    Effect of a high forage: Concentrate ratio on milk yield, blood parameters and oxidative status in lactating cows

    A feeding strategy that requires a forage: concentrate ratio equal to 70: 30, with at least five different herbs in the forage and the use of silages prohibited, has recently been introduced in Italy. Despite the benefits in terms of human health (lower ω6: ω3 ratio, higher conjugated linoleic acid level) of the obtained milk, little information regarding the possible effects on cows' health is available. The aim of this study was to evaluate the effects of such a feeding strategy in dairy cows (90 days in milk at the beginning of the trial) on milk yield and composition, and blood metabolic profile, including the evaluation of oxidative stress. The proposed feeding strategy, compared with a semi-intensive strategy, resulted in an improvement of animal oxidative status (lower levels of reactive oxygen metabolites, higher levels of antioxidant potential and anti-reactive oxygen metabolites) and a significant increase of milk urea only in the first part of the trial. No differences in milk yield and composition were detected throughout the trial

    Influence of dietary hydrogenated palm oil supplementation on serum biochemistry and progesterone levels in dairy goats

    The aim of this research was to investigate the influence of hydrogenated palm oil (HPO) added to a dairy goat diet on serum biochemistry and progesterone levels. Thirty pregnant Cilentana dairy goats were equally divided into 2 groups (control [CTR] and HPO groups). After kidding, concentrated feed for both groups was gradually increased up to 400 g/(animald),andtheHPOgroupreceived50g/(animald), and the HPO group received 50 g/(animald) of HPO. Supplementation with HPO significantly increased cholesterol levels (mg/dL, 63.80 vs. 54.68 at 30 d, P 0.05; 78.20 vs. 58.00 at 60 d, P 0.05; 83.80 vs. 57.83 at 120 d, P 0.01) compared with the CTR group although no significant differences were detected for liver and kidney function indicators. Moreover, other biochemical parameters were not affected by HPO supplementation thus suggesting no change occurred in lipid and protein metabolism. Furthermore, a significant correlation was found be- tween progesterone levels and serum cholesterol (r 1⁄4 0.65, P 0.01) although these were not signifi- cantly higher in HPO supplemented goats. The dose and time of HPO supplementation appears critical as regards assessing the limits between the risks and benefits of HPO supplementation in dairy goats. At the tested dose, HPO was well tolerated by the animals and may represent a useful tool to increase energy availability during highly demanding periods

    Evaluation of the capillary electrophoresis method for measurement of immunoglobulin concentration in ewe colostrum

    ABSTRACT Capillary electrophoresis (CE) is a technique routinely used in clinical laboratories that allows the separation and quantification of blood serum proteins in a rapid, precise, accurate, and inexpensive manner. Recently, CE has been proposed to separate and measure colostral proteins, but an evaluation of the agreement between CE and radial immunodiffusion (RID) method, currently used to quantify IgG in colostrum, is still lacking. The purpose of this study was to test the ability of a CE instrument, normally used in blood serum protein analysis, to realize the correct quantification of total Ig concentration in ewe colostrum, using RID assay as reference. Colostrum samples (n = 68) were collected from 35 multiparous Sarda ewes at first milking (n = 33) and at 24 h postpartum (n = 35). The mean ± standard deviation of IgG concentration measured by RID and whey colostrum total Ig concentration measured by CE were 54.76 ± 41.82 g/L and 54.70 ± 41.43 g/L, respectively. Lin's concordance correlation coefficient (r = 0.993; 95% confidence interval=0.989 to 0.996) and linear regression analysis results (RID = 1.0022CE − 0.063; R 2 = 0.986) showed an excellent agreement between these 2 methods. Bland-Altman analysis confirmed that CE method can be a suitable alternative to RID: the mean of the differences between CE and RID was −0.055 ± 4.95 g/L (95% confidence interval=−1.25 to 1.14 g/L) and the agreement limits were −9.75 to 9.60 g/L (low limit 95% confidence interval=−11.82 to −7.68 g/L; high limit 95% confidence interval=7.57 to 11.72 g/L). In conclusion, the current study indicates that CE method may be a reliable tool for the quantification of the total Ig concentration in ewe colostrum

    Effects of Sorghum silage in lactating buffalo cow diet: Biochemical profile, milk yield, and quality

    The influence of replacing silage with sorghum silage in the diet of dairy buffalo cows on metabolic status and on milk yield, chemical characteristics, and fatty acid profile was studied. Forty dairy buffalo cows were included in the trial and divided into two homogeneous groups (SS, sorghum silage and CS, corn silage). Blood was collected at the end of the trial (120 days), individual milk yield was registered daily. Samples of milk were monthly collected and analyzed for fat, protein, and lactose. Moreover, fatty acid profiles of silages and milk were determined. Buffalo cows fed sorghum silage showed an average milk yield higher than group CS (kg/d 10.120 vs. 9.270; p < 0.05), probably due to the lower lignin content of sorghum silage (31.0 g/kg dry matter (DM)) compared to corn silage (47.0 g/kg DM) and, by consequence, to the higher digestibility of SS diet. The percentage of linoleic acid was significantly higher in milk of group CS (C18:2: 1.27 vs. 2.05%; p < 0.01) due to the higher content of these acids in corn than in sorghum silage. The omega 6/omega 3 ratio was significantly lower in milk from buffalo cows fed sorghum than those fed corn silage (7.8 vs. 12.9; p < 0.01). Serum biochemistry showed no negative effects of the corn replacing with sorghum

    In vitro and in vivo studies of Cucurbita pepo L. flowers: chemical profile and bioactivity

    Edible flowers consumption has increased in recent years due to their rich content of healthy phytochemicals. The aim of this study was to analyse the chemical profile of Cucurbita pepo L. flowers, and to explore their antioxidant and hypoglycaemic prop- erties. Moreover, in order to assess in vivo effects, biochemical analysis, Reactive Oxygen Metabolites (d-ROMs) and Biological Antioxidant Potential (BAP) tests were performed on mice serum. High Performance Liquid Chromatography-Diode Array Detection (HPLC-DAD) analyses revealed the presence of (þ)-catechin, ()-epicatechin, rutin, and syringic acid as main constituents. 2,20- Azino-bis (3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) and Ferric Reducing Antioxidant Power (FRAP) tests showed interest- ing results. The extract exhibited the strongest inhibitory effect on a-glucosidase (IC50 of 144.77lg/mL). In vivo results confirmed the hypoglycaemic effects, also affecting lipid metabolism but did not revealed benefits on ROS production. These results may add some information supporting the use of C. pepo flowers as func- tional foods and/or nutraceuticals

    Influence of Pasture on Stearoyl-CoA Desaturase and miRNA 103 Expression in Goat Milk: Preliminary Results

    The effect of pasture on the stearoyl-CoA desaturase (SCD) and miRNA 103 expression was evaluated on dairy goats divided into two homogeneous groups (G, grazing, and S, stable). Group S was housed in a stall and received alfalfa hay as forage, while group G was led to pasture. The goats of both the groups received the same amount of concentrate. Milk yield did not di↵er statistically between the groups. Group G showed significantly higher fat (4.10% vs. 2.94%, p < 0.01) and protein percentage (3.43% vs. 3.25%; p < 0.05) than group S. Among milk fatty acids, group S showed significantly higher levels of saturated fatty acids (SFA) and lower values of mono-unsaturated fatty acid (MUFA). The percentages of polyunsaturated fatty acid (PUFA) were not different between groups even if pasture significantly affected the percentages of C18:3 and total omega 3. In group G, total CLAs were twice than in group S (0.646% vs. 0.311%; p < 0.01) mainly due to the differences in CLA cis9 trans 11 (0.623% vs. 0.304%; p < 0.01). Milk total CLA in grazing group was significantly (p < 0.01) higher in August according to the highest value of both linoleic and alfa-linolenic acids in the pasture. In grazing animals, SCD expression decreased from April to June, increased in July and decreased again in August, while it was almost unvaried along the trial in group S. By contrast, the expression of miRNA 103 showed a similar trend for both groups, decreasing from April to June, increasing in July and falling down in August. To our knowledge, this is the first observation of the effects of pasture on miRNA expression in milk from ruminant species

    Effects of ecotrofin™ on milk yield, milk quality and serum biochemistry in lactating goats

    A nutritional supplement (Ecotrofin™, by Vetoquinol Italia S.r.l) recommended in ruminants feeding to strengthen the physiological condition and improve digestive performance was tested in 20 pluriparae grazing goats divided in two groups (control and treated) to assess its possible effects on milk yield and quality and to assess eventual adverse effects. Animals from both groups also received 400&nbsp;g/day of corn meal, and the treated group was supplemented with 20&nbsp;g/head/day of the nutritional supplement. At the doses suggested by the manufacturer, despite a transient increase after 30&nbsp;days of supplementation, Ecotrofin™ did not show significant effects on milk yield and, although some changes were found in the fatty acids profile, no significant improvement of MUFA and PUFA, as well as of omega-6:omega-3 ratio and CLA content were seen. Therefore, in our experimental conditions the supplementation of diet with Ecotrofin™ did not appear useful to improve goat's performance. A significant effect on kidney health markers (27 vs. 22.5 for urea and 0.83 vs. 0.76 for creatinine, p&nbsp;&lt;&nbsp;0.05) suggested a beneficial effect on renal function but, since levels fell in the normal ranges in both groups, such hypothesis would need further studies to be addressed

    Dietary supplementation with a phytocomplex affects blood parameters and milk yield and quality in grazing goats

    A phytocomplex composed by Schizochytrium limacinum, a species of marine alga, Galega officinalis, an herbaceous plant of the Faboideae subfamily, and linseeds, was administered to lactating grazing goats in order to evaluate its effects on milk yield and quality, and to investigate possible effects on goat health status. The hypothesis was that, by using a phytocomplex including plants known to exert an improvement of milk yield and milk quality, a synergism should be possible in order to obtain such effects using low doses, thus avoiding adverse effects. Milk fat was significantly (p&lt;0.05) higher in treated group (4.02 vs. 3.61 %), in particular, levels of MUFA (24.55 vs 22.56 %), total n3 (1.34 vs 1.19 %) and total CLAs (0.52 vs. 0.40 %), were increased while n6/n3 (1.71 vs. 2.21 %) ratio was decreased. Concerning biochemical parameters, a significant (p&lt;0.05) decrease of creatinine (0.73 vs. 0.84 mg/dl) was observed in treated animals, but the levels fell within the normal range for this species. Thus, the addition of the proposed phytocomplex to the diet of grazing goats may be successfully used to improve milk fatty acids profile without negative effects on animal health

    The association of aloe and β-carotene supplementation improves oxidative stress and Inflammatory state in pregnant buffalo cows

    The complex polysaccharides of Aloe have been reported to improve immunity and oxidative status either in vitro or in vivo studies. Further, β-carotene has been shown to influence immune function and health in the cow. In this study, the effects of VigoorsanTM, a commercial supplement containing Aloe arborescence and β-carotene, on biochemical profile and oxidative/inflammatory status, were evaluatedin pregnant buffalo cows. VigoorsanTM was supplied (50 g/day/head) to pluriparae buffalo cows for 30 days before delivery. A significant increase of BAP and OXY and a decrease of d-ROMs (P<0.01) were detected in blood from cows supplemented with VigoorsanTM. Further, a decrease of TNF-α, IL1-βand MCP-1 and an increase of IL-10 were showed (P<0.01). Blood chemistry profiles showed no adverse effects on health status. Results showed that VigoorsanTM supplementation improves oxidative and inflammatory status with no negative effects on metabolism in water buffalo cow

    The Quality of Five Natural, Historical Italian Cheeses Produced in Different Months: Gross Composition, Fat-Soluble Vitamins, Fatty Acids, Total Phenols, Antioxidant Capacity, and Health Index

    Five natural historic cheeses of Southern Italy were investigated\u2014Caciocavallo Palermitano (CP), Casizolu del Montiferru (CdM), Vastedda della Valle del Bel\uecce (VVB), Pecorino Siciliano (PS), and Caprino Nicastrese (CN)\u2014which are produced with raw milk and with traditional techniques and tools, from autochthonous breeds reared under an extensive system. The effects of the month of production on gross composition, MUFA, PUFA, PUFA-\u3c96, PUFA-\u3c93, \u3b1-tocopherol, retinol, cholesterol, TPC, TEAC, and GHIC were evaluated. In CP, CLA, TPC, and GHIC were higher in April than in February. CdM showed higher values in terms of fat, saturated fatty acids, PUFA-\u3c93, \u3b1-tocopherol, TEAC, and GHIC in May than in February and September, while low values in terms of protein, moisture, and CLA were found. In VVB, MUFA, PUFA-\u3c96, and \u3b1-tocopherol increased in June compared with April; conversely, protein, FRAP, and TEAC were higher in April. In PS, protein, CLA, PUFA, PUFA-\u3c93, \u3b1-tocopherol, and GHIC increased in May compared with January; on the contrary, moisture, NaCl, and TEAC showed high values in January. CN showed higher values in terms of PUFA, PUFA-\u3c96, PUFA-\u3c93, TPC, TEAC, and GHIC in April and June compared with January. It is shown that each cheese is unique and closely linked to the production area. Cheeses produced in the spring months showed a high nutritional quality due to the greatest presence of healthy compounds originating from an extensive feeding system. For the purposes of raising awareness of five historical cheeses of Southern Italy that are less known by consumers, and of restoring dignity to the breeders and producers of these cheeses, we studied their quality in terms of chemical composition, monounsaturated fatty acid (MUFA), polyunsaturated fatty acid (PUFA), conjugated linoleic acid (CLA), PUFA-\u3c96, PUFA-\u3c93, \u3b1-tocopherol, retinol, cholesterol, polyphenol content (TPC), total antioxidant capacity (FRAP and TEAC), and health index (GHIC). Two stretched-curd bovine cheeses, Caciocavallo Palermitano (CP) and Casizolu del Montiferru (CdM), two ovine cheeses, Vastedda della Valle del Bel\uecce (VVB) and Pecorino Siciliano (PS), and one caprine cheese, Caprino Nicastrese (CN), were evaluated. These cheeses are produced in different months, with raw milk from animals reared in an extensive feeding system. In April, the CP cheese showed high values for CLA, TPC, and GHIC, while the CN cheese exhibited high PUFA, PUFA-\u3c96, PUFA-\u3c93, TEAC, and GHIC. In May, the CdM cheese exhibited high content of fat, saturated fatty acids, PUFA-\u3c93, \u3b1-tocopherol, TEAC, and GHIC, while the PS cheese showed high values of protein, CLA, PUFA, PUFA-\u3c93, \u3b1-tocopherol, and GHIC. These measured parameters characterize and distinguish each cheese due to links with numerous factors: Species, breed, feeding system, pasture biodiversity, climate, production technology, traditional tools, and ripening type. It is highlighted that, in general, the highest nutritional quality, linked to the highest presence of healthy compounds, originates from the pasture of cheese production in the spring
