    Penyaringan dan pemencilan kulat termofilik lipolitik telah dilakukan dari dua tempat iaitu kolam buangan sisa kelapa sawit dan tempat pembuangan sampah dengan menggunakan plat agar triolein . Enam strain kulat lipolitik telah dikenal pasti dengan adanya pembentukan zon keamatan di sekeliling miselia kulat tersebut. Walaubagaimanapun hanya dua strain sahaja yang menghasilkan lipase dalam media kaldu. Kulat-kulat ini dikenalpasti sebagai Rhizopus oryzae dan Rhizopus rhizopodiformis. Rhizopus oryzae didapati menghasilkan lipase ekstrasel pada aras yang maksimum selepas 72 jam pengeraman pada suhu 45'C di dalam media yang mengandungi pepton , pada pH 5 dan digoncang pada 100 psm .     New sequence variants in BRCA1 and BRCA2 genes detected by high-resolution melting analysis in an elderly healthy female population in Croatia

    Background: Mutations in BRCA1 and BRCA2 genes are associated with family predisposition to breast and ovarian cancer. Novel screening methods are required for efficient and rapid detection of sequence variants in cancer patients and their family members. Methods: The screening for variants in the breast and ovarian cancer susceptibility genes BRCA1 and BRCA2 in Croatia was performed by a high-resolution melting approach, which is based on differences in melting curves caused by variations in nucleotide sequence. This is the first screening in Croatia on elderly healthy women with no family history of cancer. BRCA1 screening was performed on 220 and BRCA2 on 115 samples. Results: In a population well beyond the average age of breast/ovarian cancer onset, 21 different sequence variants in the BRCA1 gene (one novel: c.5193q49_50delTA) and 36 variants in the BRCA2 gene (7 novel: c.459A)C, c.3318C)A, c.4412_ 4414delGAA, c.4790C)A, c.6264T)C, c.9087G)A, and c.9864A)G) were detected. Conclusions: Nine BRCA1 and seven BRCA2 known variants appeared with such high frequencies that they could be declared as harmless in this population. Eight BRCA1 high frequency variants, located further from the promoter region, appear to be strongly correlated. Three novel variants that changed the amino acid sequence of the BRCA2 protein (two missense base substitutions, c.3318C)A and c.4790C)A, and one codon deletion c.4412_4414delGAA), appearing only once, were predicted to have no potential effect on protein structure and function

Agama islam memiliki tiga aspek pokok yang harus diperhatikan, yaitu aspek Aqidah (keimanan), aspek Syariah (norma dan hukum), dan aspek Akhlak (moral atau budaya).     Extra- and intra-cellular lipases from a thermophilic Rhizopus oryzae and factors affecting their production

    A thermophilic Rhizopus oryzae was isolated, and parameters affecting its production of extra- and intra-cellular lipases were investigated. All carbon sources tested with the exception of sucrose generally inhibited the production of extracellular lipase, but enhanced the production of intracellular lipase. Peptone was the best substrate for extracellular enzyme production, but for intracellular lipase production other substrates such as tryptone, tryptic soy digest, polypeptone, and corn steep liquor gave comparable results. Among lipid substrates, glycerol was the only stimulator of extracellular enzyme production, whereas olive oil, triolein, and oleic acid had very positive effects on intracellular enzyme production. Shaking enhanced the production of both types of enzymes; the temperature optima were 45 and 37°C for extra- and intra-cellular lipases, respectively. A pH of 5.0 was optimal for production of both enzymes

    Some characteristics of lipases from thermophilic fungi isolated from palm oil mill effluent

    Rhizopus oryzae and Rhizopus rhizopodiformis are thermophilic fungi isolated from palm oil mill effluent (POME). They are able to grow up to 50°C and produce remarkable amounts of extracellular lipases. The extracellular lipases from both fungi displayed quite similar characteristics. The optimum pH and temperature for both lipases were 6.0 and 45°C, respectively. However, lipase from R. rhizopodiformis was slightly more thermostable than that of R. oryzae lipase but the latter was more stable over broader pH ranges compared to the former, specially at acidic pH. Both the enzyme showed rapid loss of activities at temperatures above 50°C and pH above 7.0. Lipase from R. oryzae showed broader specificities to substrates than R. rhizopodiformis, but both displayed little reactivity for triacetin and polyoxysorbitan (Tween 20, 40, and 60).
