    Detection of Sugar-Lectin Interactions by Multivalent Dendritic Sugar Functionalized Single-Walled Carbon Nanotubes

    We show that single walled carbon nanotubes (SWNT) decorated with sugar functionalized poly (propyl ether imine) (PETIM) dendrimer is a very sensitive platform to quantitatively detect carbohydrate recognizing proteins, namely, lectins. The changes in electrical conductivity of SWNT in field effect transistor device due to carbohydrate - protein interactions form the basis of present study. The mannose sugar attached PETIM dendrimers undergo charge - transfer interactions with the SWNT. The changes in the conductance of the dendritic sugar functionalized SWNT after addition of lectins in varying concentrations were found to follow the Langmuir type isotherm, giving the concanavalin A (Con A) - mannose affinity constant to be 8.5 x 106 M-1. The increase in the device conductance observed after adding 10 nM of Con A is same as after adding 20 \muM of a non - specific lectin peanut agglutinin, showing the high specificity of the Con A - mannose interactions. The specificity of sugar-lectin interactions was characterized further by observing significant shifts in Raman modes of the SWNT.Comment: 12 pages, 3 figure

    Garlic improves insulin sensitivity and associated metabolic syndromes in fructose fed rats

    <p>Abstract</p> <p>Background</p> <p>Type 2 diabetes mellitus, characterized by peripheral insulin resistance, is a major lifestyle disorder of the 21<sup>st </sup>Century. Raw garlic homogenate has been reported to reduce plasma glucose levels in animal models of type 1 diabetes mellitus. However, no specific studies have been conducted to evaluate the effect of raw garlic on insulin resistance or type 2 diabetes mellitus. This study was designed to investigate the effect of raw garlic on fructose induced insulin resistance, associated metabolic syndrome and oxidative stress in diabetic rats.</p> <p>Methods</p> <p>Male Sprague Dawley rats weighing 200-250 gm body weight were divided into 3 groups (n = 7 per group) and fed diet containing 65% cornstarch (Control group) and 65% fructose (Diabetic group) for 8 weeks. The third group (Dia+Garl group) was fed both 65% fructose and raw garlic homogenate (250 mg/kg/day) for 8 weeks. Whole garlic cloves were homogenized with water to make a fresh paste each day.</p> <p>Results</p> <p>At the end of 8 weeks, serum glucose, insulin, triglyceride and uric acid levels, as well as insulin resistance, as measured by glucose tolerance test, were significantly (p < 0.01) increased in fructose fed rats (Diabetic group) when compared to the cornstarch fed (Control) rats. Administration of raw garlic to fructose fed rats (Dia+Garl group) significantly (p < 0.05) reduced serum glucose, insulin, triglyceride and uric acid levels, as well as insulin resistance when compared with fructose fed rats. Garlic also normalised the increased serum levels of nitric oxide (NO) and decreased levels of hydrogen sulphide (H<sub>2</sub>S) after fructose feeding. Although body weight gain and serum glycated haemoglobin levels of fructose fed rats (Diabetic group) were not significantly different from control rats, significant (p < 0.05) reduction of these parameters was observed in fructose fed rats after garlic administration (Dia+Garl group). Significant (p < 0.05) increase in TBARS and decrease in GSH was observed in diabetic liver. Catalase was not significantly affected in any of the groups. Administration of raw garlic homogenate normalised both hepatic TBARS and GSH levels.</p> <p>Conclusions</p> <p>Our study demonstrates that raw garlic homogenate is effective in improving insulin sensitivity while attenuating metabolic syndrome and oxidative stress in fructose-fed rats.</p

    Incisional hernia following colorectal cancer surgery according to suture technique: Hughes Abdominal Repair Randomized Trial (HART).

    BACKGROUND: Incisional hernias cause morbidity and may require further surgery. HART (Hughes Abdominal Repair Trial) assessed the effect of an alternative suture method on the incidence of incisional hernia following colorectal cancer surgery. METHODS: A pragmatic multicentre single-blind RCT allocated patients undergoing midline incision for colorectal cancer to either Hughes closure (double far-near-near-far sutures of 1 nylon suture at 2-cm intervals along the fascia combined with conventional mass closure) or the surgeon's standard closure. The primary outcome was the incidence of incisional hernia at 1 year assessed by clinical examination. An intention-to-treat analysis was performed. RESULTS: Between August 2014 and February 2018, 802 patients were randomized to either Hughes closure (401) or the standard mass closure group (401). At 1 year after surgery, 672 patients (83.7 per cent) were included in the primary outcome analysis; 50 of 339 patients (14.8 per cent) in the Hughes group and 57 of 333 (17.1 per cent) in the standard closure group had incisional hernia (OR 0.84, 95 per cent c.i. 0.55 to 1.27; P = 0.402). At 2 years, 78 patients (28.7 per cent) in the Hughes repair group and 84 (31.8 per cent) in the standard closure group had incisional hernia (OR 0.86, 0.59 to 1.25; P = 0.429). Adverse events were similar in the two groups, apart from the rate of surgical-site infection, which was higher in the Hughes group (13.2 versus 7.7 per cent; OR 1.82, 1.14 to 2.91; P = 0.011). CONCLUSION: The incidence of incisional hernia after colorectal cancer surgery is high. There was no statistical difference in incidence between Hughes closure and mass closure at 1 or 2 years.     Ionic conductivity of bis(2-cyanoethyl) ether-lithium salt and poly(propylether imine)-lithium salt liquid electrolytes

    We report here a multiple-nitrile based lithium-salt liquid electrolyte. The ionic conductivity of poly (propyl ether imine) (abbreviated as PETIM) lithium salt dendrimer liquid electrolyte was observed to be a function of dendrimer generation number, n = 0 (monomer)−3. While the highest room temperature ionic conductivity value (&#8764; 10<sup>−1</sup> Sm<sup>−1</sup>) was recorded for the bis-2cyanoethyl ether monomer (i.e. zeroth generation; G<sub>0</sub>-CN), conductivity decreased progressively to lower values (&#8764; 10<sup>−3</sup> Sm<sup>−1</sup>) with increase in generation number (G<sub>1</sub>-CN &#8594; G<sub>3</sub>-CN). The G<sub>0</sub>-CN and higher dendrimer generations showed high thermal stability (&#8776;150 to 200 &#176;C), low moisture sensitivity and tunable viscosity (&#8764;10<sup>−2</sup> (G<sub>0</sub>-CN) to 3 (G<sub>3</sub>-CN) Pa s). The linker ether group was found to be crucial for ion transport and also eliminated a large number of detrimental features, chiefly moisture sensitivity, chemical instability associated typically with prevalent molecular liquid solvents. Based on the combination of several beneficial physicochemical properties, we presently envisage that the PETIM dendrimers especially the G<sub>0</sub>-CN electrolytes hold promise as electrolytes in electrochemical devices such as lithium-ion batteries

    Assessment of collagen fiber nature, spatial distribution, hue and its correlation with invasion and metastasis in oral squamous cell carcinoma and surgical margins using Picro Sirius red and polarized microscope

    Introduction: Oral squamous cell carcinoma (OSCC) comprises a bulk of all the oral malignancies and is posing a major health problem among the population. It is an established fact that tumor stroma plays a vital role in tumor progression. Therefore, methods to detect, quantify, and analyze collagen are of immense value in this regard. Picro Sirius red, which has the capability to detect thin fibers, although frequently used, is seldom exploited to the fullest extent. Aim: Our goal is not only to identify nature of fibers, but also to assess the fiber hue and the spatial distribution of different colors in various grades of OSCC and correlate it with the metastasis of the tumor. The study has also analyzed the nature of stromal elements along the clear, close and involved surgical margins of OSCC. Materials and Methods: Ten cases each of well, moderately and poorly differentiated OSCC as well as clear, close and involved margins were stained with haematoxylin eosin and picrosirius red staining for evaluation under polarized microscope. Results: In this study we found that the birefringence of the collagen fiber changed from orange red to yellowish green from well to poorly differentiated OSCC. The collagen fibers in well-differentiated carcinoma revealed polarizing colors of reddish orange around the tumor islands in the majority of the fields. To the best of our knowledge is not being studied so far in the English literature. Conclusion: In the present study, it has been observed that stromal changes at the invading front of the tumor islands and with increasing grade of the tumor can be evaluated more efficiently with the use of Picro Sirius red stain
