Efficiency of Mechanical Harvest for Immature Vegetable Soybean Pods
Since soybean (Glycine max [L.] Merr.) is low in saturated fat and active in reducing blood cholesterol, it is gaining interest as a healthy snack food. Direct consumption of vegetable soybeans is very popular in the Orient. However, the cultivars used in Asia are not adapted to U.S. production systems. The objectives of this study were to determine the efficiency of mechanical harvest and to identify vegetable soybean cultivars adapted for a mechanical harvest system. To implement the objectives, four vegetable soybean cultivars were planted in a randomized complete block design at Randolph Research Farm, Virginia State University. The cultivars were hand harvested and mechanically harvested at the green pod stage and evaluated for green pod yield (kg ha-1), one hundred pod weight (g), plant height (cm), and pod dimensions of length (cm), width (cm), and thickness (cm). A significant difference (P \u3c 0.01) was observed among the two methods of harvesting. The hand harvested beans yielded twice as many more pods as the mechanical harvested beans. However, the pods harvested mechanically were cleaner and required no further cleaning as compared to hand harvested pods. There was also significant cultivar x method harvest interaction. The common bean picker was effective in harvesting the vegetable soybean cultivars with plant height of 55 to 66 cm and pod size that ranged from 128 to 144 g 100-1 pods. This type of operation could be easily adapted by farmers using appropriate cultivars
Haptic handwriting aid for training and rehabilitation
This paper reports a method of controlling a user\u27s hand through the process of writing. Developed predominantly for enabling users to re-learn the skill of writing after a stroke, the process could also be used for teaching children hand/eye coordination, motor skills, movement and position awareness in writing. Utilising low cost haptic technology and custom control software, the system has the potential to increase writing skills in stroke sufferers in the privacy and comfort of their own home. <br /

Low temperature/short duration steaming as a sustainable method of soil disinfection
This report was presented at the UK Organic Research 2002 Conference. Soil samples containing resting structures of fungal crop pathogens (Verticillium dahliae, Sclerotinia sclerotiorum, Sclerotium cepivorum, Pythium ultimum), potato cyst nematodes (Globodera rostochiensis and Globodera pallida) and weeds (Chenopodium album and Agropyron repens) were treated with aerated steam in the laboratory at temperatures ranging from 50–80oC in a specially constructed apparatus. Steaming at 50 or 60oC for three minutes, followed by an eight-minute resting period in the steamed soil and immediate removal from the soil thereafter, resulted in 100% kill of all weeds, fungi and nematodes. Low temperature/ short duration soil steaming could become a sustainable alternative to chemical or high-temperature steam soil disinfestation
Shp2 Knockdown and Noonan/LEOPARD Mutant Shp2–Induced Gastrulation Defects
Shp2 is a cytoplasmic protein-tyrosine phosphatase that is essential for normal development. Activating and inactivating mutations have been identified in humans to cause the related Noonan and LEOPARD syndromes, respectively. The cell biological cause of these syndromes remains to be determined. We have used the zebrafish to assess the role of Shp2 in early development. Here, we report that morpholino-mediated knockdown of Shp2 in zebrafish resulted in defects during gastrulation. Cell tracing experiments demonstrated that Shp2 knockdown induced defects in convergence and extension cell movements. In situ hybridization using a panel of markers indicated that cell fate was not affected by Shp2 knock down. The Shp2 knockdown–induced defects were rescued by active Fyn and Yes and by active RhoA. We generated mutants of Shp2 with mutations that were identified in human patients with Noonan or LEOPARD Syndrome and established that Noonan Shp2 was activated and LEOPARD Shp2 lacked catalytic protein-tyrosine phosphatase activity. Expression of Noonan or LEOPARD mutant Shp2 in zebrafish embryos induced convergence and extension cell movement defects without affecting cell fate. Moreover, these embryos displayed craniofacial and cardiac defects, reminiscent of human symptoms. Noonan and LEOPARD mutant Shp2s were not additive nor synergistic, consistent with the mutant Shp2s having activating and inactivating roles in the same signaling pathway. Our results demonstrate that Shp2 is required for normal convergence and extension cell movements during gastrulation and that Src family kinases and RhoA were downstream of Shp2. Expression of Noonan or LEOPARD Shp2 phenocopied the craniofacial and cardiac defects of human patients. The finding that defective Shp2 signaling induced cell movement defects as early as gastrulation may have implications for the monitoring and diagnosis of Noonan and LEOPARD syndrome
Tooth Whitening Effects on Bracket Bond Strength In Vivo
Objective: To test the hypothesis that there is no difference between the bracket survival rate of brackets bonded to bleached and unbleached teeth.
Materials and Methods: Thirty-eight patients who required comprehensive orthodontic treatment were included in the study. A split mouth technique was used with one arch exposed to in-office whitening gel containing 38% hydrogen peroxide for 30 minutes, while the unbleached arch served as the control. Patients were divided into two groups: Brackets bonded within 24 hours after bleaching and brackets bonded 2–3 weeks after bleaching. The bracket survival rate was computed using the log-rank test (Kaplan-Meier Analysis).
Results: A significantly higher rate of bracket failure was found with bleached teeth (16.6%) compared with unbleached teeth (1.8%) after 180 days. Brackets bonded within 24 hours of bleaching resulted in significantly higher clinical failure (14.5%) compared with those bonded after 3 weeks (2.1%). Adhesive Remnant Index scores of failed brackets revealed that the majority of failure in bleached teeth occurred in the enamel/resin interface.
Conclusions: The hypothesis was rejected. Brackets bonded within 24 hours after bleaching have a significantly higher risk for bond failure. Orthodontic bonding should be delayed for 2–3 weeks if patients have a history of in-office bleaching with 38% hydrogen peroxide

Transcription controls growth, cell kinetics and cholesterol supply to sustain ACTH responses
Chronic ACTH exposure is associated with adrenal hypertrophy and steroidogenesis. The underlying molecular processes in mice have been analysed by microarray, histological and immunohistochemical techniques. Synacthen infused for 2Â weeks markedly increased adrenal mass and plasma corticosterone levels. Microarray analysis found greater than 2-fold changes in expression of 928 genes (P < 0.001; 397 up, 531 down). These clustered in pathways involved in signalling, sterol/lipid metabolism, cell proliferation/hypertrophy and apoptosis. Signalling genes included some implicated in adrenal adenomas but also upregulated genes associated with cyclic AMP and downregulated genes associated with aldosterone synthesis. Sterol metabolism genes were those promoting cholesterol supply (Scarb1, Sqle, Apoa1) and disposal (Cyp27a1, Cyp7b1). Oil red O staining showed lipid depletion consistent with reduced expression of genes involved in lipid synthesis. Genes involved in steroidogenesis (Star, Cyp11a1, Cyp11b1) were modestly affected (P < 0.05; <1.3-fold). Increased Ki67, Ccna2, Ccnb2 and Tk1 expression complemented immunohistochemical evidence of a 3-fold change in cell proliferation. Growth arrest genes, Cdkn1a and Cdkn1c, which are known to be active in hypertrophied cells, were increased >4-fold and cross-sectional area of fasciculata cells was 2-fold greater. In contrast, genes associated with apoptosis (eg Casp12, Clu,) were downregulated and apoptotic cells (Tunel staining) were fewer (P < 0.001) and more widely distributed throughout the cortex. In summary, long-term steroidogenesis with ACTH excess is sustained by genes controlling cholesterol supply and adrenal mass. ACTH effects on adrenal morphology and genes controlling cell hypertrophy, proliferation and apoptosis suggest the involvement of different cell types and separate molecular pathways
