    De novo design of heat-repressible RNA thermosensors in E-coli

    RNA-based temperature sensing is common in bacteria that live in fluctuating environments. Most naturally-occurring RNA thermosensors are heat-inducible, have long sequences, and function by sequestering the ribosome binding site in a hairpin structure at lower temperatures. Here, we demonstrate the de novo design of short, heat-repressible RNA thermosensors. These thermosensors contain a cleavage site for RNase E, an enzyme native to Escherichia coli and many other organisms, in the 5ā€² untranslated region of the target gene. At low temperatures, the cleavage site is sequestered in a stemā€“loop, and gene expression is unobstructed. At high temperatures, the stemā€“loop unfolds, allowing for mRNA degradation and turning off expression. We demonstrated that these thermosensors respond specifically to temperature and provided experimental support for the central role of RNase E in the mechanism. We also demonstrated the modularity of these RNA thermosensors by constructing a three-input composite circuit that utilizes transcriptional, post-transcriptional, and post-translational regulation. A thorough analysis of the 24 thermosensors allowed for the development of design guidelines for systematic construction of similar thermosensors in future applications. These short, modular RNA thermosensors can be applied to the construction of complex genetic circuits, facilitating rational reprogramming of cellular processes for synthetic biology applications

    Using the 5-Item Medication Adherence Report Scale (MARS-5) to Screen for Non-adherence to Vitamin and Mineral Supplementation After Bariatric Surgery

    INTRODUCTION: Poor adherence to recommended vitamin and mineral supplementation after bariatric surgery is common and challenging for healthcare professionals to identify. There are several questionnaires for self-reporting of adherence to chronic medication, but none has so far been evaluated for assessment of adherence to vitamin and mineral supplementation after bariatric surgery. The aim of this study was to assess the accuracy of the 5-item Medication Adherence Report Scale (MARS-5) in measuring adherence to vitamin and mineral supplementation post bariatric surgery (gastric bypass or sleeve gastrectomy). METHOD: The psychometric properties of MARS-5 for vitamin and mineral supplementation were validated in two cohorts: one at 1 year post bariatric surgery (n = 120) and the other at 2 years post-surgery (n = 211). MARS-5 was compared to pharmacy refill data for vitamin B12 and combined calcium/vitamin D as reference. RESULTS: Correlation analyses demonstrated that the MARS-5 had acceptable validity compared to objectively measured adherence rates from pharmacy refill data (calculated as continuous, multiple-interval measures of medication availability/gaps-coefficient ranged from 0.49 to 0.54). Internal reliability (Cronbach's Ī±) was high: 0.81 and 0.95, respectively. There was a clear ceiling effect where one out of three had a maximum score on MARS-5. CONCLUSION: MARS-5 demonstrated acceptable psychometric properties for assessment of adherence to vitamin and mineral supplementation post bariatric surgery

    Effects of resistance training on classic and specific bioelectrical impedance vector analysis in elderly women

    Raw bioelectrical impedance analysis (BIA) data [resistance (R); reactance (Xc)] through bioelectrical impedance vector analysis (BIVA) and phase angle (PhA) have been used to evaluate cellular function and hydration status. The purpose of this investigation was to examine the effects of resistance training (RT) on classic and specific BIVA in elderly women. Twenty women (mean Ā± SD; age: 71.9 Ā± 6.9 years; BMI: 24.5 Ā± 3.0 kg m(-2)) completed a 6-month RT program. Whole-body, single-frequency BIA, body geometry, and leg strength (5RM) measures were completed at baseline (t0), 3 months (t3), and 6 months (t6). The mean impedance vector displacements were compared using Hotelling's T(2) test to evaluate changes in R and Xc relative to height (R/ht; Xc/ht) or body volume (Rsp; Xcsp) estimated from the arms, legs, and trunk. 5RM, PhA, and BIVA variables were compared using ANOVA. PhA improved at t6 (p < 0.01), while 5RM improved at t3 and t6 (p < 0.01). Using classic BIVA, 6 months (T(2) = 31.6; p < 0.01), but not 3 months of RT (T(2) = 4.5; p = 0.20), resulted in significant vector migration. Using specific BIVA, 6 months (T(2) = 24.4; p < 0.01), but not 3 months of RT (T(2) = 5.5; p = 0.10), also resulted in significant vector migration. 5RM was correlated to both PhA (r = 0.48-56) and Xcsp (r = 0.45-53) at all time points. Vector displacements were likely the result of improved cellular integrity (Xcsp) and cellular health (PhA)
