine-cell grains can be either enriched in solute or be close to the nominal composition. It is argued that their composition results from the interplay between thermo-solutal and shrinkage-induced flows. Copyright @ 2008 Elsevier B.V.Semi-continuous direct-chill (DC) casting holds a prominent position in commercial aluminium alloy processing, especially in production of large sized ingots. Macrosegregation, which is the non-uniform chemical composition over the length scale of a casting, is one of the major defects that occur during this process. The fact that macrosegregation is essentially unaffected by subsequent heat treatment (hence constitutes an irreversible defect) leaves us with little choice but to control it during the casting stage. Despite over a century of research in the phenomenon of macrosegregation in castings and good understanding of underlying mechanisms, the contributions of these mechanisms in the overall macrosegregation picture; and interplay between these mechanisms and the structure formation during solidification are still unclear. This review attempts to fill this gap based on the published data and own results. The following features make this review unique: results of computer simulations are used in order to separate the effects of different macrosegregation mechanisms. The issue of grain refining is specifically discussed in relation to macrosegregation. This report is structured as follows. Macrosegregation as a phenomenon is defined in the Introduction. In “Direct-chill casting – process parameters, solidification and structure patterns” section, direct-chill casting, the role of process parameters and the evolution of structural features in the as-cast billets are described. In “Macrosegregation in direct-chill casting of aluminium alloys” section, macrosegregation mechanisms are elucidated in a historical perspective and the correlation with DC casting process parameters and structural features are made. The issue of how to control macrosegregation in direct-chill casting is also dealt with in the same section. In “Role of grain refining” section, the effect of grain refining on macrosegregation is introduced, the current understanding is described and the contentious issues are outlined.     Influence of melt feeding scheme and casting parameters during direct-chill casting on microstructure of an AA7050 billet

    © The Minerals, Metals & Materials Society and ASM International 2012Direct-chill (DC) casting billets of an AA7050 alloy produced with different melt feeding schemes and casting speeds were examined in order to reveal the effect of these factors on the evolution of microstructure. Experimental results show that grain size is strongly influenced by the casting speed. In addition, the distribution of grain sizes across the billet diameter is mostly determined by melt feeding scheme. Grains tend to coarsen towards the center of a billet cast with the semi-horizontal melt feeding, while upon vertical melt feeding the minimum grain size was observed in the center of the billet. Computer simulations were preformed to reveal sump profiles and flow patterns during casting under different melt feeding schemes and casting speeds. The results show that solidification front and velocity distribution of the melt in the liquid and slurry zones are very different under different melt feeding scheme. The final grain structure and the grain size distribution in a DC casting billet is a result of a combination of fragmentation effects in the slurry zone and the cooling rate in the solidification range

    Designing liposomal adjuvants for the next generation of vaccines

    Liposomes not only offer the ability to enhance drug delivery, but can effectively act as vaccine delivery systems and adjuvants. Their flexibility in size, charge, bilayer rigidity and composition allow for targeted antigen delivery via a range of administration routes. In the development of liposomal adjuvants, the type of immune response promoted has been linked to their physico-chemical characteristics, with the size and charge of the liposomal particles impacting on liposome biodistribution, exposure in the lymph nodes and recruitment of the innate immune system. The addition of immunostimulatory agents can further potentiate their immunogenic properties. Here, we outline the attributes that should be considered in the design and manufacture of liposomal adjuvants for the delivery of sub-unit and nucleic acid based vaccines

    Dynamic wavefront shaping with an acousto-optic lens for laser scanning microscopy

    Acousto-optic deflectors (AODs) arranged in series and driven with linearly chirped frequencies can rapidly focus and tilt optical wavefronts, enabling high-speed 3D random access microscopy. Non-linearly chirped acoustic drive frequencies can also be used to shape the optical wavefront allowing a range of higher-order aberrations to be generated. However, to date, wavefront shaping with AODs has been achieved by using single laser pulses for strobed illumination to 'freeze' the moving acoustic wavefront, limiting voxel acquisition rates. Here we show that dynamic wavefront shaping can be achieved by applying non-linear drive frequencies to a pair of AODs with counter-propagating acoustic waves, which comprise a cylindrical acousto-optic lens (AOL). Using a cylindrical AOL we demonstrate high-speed continuous axial line scanning and the first experimental AOL-based correction of a cylindrical lens aberration at 30 kHz, accurate to 1/35th of a wave at 800 nm. Furthermore, we develop a model to show how spherical aberration, which is the major aberration in AOL-based remote-focusing systems, can be partially or fully corrected with AOLs consisting of four or six AODs, respectively

    Lifetime prevalence of novel psychoactive substances use among adults in the USA: Sociodemographic, mental health and illicit drug use correlates. Evidence from a population-based survey 2007–2014

    INTRODUCTION: As Novel psychoactive substances (NPS) are conceived to mimic the effects of common illicit drugs, they represent a serious public health challenge due to the spike in intoxications and fatalities that have been linked to their use. This study aims to provide epidemiological data on NPS use in the USA, determining lifetime prevalence of use and defining demographic, socioeconomic, drug use patterns and mental health correlates. METHODS: This study uses secondary data from the US National Survey on Drug Use and Health (NSDUH), which is a large cross-sectional population-based survey carried out annually in the USA. We analysed data from 2007-14 (N = 307,935) using bivariate descriptive analysis and binary logistic regression to calculate prevalence and determine factors underlying NPS consumption. Adjusted odds ratios (OR) with 95% CI's were calculated for a set of selected independent variables. RESULTS AND DISCUSSION: Our analysis NSDUH from 2007-14 highlights an increase in NPS use among adults, especially among white young men aged 18 to 25. Although the level of education of NPS users was relatively higher as compared to non-users, NPS users seemed to have a less wealthy situation. However, socioeconomic vulnerability appeared to be less important than mental health issues as a correlate to NPS use. NPS users seem to have followed a pattern of polysubstance use throughout their life, which involves both traditional illicit drugs and classic synthetic drugs. As NPS use seemed to be more prevalent among people having mental health issues, the rise in their use may have a negative impact on population mental health outcomes. CONCLUSION: Further comparative research on trends in NPS use and potential public health responses would be instrumental for developing appropriate health interventions, including drug checking, education for users and training for healthcare professionals working both within emergency wards and in/outpatient addiction and mental health services

    The Cadmium Zinc Telluride Imager on AstroSat has proven to be a very effective all-sky monitor in the hard X-ray regime, detecting over three hundred GRBs and putting highly competitive upper limits on X-ray emissions from gravitational wave sources and fast radio bursts. We present the algorithms used for searching for such transient sources in CZTI data, and for calculating upper limits in case of non-detections. We introduce CIFT: the CZTI Interface for Fast Transients, a framework used to streamline these processes. We present details of 88 new GRBs detected by this framework that were previously not detected in CZTI

    Capecitabine in combination with docetaxel and mitomycin C in patients with pre-treated tumours: results of an extended phase-I trial

    Preclinical data suggest that the anti-tumour activity of capecitabine is enhanced by taxanes and mitomycin C through up-regulation of thymidine phosphorylase (TP). Here, we studied safety and efficacy of the combination of capecitabine with docetaxel and mitomycin C. Two dose levels (DL) were investigated: capecitabine 1000 mg m−2 b.i.d. on days 1–14, docetaxel 40 mg m−2 i.v. day 1, mitomycin C 4 or 6 mg m−2 i.v. day 1 (DL I or II). Cycles were repeated every 3 weeks. The primary aim was to determine the dose-limiting toxicities (DLT) during the first two treatment cycles and the maximum tolerated dose (MTD). A total of 46 patients (pts) refractory to standard therapies were enrolled, of whom the majority had gastrointestinal tumours (n=40). 14 pts had received ⩾3 lines of prior chemotherapy. At DL I, one out of six pts experienced DLT. At DL II, two out of six pts had DLT (mucositis grade 3). Thus, DL I was determined as MTD. Among a total of 37 pts treated on DL I the following toxicities were observed during cycles 1 and 2 (number of patients with grade 1/2/3/4 toxicity; NCI-CTC v. 3.0): anaemia 10/8/3/0, leucocytopenia 4/11/1/2, thrombocytopenia 0/1/2/0, diarrhoea 8/1/2/0, stomatitis/mucositis 3/3/1/0, nausea/vomiting 10/2/0/0, and hand-foot skin reaction 5/1/1/0. Of 30 pts who received at least two treatment cycles nine achieved complete or partial remissions, six pts achieved minor remissions, and seven pts had stable disease (tumour control rate 73%). Of note, four out of 10 patients with pancreatic cancer had partial remissions. In conclusion, capecitabine can safely be combined with docetaxel (40 mg m−2) and mitomycin C (4 mg m−2). The established regimen was well tolerated and the preliminary efficacy data in this heavily pre-treated patients population appears to be promising
