    This paper develops a short-run economic damage assessment model. The model contains two sub models. One is the conventional traffic assignment model, which calculates traveling time under the damaged transportation infrastructure. The other is the economic damage assessment model, which determines the decrease in production level in the short run. This method facilitates the identification of critical infrastructure that could reduce the economic damage when the disaster occurs. As a case study, we applied this model to several recovery plans for transport facilities. The results suggest that the proposed comprehensive model should be considered as a prevention plan

    Study on Dynamic Characteristics of a Pile Group Foundation

    The earthquake response of a structure on group of piles is investigated. The test model, which is employed in this study, is a foundation on four piles. Three conditions are prepared for examination of the contact effects of the foundation bottom and the backfilling effects on dynamic characteristics of the foundation on the piles. Forced vibration tests are carried out for ascertaining the impedance functions as the inertial interaction, and earthquake observations for the earthquake input motion as the kinematic interaction. On the basis of the test and the observation results, the correlation analyses are executed for examining the applicability of the analytical method based on the substructure method in which the three-dimensional wave propagation theory is applied for calculation of the Green\u27s functions. The analytical method is concluded to have sufficient applicability for the practical design procedure of a structure on pile group

    Ventricular fibrillation after bortezomib therapy in a patient with systemic amyloidosis

    A 64-year-old female was diagnosed with systemic amyloidosis associated with multiple myeloma. Bortezomib and dexamethasone-therapy was initiated; however, she developed lethal ventricular fibrillation (VF) and cardiac arrest after 84 hours of therapy. Cardiopulmonary resuscitation using direct current shocks with epinephrine and amiodarone was initiated but failed to receive cardiac function. Although her arterial pulsations recovered immediately after the injection of vasopressin, she died of heart failure 8 hours after the onset of VF. Cardiac amyloidosis was verified by autopsy. Although the direct association of bortezomib with lethal VF remained to be clarified in our patient, the current report emphasizes on bortezomib as a substantial risk factor for cardiomyocyte damage. The potential risk of lethal events associated with cardiac amyloidosis should be carefully considered during bortezomib treatment for patients with AL amyloidosis

    Effectiveness of Isoprothiolane in Preventing Fatty Liver in Dairy Cows

    In this study, we evaluated the effectiveness of isoprothiolane in preventing fatty liver in dairy cows. Eight pregnant Holstein cows with high body condition scores were studied. Four cows were administered isoprothiolane via feed at 10 g/day/head from 21 days before delivery (treated group), while the remaining four cows received no treatment (non-treated group). Liver samples from both groups were obtained 21 days before delivery and 2 days following delivery. No accumulation of lipid droplets in hepatocytes was observed in any cows prior to delivery. Following delivery, the accumulation of lipid droplets in hepatocytes was significantly reduced in the treated group compared to the non-treated group. The number of mitochondria per unit area was also significantly smaller in the treated group than in the non-treated group. Blood and biochemical test results from cows in the treated group showed an increase in mGOT levels and stable intrahepatic ATP production. These results suggest that isoprothiolane treatment intensified beta-oxidation in liver mitochondria, thereby activating the TCA cycle and maintaining intrahepatic ATP production

    Genetic insights into ossification of the posterior longitudinal ligament of the spine

    Ossification of the posterior longitudinal ligament of the spine (OPLL) is an intractable disease leading to severe neurological deficits. Its etiology and pathogenesis are primarily unknown. The relationship between OPLL and comorbidities, especially type 2 diabetes (T2D) and high body mass index (BMI), has been the focus of attention; however, no trait has been proven to have a causal relationship. We conducted a meta-analysis of genome-wide association studies (GWASs) using 22,016 Japanese individuals and identified 14 significant loci, 8 of which were previously unreported. We then conducted a gene-based association analysis and a transcriptome-wide Mendelian randomization approach and identified three candidate genes for each. Partitioning heritability enrichment analyses observed significant enrichment of the polygenic signals in the active enhancers of the connective/bone cell group, especially H3K27ac in chondrogenic differentiation cells, as well as the immune/hematopoietic cell group. Single-cell RNA sequencing of Achilles tendon cells from a mouse Achilles tendon ossification model confirmed the expression of genes in GWAS and post-GWAS analyses in mesenchymal and immune cells. Genetic correlations with 96 complex traits showed positive correlations with T2D and BMI and a negative correlation with cerebral aneurysm. Mendelian randomization analysis demonstrated a significant causal effect of increased BMI and high bone mineral density on OPLL. We evaluated the clinical images in detail and classified OPLL into cervical, thoracic, and the other types. GWAS subanalyses identified subtype-specific signals. A polygenic risk score for BMI demonstrated that the effect of BMI was particularly strong in thoracic OPLL. Our study provides genetic insight into the etiology and pathogenesis of OPLL and is expected to serve as a basis for future treatment development
