    Analysis of the Cellular Proteins, TIA-1 and TIAR, and their Interaction with the West Nile Virus (WNV) 3\u27 SL Minus-Strand RNA

    The 3\u27 terminal stem loop of the WNV minus-strand [WNV3\u27(-) SL] RNA was previously shown to bind the cell protein, T-cell intracellular antigen-1 (TIA-1), and the related protein, TIAR. These two proteins are known to bind AU-rich sequences in the 3\u27 UTRs of some cellular mRNAs. AU stretches are located in three single-stranded loops (L1, L2, and L3) of the WNV3\u27(-) SL RNA. The RNA binding activity of both proteins was reduced when L1 or L2, but not L3, AU sequences were deleted or substituted with Cs. Deletion or substitution with Cs of the entire AU-rich sequence in either L1 or L2 in a WNV infectious clone was lethal for the virus while mutation of some of these nt decreased the efficiency of virus replication. Mutant viral RNAs with small plaque or lethal phenotypes had similar translational efficiencies to wildtype RNA, but showed decreased levels of plus-strand RNA synthesis. These results correlated well with the efficiency of TIA-1 and/or TIAR binding in in vitro assays. In normal cells, TIA-1 and TIAR are evenly distributed in the cytoplasm and nucleus. Between 6 and 24 hr after WNV infection, TIAR concentrated in the perinuclear region and TIA-1 localization to this region began by 24 hr. Similar observations were made in DV2 infected cells but at later times after infection. In infected cells, both proteins colocalized with dsRNA, a marker for viral replication complexes, and with viral non-structural proteins. Anti-TIAR or anti-TIA-1 antibody coimmunoprecipitated viral NS3 and possibly other viral nonstructural proteins. In response to different types stress, TIA-1 and TIAR recruit cell mRNA poly(A)+ into cytoplasmic stress granules (SG) leading to general translational arrest in these cells. SG were not induced by flavivirus infection and cells became increasingly resistant to arsenite induction of SG with time after infection. Processing Body (PB) assembly was also decreased beginning at 24 hr. These data suggest that the sequestration of first TIAR and then TIA-1 via their interaction with viral components in flavivirus infected cells inhibits SG formation and prevents the shutoff of host translation

    Enhancement of circular RC columns using steel mesh as internal or external confinement under the influence of axial compression loading

    Reinforced concrete (RC) columns cannot get supreme confinement by using the customary steel stirrups reinforcement because of the requirements for the spacing distances between the stirrups in addition to concrete continuance trouble. For this, Steel Mesh (SM) externally wrapped around the outer perimeter of the column as contributory confinement are being widely used due to its features. Limited tests focused on using SM for the internal confinement around the reinforcing cage of RC columns. Moreover, no experimental comparison was presented between RC columns internally and externally confined using SM. This paper investigates experimentally the behavior of circular RC columns confined internally or externally by SM. Six short RC columns have been subjected to axial loading until failure. The main studied parameters were SM schemes, number of SM wraps, SM position (internally or externally), and the steel stirrups existence. Results demonstrated that SM could decrease the crack opening, diminish the concrete spalling, increase the maximum failure load, and enhance the ductility, energy absorption, and column stiffness. Furthermore, the partially internal confinement using two wraps of SM around the steel ties presented the maximum capacity with reasonable ductility. In general, internally confined columns showed better behavior than the externally confined one

    Prediction of self compacted rubberized concrete properties using Taguch methods

    The effect and optimization of using self-compacting rubberized concrete was investigated by using Taguchi method. Design of experiment was performed via orthogonal array to accommodate four factors with four levels. These factors were the percentage of fine rubber, coarse rubber, fly ash and viscocrete in the concrete mix. The signal-to-noise (S/N) ratio and the analysis of variance (ANOVA) were employed to study the performance characteristics of self-compacting rubberized concrete (SCRC). Rubberized concrete can be improved using the concrete proportioned as self-compacting concrete. The results indicate that there was a reduction in the strength with increasing rubber content but there was an increase in impact resistance. However, the replacement of 10% of coarse aggregate with coarse rubber gave more strength than that of zero rubber mix by 124% at 90 days. Replacement of 20% of both fine and coarse aggregates with fine and coarse rubber respectively, increased impact resistance by 453% compared to the corresponding SCRC control mix

    Hydrographical conditions and benthic assemblages in the Suez Gulf, Egypt

    The coastal development and human activities along the Suez Gulf leading to sedimentation, degrade the quality of water, disturbing the natural structure and functions of aquatic communities. The Suez Gulf is a large semi-closed area with a 346 km long coastline on the western beach side. The prevailing physicochemical parameters in shallow intertidal waters were measured seasonally over the year. Benthic faunas in the sampling sites were studied indicating their regional distribution in relation to the impact of different environmental parameters in the intertidal region. The concentration of copper in seawater reached high level at St. IV (4.57 ug/1), which is exposed to sewage and petroleum hydrocarbons. The grain size of the sediment is a determining factor for the organic carbon concentration and the sandy substrate enhances organic matter degradation processes. A large number of oil fields are present along the western coast of the Suez Gulf, therefore, cadmium and organic matter appeared to be high. The values of pH did not vary greatly among the different sampling sites. It was high at EI-Ein, El-Sukhna and Ras-Shukeir due to the disposal of mainly acidic sewage and industrial effluents of the two stations Adabiya and Ras-Gharib respectively. The macrobenthos included 71 species embraced mainly from Mollusca (53.5% Gastropoda and 12.7% Bivalvia) and the other invertebrates included 7 groups namely, Rhizostoma, Polychaeta, Cirripedia, Amphipoda, Isopoda, Decapoda and Echinodermata. The distribution of benthos is affected by the temperature and salinity of seawater. The concentration of organic matter in seawater and in sediments in shallow waters shows high values in the central part of the Gulf of Suez

    Ecology and settlement of marine fouling in the Suez Bay, Egypt

    This study deals with seasonal variations, natural correlations and similarities of fouling assemblages on exposure panels in the Suez Bay during January 1992 to January 1993. Three main sources of pollutions flow into the bay; industrial waste products, domestic drainage of Suez city and ships' oil and refuse.The fouling assemblages on the test pan els after various periods (1, 2 and 3 months) belonged mainly to the algae (Ulva rigida), polychaetes (Hydroides elegans), Cirripedes (Balanus amphitrite) and amphipods. The fouling at the lst station was relatively more dense than at the 2nd station during the summer and autumn seasons. The lowest productivity was achieved at the 3rd station which was considered less polluted being offshore water. The overall paucity of fouling in the bay is because of the silt covering the submerged surfaces, particularly at the 2nd station, leading to the prevention of the settlements or establishment of fouling organisms. The seasonal changes in the intensity of fouling assemblages on submerged surfaces in seawater seems to be closely related to seasonal variations in water temperature. The great fouling communities on the buoys and long exposure panels showed a remarkable variety of species and density rather than on short term exposures, which were more dense during warmer months

    Increases in coastal development and human activities leading to sedimentation degrade the quality of water; disturb the natural structure and functions of aquatic communities. The Suez Gulf is a large semi-closed area (~625 km long coastline).The assemblages of bottom fauna were studied qualitatively and quantitatively in the shallow intertidal waters along the western coast of the Suez Gulf. The quality of seawater and sediment structures were analyzed. The distribution of macro-benthos included a total of 38 species of Gastropoda and 9 Bivalvia; and 25 species from the other invertebrates included 7 groups namely, Rhizostoma, Polychaeta, Cirripedia, Amphipoda, Isopoda, Decapoda and Echinodermata. The most dominant group among invertebrate groups was the Polychaeta which included 4 species: Hydroides elegans, Perinereis cultilifera, Perinereis nuntia and Ophelina acuminata. The Cirripedia were represented by 3 species namely, Balanus amphitrite, Chithamalus challengeri and Tetraclita squamosa. The variations in the numerical abundance and biomass of bottom fauna studied between the observation periods and at sampling sites. There was a marked increase in benthos biomass at St. IV (Ras Gharib) yielding an average of 318.8 g/m² in which the gastropod community represented the dominant species in collected samples reaching 270.28 g/m² (84.4% of the total biomass) and numerically numbered 116 ind./m². Veliger larvae of bivalves and gastropods appeared to be present in the plankton for long periods and their production seems to be continuous throughout the year. In the intertidal zone of the Suez Gulf, the values of pH varied within narrow limits. Water temperature and salinity seemed to be important in the distribution and abundance of the macro-benthos communities in the study areas. The organic content in shallow intertidal waters and sediments indicated high values in the central part of the Gulf of Suez

    Numerical Assessment of Reinforced Concrete Beams Strengthened with CFRP Sheets under Impact Loading

    This paper investigates numerically the behavior of Reinforced Concrete (RC) beams, strengthened using Carbon Fiber Reinforced Polymers (CFRP) sheets, subjected to impact loading. Three-dimensional finite element analysis was performed and its results were verified against experimental ones available in the literature showing good agreement. Then, a comprehensive parametric study was performed to investigate the effect of studied parameters on the strengthened RC beams. The main studied parameters were type and size of reinforcing bars, geometric characteristics of externally bonded CFRP sheets (width, length, and thickness), impact velocity, and the position of the impactor with respect to the beam. Results showed that the use of externally bonded CFRP sheets enhanced the beam capacity and failure mode, and reduced the mid-span deflection. Moreover, a reduction in the mid-span deflection was observed due to the use of CFRP bars as internal reinforcement. On the other hand, the deflection was increased due to the increase of the impact velocity, and the change of the impact load position
