    Imaging Pulmonary NF-kappaB Activation and Therapeutic Effects of MLN120B and TDZD-8

    NF-ÎșB activation is a critical signaling event in the inflammatory response and has been implicated in a number of pathological lung diseases. To enable the assessment of NF-ÎșB activity in the lungs, we transfected a luciferase based NF-ÎșB reporter into the lungs of mice or into Raw264.7 cells in culture. The transfected mice showed specific luciferase expression in the pulmonary tissues. Using these mouse models, we studied the kinetics of NF-ÎșB activation following exposure to lipopolysaccharide (LPS). The Raw264.7 cells expressed a dose-dependent increase in luciferase following exposure to LPS and the NF-ÎșB reporter mice expressed luciferase in the lungs following LPS challenge, establishing that bioluminescence imaging provides adequate sensitivity for tracking the NF-ÎșB activation pathway. Interventions affecting the NF-ÎșB pathway are promising clinical therapeutics, thus we further examined the effect of IKK-2 inhibition by MLN120B and glycogen synthase kinase 3 beta inhibition by TDZD-8 on NF-ÎșB activation. Pre-treatment with either MLN120B or TDZD-8 attenuated NF-ÎșB activation in the pulmonary tissues, which was accompanied with suppression of pro-inflammatory chemokine MIP-1ß and induction of anti-inflammatory cytokine IL-10. In summary, we have established an imaging based approach for non-invasive and longitudinal assessment of NF-ÎșB activation and regulation during acute lung injury. This approach will potentiate further studies on NF-ÎșB regulation under various inflammatory conditions

    Elevated c-Src is linked to altered cell–matrix adhesion rather than proliferation in KM12C human colorectal cancer cells

    Elevated expression and/or activity of c-Src, the prototype of the Src family of protein tyrosine kinases, is associated with the development of human colon cancer. However, despite the known pleiotropic effects of these kinases in promoting (a) cell growth downstream of growth factor receptors, and (b) the dynamic regulation of integrin adhesions in fibroblast model systems, their precise role in epithelial cancer cells is unknown. Here we addressed whether elevated expression and activity of cellular Src alters cell proliferation and/or cell–matrix adhesion in cancer cells from the Fidler model of colorectal metastasis. Although elevated Src correlates with ability to metastasise to the liver after intrasplenic injection, we found that this was not linked to enhanced growth, either in vitro or in vivo as sub-cutaneous tumours. However, elevated Src was associated with enhanced attachment to extracellular matrix. In addition, adhesion to fibronectin, was suppressed by agents that inhibited Src activity, while enforced elevation of Src in non-metastatic cells was sufficient to stimulate adhesion to fibronectin and enhanced assembly of adhesion complexes, without influencing cell growth. Thus, we conclude that one role of elevated Src in human colon cancer cells is to modulate integrin-dependent cell–matrix attachment and formation of adhesion structures, which may, in turn, influence cell motility and integrin-dependent cellular responses

    Communities and other Social Structures for Knowledge Sharing — A Case Study in an Internet Consultancy Company

    Abstract. This research aims at understanding how people share knowledge in their everyday work in a project-based company. The social structures for knowledge sharing are characterised as formal, informal, and quasi-informal structures. They vary from those with high formalisation to the informal, and even include structures which are invisible and unrecognised in the organisation. They also vary in their composition. They may share the same or different space, and communication is based on face-to-face or virtual interaction. Data was collected by means of documents and interviews (n=18) during the autumn of 2002 and the winter of 2003 from an Internet consultancy company. The study shows the great variety of formal, informal, and quasi-informal social structures that are used for knowledge sharing in the case company. In all, sixteen different structures were found. The number of formal structures is smaller than the number of informal ones. Their analysis in terms of five dimensions also shows their great heterogeneity
