    Metabolomics for mitochondrial and cancer studies

    AbstractMetabolomics, a high-throughput global metabolite analysis, is a burgeoning field, and in recent times has shown substantial evidence to support its emerging role in cancer diagnosis, cancer recurrence, and prognosis, as well as its impact in identifying novel cancer biomarkers and developing cancer therapeutics. Newly evolving advances in disease diagnostics and therapy will further facilitate future growth in the field of metabolomics, especially in cancer, where there is a dire need for sensitive and more affordable diagnostic tools and an urgency to develop effective therapies and identify reliable biomarkers to predict accurately the response to a therapy. Here, we review the application of metabolomics in cancer and mitochondrial studies and its role in enabling the understanding of altered metabolism and malignant transformation during cancer growth and metastasis. The recent developments in the area of metabolic flux analysis may help to close the gap between clinical metabolomics research and the development of cancer metabolome. In the era of personalized medicine with more and more patient specific targeted therapies being used, we need reliable, dynamic, faster, and yet sensitive biomarkers both to track the disease and to develop and evolve therapies during the course of treatment. Recent advances in metabolomics along with the novel strategies to analyze, understand, and construct the metabolic pathways opens this window of opportunity in a very cost-effective manner.     Industry 4.0 Asset Administration Shell (AAS): Interoperable Skill-Based Service-Robots

    This paper describes our use of Industry 4.0 Asset Administration Shells (AASs) in the context of service robots. We use AASs with software components of service robots and with complete service robot systems. The AAS for a software component serves as a standardized digital data sheet. It helps sysem builders at design time in finding and selecting software components that match system-level requirements of the systems to be built. The AAS for a system comprises a data sheet for the system and furthermore collects at runtime operational data and it allows for skill-level commanding of the service robot. AASs are generated and filled as part of our model-driven development and composition workflow for service robotics.     Metabolic Profiling Based Quantitative Evaluation of Hepatocellular Metabolism in Presence of Adipocyte Derived Extracellular Matrix

    The elucidation of the effect of extracellular matrices on hepatocellular metabolism is critical to understand the mechanism of functional upregulation. We have developed a system using natural extracellular matrices [Adipogel] for enhanced albumin synthesis of rat hepatocyte cultures for a period of 10 days as compared to collagen sandwich cultures. Primary rat hepatocytes isolated from livers of female Lewis rats recover within 4 days of culture from isolation induced injury while function is stabilized at 7 days post-isolation. Thus, the culture period can be classified into three distinct stages viz. recovery stage [day 0–4], pre-stable stage [day 5–7] and the stable stage [day 8–10]. A Metabolic Flux Analysis of primary rat hepatocytes cultured in Adipogel was performed to identify the key metabolic pathways modulated as compared to collagen sandwich cultures. In the recovery stage [day 4], the collagen-soluble Adipogel cultures shows an increase in TriCarboxylic Acid [TCA] cycle fluxes; in the pre-stable stage [day 7], there is an increase in PPP and TCA cycle fluxes while in the stable stage [day 10], there is a significant increase in TCA cycle, urea cycle fluxes and amino acid uptake rates concomitant with increased albumin synthesis rate as compared to collagen sandwich cultures throughout the culture period. Metabolic analysis of the collagen-soluble Adipogel condition reveals significantly higher transamination reaction fluxes, amino acid uptake and albumin synthesis rates for the stable vs. recovery stages of culture. The identification of metabolic pathways modulated for hepatocyte cultures in presence of Adipogel will be a useful step to develop an optimization algorithm to further improve hepatocyte function for Bioartificial Liver Devices. The development of this framework for upregulating hepatocyte function in Bioartificial Liver Devices will facilitate the utilization of an integrated experimental and computational approach for broader applications of Adipogel in tissue e engineering and regenerative medicine

    Determinants of Sugar-Sweetened Beverage Consumption among Indian Adults: Findings from the National Family Health Survey-4

    BACKGROUND: Sugar-sweetened beverages (SSBs) are associated with obesity, and various other noncommunicable diseases (NCDs). The aim of the study was to study the patterns of consumption of SSBs and association of SSB consumption with various socioeconomic factors and fried food consumption. METHODOLOGY: We used data of the 4th round of National Family Health Survey. We used multiple logistic regression to estimate the extent of the relationship between consumption of aerated drinks and various predictors. Furthermore, generalized structural equation modeling (GSEM) was used to derive a path diagram that showed a significant linkage between aerated drinks and observed variables. RESULTS: Our study showed a clear association between consumption of aerated drinks with socioeconomic variables age, sex, marital status, and wealth index. The consumption of aerated drinks was also significantly associated with watching television and eating fried foods. CONCLUSION: Aerated drinks are a popular source of added sugar in the Indian diet. Limiting such factors can prove to be beneficial in reducing their consumption and further help in reducing the burden of NCDs

    Propane to Acrylic Acid

    Acrylic acid is an essential polymer raw material for many industrial and consumer products. Currently, acrylic acid is manufactured from propylene, which is created as a by- product from fossil fuels manufacture and industrial cracking of heavy hydrocarbons. However, the discovery of new natural gas reserves presents new opportunities for the production of acrylic acid. A design feasibility study is presented to analyze the economics behind producing acrylic acid from the selective oxidation of propane to propylene over a mixed metal oxide catalyst, Mo1V0.30Te0.23Nb0.125Ox. The proposed plant is located in the U.S. Gulf Coast and produces 200MM lb/yr acrylic acid. Since the catalytic oxidation process has low propane conversion per pass, the process recycles unconverted propane and propylene back to the reactor to increase overall conversion. The acrylic acid is then separated and purified to the glacial-grade industrial standard for polymer raw material of 99.7% acrylic acid by mass. A major challenge of the separation process is the non-ideal behavior of the components, which produces three different azeotropes: water with acrylic acid, water with acetic acid, and acetic acid with acrylic acid. The separation process utilizes four distillation towers to navigate around the azeotropes. After a thorough economic analysis, the proposed process is found to be economically viable. It has an estimated IRR of 84.9% and NPV of 384,963,400ata15384,963,400 at a 15% discount rate using an acrylic acid price of 1.75/lb. The process is predicted to become profitable in year 3. If the product price decreases by 45% to 1.20/lb(thecurrentmarketpriceofacrylicacid),theestimatedIRRwillbe451.20/lb (the current market price of acrylic acid), the estimated IRR will be 45% with a NPV of 114,552,700 at a 15% discount rate. The process will then become profitable in year 4

    Inertial Focusing of Particles in Curved Micro-channels

    Inertial focusing is the migration of particles in flow laterally across a channel into well-defined equilibrium positions. In microfluidic channels, inertial focusing takes advantage of hydrodynamic interactions even at high flow speeds. Particle isolation through inertial focusing is a high throughput method of processing biological samples for point-of-care diagnostics. While photos provide qualitative analyses of inertial focusing, we desired quantitative characterization of these systems. In this study, we ran flow experiments, first with fluorescent polystyrene beads and later with cells in solution, through curved micro-channels at controlled rates using a syringe pump. Our results from polystyrene bead experiments confirmed previous studies on flow through curved micro-channels, in which particles are focused along both sides of the channel at low flow rates and transition towards the center of the channel as the flow rate increases. FWHM analysis also showed that the streamline width is minimized at an intermediate flow rate, indicating inertial focusing is optimized under that condition. As this method of analysis was confirmed with polystyrene beads, we further used this analysis method to characterize the focusing of cells in solution. To maximize both throughput and purity, microfluidic devices must be designed to operate at the highest flow rate at which effective separation from bulk fluid can occur. Será discutido o que leva parlamentares considerarem a manutenção de seus cargos no Congresso Nacional. Variáveis como números de mandatos, estado (os) que representam/representaram, nível de escolaridade, atividade profissional fora do Congresso Nacional e Comissões que integram ou integraram foram levados em consideração para encontrar relação entre esses dados e a teoria informacional
