    Effects of ammonia toxicity on growth performance, cortisol, glucose and hematological response of Nile Tilapia (Oreochromis niloticus)

    Ammonia is a production limiting factor in the aquaculture media affecting fish production. A study was designed to scrutinize effects of ammonia on growth performance, survival, cortisol and hematological parameters of Tilapia fish. The study examined effects of 96 h-incubation of male and female Tilapia with 3 mg ammonium chloride per a liter of water compared to control. The study has been carried out in the physiology laboratory of the department of animal and fish production, Alexandria University. Fourteen aquaria were used (6 control and 8 ammoniated). Each aquarium contained 6 fish (half the population males and the other have females). Duration of the control reared fish was 30 days, however the duration for ammoniated group was 4 days. In all stressed fish, there found decreases in final body weight, average daily gain and specific growth rate as compared to controls. Hematological parameters revealed increases (P0.05) in total leukocyte counts in both males and females exposed to stressors. There were significant decreases (P0.05) in red blood cell, hematocrit value and hemoglobin concentration in both males and females. There were non-significant differences (P0.10) in these parameters between males and females. Exposing both male and female tilapia to ammonia, resulted in increases (P0.05) in mean corpuscular volume (MCV). Mean corpuscular hemoglobin (MCH) didn’t change in male tilapia, while females expressed increased MCH values in the ammonia condition. Mean corpuscular hemoglobin concentration (MCHC) decreased (P0.05) under ammonia with no differences between males and females. Differential leukocyte count exhibited increases (P0.05) in neutrophils in ammonia-exposed males and females and decreases (P0.05) in eosinophils and monocytes in males, but not in females. However, lymphocytes decreased (P0.05) in both females and males exposed to ammonia. Cortisol level increased (P0.05) by about 2 folds in both sexes of fish exposed to ammonia (2.95 and 2.72 vs6.40 and 6.48 ng/ml in control males and females vs ammonia-exposed males and females).Rearing tilapia fish in media containing high level (3mg/l water) of ammonium chloride not only deteriorated growth rate but it also negatively affected the health wellbeing.

    Effectiveness of a recombinant human follicle stimulating hormone on the ovarian follicles, peripheral progesterone, estradiol-17β, and pregnancy rate of dairy cows

    Aims: This study aimed at elucidating the effects of recombinant human follicle stimulating hormone (r-hFSH) on the ovarian follicular dynamics, progesterone, estradiol-17β profiles, and pregnancy of dairy cows. Materials and Methods: Three groups (G, n=5 cows) of multiparous dairy cows were used. G1 (C) control cows were given controlled internal drug release (CIDR) and prostaglandin F2α; G2 (L) cows were given low dose (525 IU and G3 (H) cows were given high dose (1800 IU) of r-hFSH on twice daily basis at the last 3 days before CIDR removal. All cows were ultrasonically scanned for follicular growth and dynamics, and blood samples were collected every other day for two consecutive estrus cycles for the determination of estradiol-17β and progesterone. Results: Estrus was observed in all C and L but not in H cows. Dominant follicle was bigger in L compared to C and H cows. Dominant follicle in C (16.00±2.5 mm) and L cows (17.40±2.3 mm) disappeared at 72 h after CIDR removal. However, in H cows, no ovulation has occurred during 7 days post-CIDR removal. Progesterone was not different (p>0.10) among groups, whereas estradiol-17β revealed significant (p<0.01) reduction in H (15.96±2.5 pg/ml) cows compared to C (112.26±26.1 pg/ml) and L (97.49±15.9 pg/ml) cows. Pregnancy rate was higher in L cows (60%) compared with C cows (20%). However, H cows were not artificially inseminated due to non-ovulation. Only a cow of C group has calved one calf, however, 2 of the L cows gave birth of twins and a cow gave single calf. Conclusion: Administration of a low dose (525 IU) of r-hFSH resulted in an optimal size of dominant follicle, normal values of progesterone and estradiol-17β, and 40% twinning rate, howeverusing 1800 IU of r-hFSH, have adverse effects on ovarian follicular dynamics and hormonal profiles with non-pregnancy of dairy cows raised under hot climate

    The objectives of this study were to elucidate the impact of the administration of recombinant bovine somatotropin (rbST), in combination with Ovsynch protocol on the preovulatory follicle (POF) diameter, a daily growth rate of the dominant follicle (DF), ovulation rate, and the subsequent conception rate in buffaloes. Fifty buffaloes (Bubalus bubalus) were utilized in this study and divided into two groups {rbST group (rbSTG), n = 25; and control group (CG), n = 25}. Ultrasonography of ovaries revealed that the mean diameter of POF was larger (P < 0.05) in rbSTG compared to CG buffaloes (14.23 ± 0.34 mm vs. 10.67 ± 0.95 mm). Also, the daily growth rate of the DF from Day 7 till the day of ovulation was higher (P < 0.05) in rbSTG compared with CG buffaloes (1.24 ± 0.15 vs. 0.64 ± 0.10 mm/day). Besides, the ovulation rate in rbSTG was higher [(88%), P < 0.05] compared with the CG cows (64%). Thus, the conception rate in rbSTG buffalo was higher (56%; P < 0.05) compared with CG counterparts (40%). In conclusion, rbST combination with Ovsynch protocol for the synchronization of estrus and ovulation significantly improved the follicular dynamics and subsequent conception rate of buffaloes

    Gestation in sheep necessitates the support of nutrients to avoid early embryonic mortalities. Therefore, this study investigates the effects of supplementing either L-arginine or Spirulina alga in the first trimester on the pregnancy rate, litter size, inhibin, insulin, IGF-I, CO Q10, and bone morphogenetic peptide 15 (BMP15) in maternal circulation. Animals were offered barley (500 g/head/day, 14% CP), alfalfa hay (1 kg/head/day, 12% CP), clean water, and balanced salt block licks as free choices. Forty Noemi and Najdi ewes were randomly allotted into three groups: control (C, n = 8), L-arginine (ARG, n = 16), and Spirulina (SP, n = 16). All females were implanted with CIDR for ten days. On days 8, 9, and 10, treated ewes were given a protocol comprised of human recombinant FSH at descending doses (50, 50, 40, 40, and 30, 30 IU, A.M, and P.M, respectively). At the fifth dose, animals were given an equivalent dose of hCG (240 IU). After CIDR withdrawal, ewes were exposed to fertile rams for mating. SP-ewes were orally given 50 ml (2%) Spirulina, and ARG-ewes were given 50 ml (35 mg/kg BW) L-arginine daily for 50 days postbreeding. Blood inhibin, insulin, IGF-I, CO Q10, and BMP15 were determined throughout gestation until parturition. The findings indicated that the conception rates were 25, 75, and 87.5% in C, ARG, and SP, respectively (P<0.05). The percent of ewes giving birth to twins was 0, 25, and 50% in C, ARG, and SP, respectively (P<0.05). The survival rates were 100, 81.8, and 83.3%, respectively. Birth weight was 5.6, 3.2, and 3.4 kg in C, ARG, and SP, respectively. Weaning weights were 28.3, 25.6, and 27.2 kg in C, ARG, and SP, respectively. BMP-15 was reduced (P<0.05) in ARG than in C and SP. However, SP decreased (P<0.05) inhibin more than in C and ARG. ARG and SP increased (P<0.05) insulin than in C, whereas SP decreased (P<0.05) IGF-I. SP increased CO Q10 compared with ARG. Ewes bearing twins revealed higher (P<0.05) IGF-I (8.57 ng/ml) than those bearing singles (4.63 ng/ml); however, BMP-15 was higher in single (796.6 pg/ml) than in twin-bearing (387.5 pg/ml) ewes. In conclusion, providing early-gestating ewes with Spirulina enhances maternal health, productivity, and reproductive outcomes

    Is MRI diffusion-weighted imaging a reliable tool for the diagnosis and posttherapeutic follow-up of extremity soft tissue neoplasms?

    Purpose: The aim of this study was to evaluate the benefit of using quantitative diffusion-weighted imaging (DWI) with apparent diffusion coefficient (ADC) mapping in the initial diagnosis and post-therapeutic follow-up of extremity soft tissue masses. Patients and Methods: This study included 90 patients with extremity soft tissue masses. The DWI was obtained with 3 b values, including 0, 400, and 800 s/mm2. Calculation of the ADC value of the lesion was done by placing the region of interest (ROI) to include the largest area of the lesion. ADC values were compared with the histopathology. Eighteen patients had posttherapeutic magnetic resonance imaging (MRI). Results: Benign masses, fibromatosis, and malignant soft tissue masses had mean ADC values of 1.18 ± 1.0191 × 10−3 mm2/s; 1.31 ± 0.245 × 10−3 mm2/sec; and 1.3 ± 0.7 × 10−3 mm2/s, respectively. Myxomatous malignant masses had an ADC value of 2.6 ± 0.55 × 10−3 mm2/s, while nonmyxomatous malignant masses had an ADC value of 1.1 ± 0.8 × 10−3 mm2/s. ADC cutoff value between benign and non-benign (including malignant and locally aggressive masses) was 0.6 × 10−3 mm2/sec with 98.3% sensitivity and 50% specificity (P = 0.5123). The statistical difference between malignant soft tissue masses (mean ADC 1.309 ± 0.723 × 10−3 mm2/s) and fibromatosis masses (mean ADC value 1.31 ± 0.245 × 10−3 mm2/s) using a comparative T-test proved to be of poor significance level (P value ~ 0.9757). Nine patients with soft tissue sarcomas (STSs) had pre and post-therapeutic MRI examinations showing a mean increase of the recorded ADC values by about 0.28 × 10−3 mm2/s in the post-therapy study as compared with the recorded initial pretreatment values. Analysis of the post-therapy follow-up studies of fibromatosis showed that lesions with favorable response to chemotherapy or radiotherapy (8/12) exhibited significantly lower ADC values than those showing progressive disease course. Conclusion: DWI with ADC mapping of extremity soft tissue tumors are so complicated that they alone may not be of much value in differentiating between benign and malignant tumors; however, it can be used as a tool for monitoring response to treatment

    Aim: The aim of this study was to investigate the effectiveness of using recombinant human follicle-stimulating hormone (FSH) compared with pituitary-derived porcine FSH given as one dose or multiple doses on the neonatal traits, follicular dynamics, and maternal blood biochemical constituents in Noemi ewes. Materials and Methods: A 3×2 factorial arrangement was designed utilizing 60 adults Noemi ewes to test the effects of using two sources of FSH (human vs. porcine) in addition to control, either given as a single total dose or six descending doses to provoke twinning. Six treatments (T) were tested (n=10 ewes/T). C1 and C6 served as control ewes given saline as one dose and six doses, respectively; H1 and H6 ewes were given human FSH as one and six doses; and P1 and P6 ewes were given porcine FSH similar to the above treatments. Saline and/or FSH administration were administered at days 8, 9, and 10 of the 10-day controlled internal drug release (CIDR) implant. At CIDR removal, fertile rams were used for natural mating. Blood samples for the assessment of serum metabolites were collected. Results: Twinning increased in FSH-treated ewes than control. However, giving FSH of either source as a single dose resulted in a higher incidence of stillbirths. Pregnancy rates were 30, 40, 50, 60, 70, and 80% in C1, C6, P1, P6, H1, and H6, respectively. Respective percent of ewes delivering twins/multiple birth was 0, 0, 80, 66.7, 71.4, and 87.5%. FSH of human source was more efficient for folliculogenesis than porcine FSH. Administration of FSH increased blood cholesterol, decreased high-density lipoprotein; however, low-density lipoprotein levels were not different than control. Moreover, an interaction (p<0.05) exists between source and type of FSH administration on blood glucose. Six doses of FSH elevated blood protein. Blood albumin decreased by porcine-FSH but not affected by human-FSH. Blood globulins were not different due to source of FSH, whereas giving FSH as six doses increased globulins than in single-dose protocol. Contrariwise, an interaction was found between source and type of FSH administration on elevating the activity of alanine aminotransferase and reducing the activity of aspartate aminotransferase. Conclusion: Administration of human FSH at 180 IU in six descending doses resulted in the best neonatal outcomes and maternal health in Noemi ewes

    Consolidating probiotic with dandelion, coriander and date palm seeds extracts against mercury neurotoxicity and for maintaining normal testosterone levels in male rats

    Objective: Heavy metals are major elements polluting our universe. The inhalation, ingestion or even contacting human body with these elements results in huge health problems. The most common pollutant in our surrounding is mercury. Therefore, the present study aimed to elucidating the protective ability of hot water extracts of dandelion (DA), coriander (CO), date palm seeds (DS), probiotic supernatant (PS) and their combined mixture against mercury-induced neurotoxicity and altered testosterone levels in male rats. Methods: Fifty six male rats were randomly allotted into seven groups (n = 8 rats/group). Group1 (negative control; NC) animals were fed on the basal diet only, group2 (positive controls; PC) animals were fed on the basal diet and given an aqueous solution of mercuric chloride (25 ppm mercuric) in drinking water. Animals of the antioxidant-treated groups (3–7) were fed on the basal diet and given an aqueous solution of mercuric chloride (25 ppm mercuric) in drinking water together with the herbal antioxidant extracts and probiotics (25 ml/rat/day) throughout the experimental period. Where, group3 (Hg/CO) given coriander extract, group4 (Hg/DA) given dandelion extract, group5 (Hg/DS) given date palm seeds extract, group6 (Hg/PS) given probiotic supernatant, and group7 (Hg/Mix) given mixture of equal quantities of probiotic supernatant together with the three herbal extracts. The treatment lasted for 6 weeks, animals were sacrificed and blood samples were collected. Blood testosterone, enzyme activity and histopathological sections were performed. Results: The obtained data exhibited that mercury intoxication revealed increases of lactic dehydrogenase and decreases of glutathione-s-transferase and testosterone. Light microscopic investigations of the brain cortex and cerebellum were suggestive of multiple foci of inflammation, cellular infiltration, gliosis and degeneration. Moreover, decreased glial fibrillary acidic protein (GFAP)-immunoreactivity and potential astrocyte toxicity both reflected impaired neuro-protective function of astrocytes necessary for maintaining the brain structure and function. Conclusion: Administration of the herbal extracts and their mixture with probiotics enhance the body defense and contain protective factor against mercury neurotoxicity and for maintaining normal testosterone levels in male rats. Also, treatment restored the normal control levels of biochemical attributes and histological architecture.
