    Protein composition of interband regions in polytene and cell line chromosomes of Drosophila melanogaster

    <p>Abstract</p> <p>Background</p> <p>Despite many efforts, little is known about distribution and interactions of chromatin proteins which contribute to the specificity of chromomeric organization of interphase chromosomes. To address this issue, we used publicly available datasets from several recent Drosophila genome-wide mapping and annotation projects, in particular, those from modENCODE project, and compared molecular organization of 13 interband regions which were accurately mapped previously.</p> <p>Results</p> <p>Here we demonstrate that in interphase chromosomes of <it>Drosophila </it>cell lines, the interband regions are enriched for a specific set of proteins generally characteristic of the "open" chromatin (RNA polymerase II, CHRIZ (CHRO), BEAF-32, BRE1, dMI-2, GAF, NURF301, WDS and TRX). These regions also display reduced nucleosome density, histone H1 depletion and pronounced enrichment for ORC2, a pre-replication complex component. Within the 13 interband regions analyzed, most were around 3-4 kb long, particularly those where many of said protein features were present. We estimate there are about 3500 regions with similar properties in chromosomes of <it>D. melanogaster </it>cell lines, which fits quite well the number of cytologically observed interbands in salivary gland polytene chromosomes.</p> <p>Conclusions</p> <p>Our observations suggest strikingly similar organization of interband chromatin in polytene chromosomes and in chromosomes from cell lines thereby reflecting the existence of a universal principle of interphase chromosome organization.</p

    Salivary gland polytene chromosomes demonstrate banding pattern, genetic meaning of which is an enigma for decades. Till now it is not known how to mark the band/interband borders on physical map of DNA and structures of polytene chromosomes are not characterized in molecular and genetic terms. It is not known either similar banding pattern exists in chromosomes of regular diploid mitotically dividing nonpolytene cells. Using the newly developed approach permitting to identify the interband material and localization data of interband-specific proteins from modENCODE and other genome-wide projects, we identify physical limits of bands and interbands in small cytological region 9F13-10B3 of the X chromosome in D. melanogaster, as well as characterize their general molecular features. Our results suggests that the polytene and interphase cell line chromosomes have practically the same patterns of bands and interbands reflecting, probably, the basic principle of interphase chromosome organization. Two types of bands have been described in chromosomes, early and late-replicating, which differ in many aspects of their protein and genetic content.     CHARACTERISTICS OF AUTOIMMUNE INFLAMMATION IN THE PATIENTS WITH LUNG TUBERCULOSIS

    Tuberculosis is a granulomatous disease caused by Mycobacterium tuberculosis, being characterized by the development of caseous granulomas in various organs, mainly in lungs. M. tuberculosis is known to be a trigger for autoimmune inflammation, due to the possible mimicry of bacterial proteins as autoantigens. Recently, a significance of mesenchymal vimentin as an autoantigen in mycobacterial infections has been actively discussed. The aim of the present study was to determine autoantibodies for various vimentin modifications in the patients with tuberculosis.The study was performed in 2014-2017 and included 28 patients with pulmonary tuberculosis (group I), 30 patients with nonspecific lung diseases (group II): 15 with granulomatous polyangiitis, and 15 with different alveolites. Control group consisted of healthy subjects (n = 40). Concentration of antibodies to mutated citrullinated vimentin (anti-MCV) was measured using ELISA (ORGENTEC, Germany). The patients with elevated anti-MCV levels were tested for antibodies to cyclic citrullinated peptide (anti-CCP) using ELISA technique (EUROIMMUN, Germany). Statistical analysis was carried out using GraphPad Prism 6 (GraphPad Software, USA), Statistica 10 (Statsoft, USA) using nonparametric analysis of samples with Mann-Whitney and Chi-square criteria, and Spearman method for correlation analysis. The differences were considered statistically significant at p &lt; 0.05.The anti-MCV concentrations were significantly higher in patients with tuberculosis (group I, 60.7% of cases, 17/28) than in group II, and control group (23.6 and 25.0% of cases, respectively). No statistically significant differences were revealed between the results of anti-MVC and anti-CCP levels in comparison group with the control group (p = 0.18).High levels of anti-MCV antibodies in the patients with pulmonary tuberculosis reflect an opportunity of developing autoimmune process in the disease pathogenesis. Measurement of plasma anti-MCV antibody concentrations may be important for correction of the therapy, especially upon administration of immunosuppressive and hormonal corticosteroid drugs. It has been shown that anti-CCP are not characteristic to the lung diseases

    Effect of electron irradiation on vortex dynamics in YBa_2Cu_3O_{7-x} single crystals

    We report on drastic change of vortex dynamics with increase of quenched disorder: for rather weak disorder we found a single vortex creep regime, which we attribute to a Bragg-glass phase, while for enhanced disorder we found an increase of both the depinning current and activation energy with magnetic field, which we attribute to entangled vortex phase. We also found that introduction of additional defects always increases the depinning current, but it increases activation energy only for elastic vortex creep, while it decreases activation energy for plastic vortex creep.Comment: 4 pages, 3 figures, submited to Phys.     Assessment of the course of the gestational process using survey method and depending on the laboratory confirmed prenatal alcohol use (cross-section study)

    Background. Studying the problem of alcohol consumption by pregnant women using modern laboratory research methods has valuable theoretical and practical significance. The aim of the study. To determine the characteristics of the pregnancy course in women consuming alcohol in the prenatal period confirmed by survey and laboratory tests, depending on the phosphatidylethanol levels. Materials and methods. We examined 863 women under observation at the Irkutsk Regional Perinatal Center for the period from 2014 to 2021. To confirm the fact of alcohol consumption in the prenatal period, 545 women were surveyed, 318  women were examined using laboratory analysis. The diagnostic biomarker for  alcohol was  PEth:16:0/18:1. To assign pregnant women to  the control group, a PEth concentration of ≤ 8 ng/ml was taken. If the PEth concentration was &gt; 8 ng/ml, pregnant women were classified as heavy drinkers. Clinical and laboratory indicators of the course of pregnancy and childbirth were carried out in comparative groups. Results. It has been established that every second woman of reproductive age took alcohol before pregnancy. 24.2 % of women did not stop consuming alcohol in  the  prenatal period. At the same time, the risk of congenital malformations was high, since 20.4 % of women consumed alcohol in the first trimester of pregnancy. Based on the results of the survey, it was revealed that in women who consumed alcohol in the prenatal period, the following pathological conditions are statistically significantly more common: anemia, congenital heart defects in fetuses, prematurity of gestational age, labor anomalies, uterus subinvolution. Based on the results of laboratory confirmation of alcohol consumption, it was established that parity of birth, intrauterine growth retardation, and  premature birth were statistically significantly more often in pregnant women who drink. Conclusion. Thus, in order to obtain the most meaningful and high-quality results, it is necessary to  conduct larger studies. In addition, maternal blood biomarkers should be used to confirm levels of alcohol consumption throughout all trimesters of pregnancy

    Radial-shear rolling is prominent in production of titanium alloys at «VSMPO-AVISMA» Open Society (V. Salda city) and has a number of advantages over other plastic metal working processes. In the present investigation on the basis of engineering design procedures, a complex of software has been developed for forming simulation and heat transfer in the metal volume,and with its help the rolling of VT-6 alloy bar of 155 mm in diameter has been studied. «ThermoRSR» program has been developed that allows us to visualize design data in «Mathcad» medium; rolling process simulation has been carried out by means of DEFORM-3D package; and the conditions of its application are shown. Prospective ways of rolled stock temperature and rolling torque measurement are considered.Радиально-сдвиговая прокатка (РСП) играет большую роль в производстве титановых сплавов в ОАО «ВСМПО-АВИСМА» (г. В. Салда), имея ряд преимуществ перед другими процессами обработки металлов давлением. В настоящем исследовании на базе инженерных методик расчета разработан комплекс программных средств для моделирования формоизменения и теплопередачи в объеме металла и с его помощью изучена прокатка прутка диаметром 155 мм из сплава ВТ-6. Разработана программа «ThermoRSR», позволяющая визуализировать расчетные данные в среде «Mathcad», проведено моделирование процесса прокатки с помощью пакета DEFORM-3D и показаны условия его применимости. Рассмотрены перспективные способы измерения температуры проката и крутящего момента на шпинделе прокатного стана
