    Experimental Study of Photonuclear Reactions of ^4He below Pion Threshold(I. Nuclear Physics)

    An experimental method for the precise measurement of the photonuclear reactions of ^4He below the pion threshold has been tested. We used a tagged photon beam and a time projection chamber containing helium gas, which served as an active target. It was proved that the chamber could successfully detect the tracks of the charged particles from the photonuclear reactions in a high radiation level due to the irradiation of a high-intensity photon beam. It was found that the background was mainly due to the noise of the chamber, and could be suppressed by taking coincidence of the signals from the chamber and the tagging counter

    Asthma Pregnancy Alters Postnatal Development of Chromaffin Cells in the Rat Adrenal Medulla

    Background: Adrenal neuroendocrine plays an important role in asthma. The activity of the sympathoadrenal system could be altered by early life events. The effects of maternal asthma during pregnancy on the adrenal medulla of offspring remain unknown. Methodology/Principal Findings: This study aims to explore the influence of maternal asthma during pregnancy on the development and function of adrenal medulla in offspring from postnatal day 3 (P3) to postnatal day 60 (P60). Asthmatic pregnant rats (AP), nerve growth factor (NGF)-treated pregnant rats (NP) and NGF antibody-treated pregnant rats (ANP) were sensitized and challenged with ovalbumin (OVA); NP and ANP were treated with NGF and NGF antibody respectively. Offspring rats from the maternal group were divided into four groups: offspring from control pregnant rats (OCP), offspring from AP (OAP), offspring from NP (ONP), and offspring from ANP (OANP). The expressions of phenylethanolamine N-methyltransferase (PNMT) protein in adrenal medulla were analyzed. The concentrations of epinephrine (EPI), corticosterone and NGF in serum were measured. Adrenal medulla chromaffin cells (AMCC) were prone to differentiate into sympathetic nerve cells in OAP and ONP. Both EPI and PNMT were decreased in OAP from P3 to P14, and then reached normal level gradually from P30 to P60, which were lower from birth to adulthood in ONP. Corticosterone concentration increased significantly in OAP and ONP. Conclusion/Significance: Asthma pregnancy may promote AMCC to differentiate into sympathetic neurons in offspring rats and inhibit the synthesis of EPI, resulting in dysfunction of bronchial relaxation

    Validation of the Burden Index of Caregivers (BIC), a multidimensional short care burden scale from Japan

    BACKGROUND: We constructed a concise multidimensional care burden scale that reflects circumstances unique to Japan, with a focus on intractable neurological diseases. We surveyed 646 family caregivers of patients with intractable neurological diseases or stroke using 28 preliminary care burden scale items obtained from qualitative research. The results were used to finalize the feeling of care burden scale (BIC: burden index of caregivers), and verify its reliability and validity. METHODS: The survey was conducted among caregivers providing home health care to patients with intractable neurological diseases (PD [Parkinson's disease], SCD [spinocerebellar degeneration], MSA [multiple system atrophy], and ALS [amyotrophic lateral sclerosis]) or CVA (cerebrovascular accident) using a mailed, self-administered questionnaire between November, 2003 and May, 2004. RESULTS: Response rates for neurological and CVA caregivers were 50% and 67%, respectively, or 646 in total (PD, 279; SCD, 78; MSA, 39; ALS, 30; and CVA, 220). Item and exploratory factor analyses led to a reduction to 11 items, comprising 10 items from the 5 domains of time-dependent burden, emotional burden, existential burden, physical burden, and service-related burden; and 1 item on total burden. Examination of validity showed a moderate correlation between each domain of the BIC and the SF-8 (Health related quality of life scale, Short Form-8), while the correlation coefficient of the overall BIC and CES-D was 0.62. Correlation between the BIC and ZBI, a preexisting care burden scale, was high (r = 0.84), while that with the time spent on providing care was 0.47. The ICC (Intraclass correlation coefficient) by test-retest reliability was 0.83, and 0.68 to 0.80 by individual domain. CONCLUSION: These results show that the BIC, a new care burden scale comprising 11 items, is highly reliable and valid

    Internal Ribosomal Entry Site-Mediated Translation Is Important for Rhythmic PERIOD1 Expression

    The mouse PERIOD1 (mPER1) plays an important role in the maintenance of circadian rhythm. Translation of mPer1 is directed by both a cap-dependent process and cap-independent translation mediated by an internal ribosomal entry site (IRES) in the 5β€² untranslated region (UTR). Here, we compared mPer1 IRES activity with other cellular IRESs. We also found critical region in mPer1 5β€²UTR for heterogeneous nuclear ribonucleoprotein Q (HNRNPQ) binding. Deletion of HNRNPQ binding region markedly decreased IRES activity and disrupted rhythmicity. A mathematical model also suggests that rhythmic IRES-dependent translation is a key process in mPER1 oscillation. The IRES-mediated translation of mPer1 will help define the post-transcriptional regulation of the core clock genes

    Structural insights into Atg10-mediated formation of the autophagy-essential Atg12-Atg5 conjugate

Despite its importance, the molecular mechanism of the Atg12-Atg5 conjugate formation has not been elucidated. Here, we report the solution and crystal structures of Atg10 and Atg5 homologs from Kluyveromyces marxianus (Km), a thermotolerant yeast. KmAtg10 comprises an E2-core fold with characteristic accessories, including two Ξ² strands, whereas KmAtg5 has two ubiquitin-like domains and a helical domain. The nuclear magnetic resonance experiments, mutational analyses, and crosslinking experiments showed that KmAtg10 directly recognizes KmAtg5, especially its C-terminal ubiquitin-like domain, by its characteristic two Ξ² strands. Kinetic analysis suggests that Tyr56 and Asn114 ofΒ KmAtg10 may place the side chain of KmAtg5 Lys145 into the optimal orientation for its conjugation reaction with Atg12. These structural features enable Atg10 to mediate the formation of the Atg12-Atg5 conjugate without a specific E3 enzyme

    Fluctuation suppression during confinement potential formation in GAMMA 10

    The suppression of the potential fluctuation was clearly observed by a gold neutral beam probe (GNBP) during the higher potentials and positive electric fields driven by electron cyclotron heating. The particle flux evaluated from the phase difference between the potential and density fluctuations measured by GNBP showed the radial outward flux and correlates with the decrease in the stored energy. When flux is suppressed by the potential formation, the quick plasma stored energy increase is observed

    In the tandem mirror GAMMA 10, plasma confinement is achieved not only by the magnetic mirror configuration but also by the high potentials at both end regions. Before applying plug - electron cyclotron resonance heating (P-ECH), drift-type fluctuations were observed in the potential and density measurements. The suppression potential and density of these fluctuations were clearly determined by using a gold neutral beam probe (GNBP) system during the potential formation by the application of P-ECH. We studied the relationship between the suppression levels of the potential fluctuations and the effects of 'potentialproduced by changing the applied electron cyclotron heating power. Moreover, the particle-flux-related values obtained by examining the phase difference between the potential and density fluctuations were measured by using the GNBP. We clearly show that radial anomalous transport induces radial particle transport, which decreases the stored energy of the plasma
