    Clozapine as add-on medication in the maintenance treatment of bipolar and schizoaffective disorders - A case series

    Atypical neuroleptics are increasingly used in the treatment of bipolar and schizoaffective disorders. Currently, numerous controlled short-term studies are available for clozapine, olanzapine, risperidone or quetiapine, but long-term data are still missing. Three patients (2 with bipolar disorder, 1 with schizoaffective disorder) are described who showed a marked reduction of affective symptomatology after clozapine had been added to mood stabilizer pretreatment. The patients were seen once a month before and after the introduction of clozapine for at least 6 months. Treatment response was evaluated using different rating scales (IDS, YMRS; GAF; CGIBP) and the NIMH Life Chart Methodology. All patients showed a marked improvement after the add-on treatment with clozapine had been initiated. Clozapine was tolerated well with only transient and moderate weight gain and fatigue as only side effects. This case series underlines the safety and efficacy of clozapine as add-on medication in the treatment of bipolar and schizoaffective disorders. Copyright (C) 2002 S. Karger AG, Basel

    Calorimetric study of water's two glass transitions in the presence of LiCl

    A DSC study of dilute glassy LiCl aqueous solutions in the water-dominated regime provides direct evidence of a glass-to-liquid transition in expanded high density amorphous (eHDA)-type solutions. Similarly, low density amorphous ice (LDA) exhibits a glass transition prior to crystallization to ice Ic. Both glass transition temperatures are independent of the salt concentration, whereas the magnitude of the heat capacity increase differs. By contrast to pure water, the glass transition endpoint for LDA can be accessed in LiCl aqueous solutions above 0.01 mole fraction. Furthermore, we also reveal the endpoint for HDA's glass transition, solving the question on the width of both glass transitions. This suggests that both equilibrated HDL and LDL can be accessed in dilute LiCl solutions, supporting the liquid-liquid transition scenario to understand water's anomalies.Fil: Ruiz, Guadalupe N..     GHG Mitigation Potentials in Annex I Countries. Comparison of Model Estimates for 2020

    Robust quantification of the future potentials and costs for mitigating greenhouse gases in different countries could provide important information to the current negotiations on a post-2012 climate agreement. However, such information is not readily available from statistical sources, but requires the use of complex models that combine economic, technological and social aspects. In March 2009, the International Institute for Applied Systems Analysis (IIASA) invited leading modelling teams to a comparison of available model estimates of GHG mitigation potentials and costs in the Annex I countries for the year 2020. Eight modelling teams provided input to this comparison exercise. Although at face value estimates of mitigation potentials and costs show wide variation across models differences (i) in assumptions on the baseline economic development, (ii) in the definition of which mitigation measures are considered part of the baseline, and (iii) in the time window assumed for the implementation of mitigation measures explain much of the variation in model results. The paper presents a check-list of factors that need to be considered when interpreting model results. Once corrected for these key factors, two clusters of cost curves emerge for the year 2020: Models that include consumer demand changes and macro-economic feedbacks agree on a mitigation potential of up to 40% reduction below 2005 levels (that is approximately 45% below the 1990 level) for total Annex I emissions in 2020 for a carbon price of 50 to 150 US-$/tCO2. Bottom-up models that restrict their analysis to technical measures show only half of this potential. The model intercomparison demonstrates that future economic development has a strong impact on the eforts necessary to achieve given emission reduction levels. Any delay in the start of implementation of mitigation measures will reduce the mitigation potential that is achievable in the near term and inrease the costs. The introduction of measures that mobilize demand adjustments through structural or behavioural changes may increase the short-term mitigation potential significantly

    Novel potential interacting partners of fibronectin in spontaneous animal model of interstitial cystitis

    Feline idiopathic cystitis (FIC) is the only spontaneous animal model for human interstitial cystitis (IC), as both possess a distinctive chronical and relapsing character. Underlying pathomechanisms of both diseases are not clearly established yet. We recently detected increased urine fibronectin levels in FIC cases. The purpose of this study was to gain further insight into the pathogenesis by assessing interacting partners of fibronectin in urine of FIC affected cats. Several candidate proteins were identified via immunoprecipitation and mass spectrometry. Considerable changes in FIC conditions compared to physiological expression of co-purified proteins were detected by Western blot and immunohistochemistry. Compared to controls, complement C4a and thioredoxin were present in higher levels in urine of FIC patients whereas loss of signal intensity was detected in FIC affected tissue. Galectin-7 was exclusively detected in urine of FIC cats, pointing to an important role of this molecule in FIC pathogenesis. Moderate physiological signal intensity of galectin-7 in transitional epithelium shifted to distinct expression in transitional epithelium under pathophysiological conditions. I-FABP expression was reduced in urine and urinary bladder tissue of FIC cats. Additionally, transduction molecules of thioredoxin, NF-κB p65 and p38 MAPK, were examined. In FIC affected tissue, colocalization of thioredoxin and NF-κB p65 could be demonstrated compared to absent coexpression of thioredoxin and p38 MAPK. These considerable changes in expression level and pattern point to an important role for co-purified proteins of fibronectin and thioredoxin-regulated signal transduction pathways in FIC pathogenesis. These results could provide a promising starting point for novel therapeutic approaches in the future

    Bistability and all-optical memory in dual-mode diode lasers with time-delayed optical feedback

    A proposal for an all-optical memory based on a bistability of single-mode states in a dual-mode diode laser with time-delayed optical feedback is presented. The system is modeled using a multimode extension of the Lang-Kobayashi equations with injected optical pulses. We uncover the bifurcation structure by deriving analytical expressions for the boundaries of the bistable region and demonstrate how the delay time in the external cavity determines an optimal pulse duration for efficient switching of the memory element. We also show the relevant role played by gain saturation and by the dual-mode solutions of the Lang-Kobayashi equations for the existence of the bistable regions. Our results demonstrate that feedback induced bistability can lead to significant performance improvements when compared to memory elements based on the injection locking bistability in dual-mode devices

    United Nations Secretary General Ban Ki-moon has invited world leaders to come to the Climate Summit on September 23, 2014 to deliver "bold pledges" to tackle climate change. This paper was prepared at the request of the Republic of Nauru, Chair of the Alliance of Small Island States, as part of their answer to that call. We believe the path to the global low-carbon transformation needed to tackle the climate crisis is within reach, but requires decisive political action from leaders around the world, now. This paper is unabashedly prescriptive on the need for action, but recognizes that there are multiple approaches and models from around the world that can be scaled up and adapted to national circumstances. Cost-effective technologies for a low-carbon economy are being implemented throughout the world, but at nowhere the scale and speed necessary. Emissions continue to rise. With every year of delay, human suffering, biodiversity loss, and the costs of mitigation and adaptation increase. We are running out of time

    Suspension cultures of Berberis species are useful sources for the detection and isolation of a new enzyme which transfers the methyl group from S-adenosyl-L-methionine specifically to the 9-position of the (S)-enantiomer of scoulerine, producing (S)-tetrahydrocolumbamine. The enzyme was enriched 27-fold; it is not particle bound, has a pH optimum of 8.9, a molecular weight of 63 000 and shows a high degree of substrate specificity
