    Efficacy of Epinastine Hydrochloride for Antigen-Provoked Nasal Symptoms in Subjects with Orchard Grass Pollinosis

    ABSTRACTBackgroundAmong the gramineae species, orchard grass is a typical causative pollen that provokes seasonal rhinitis. The purpose of this study was to examine the protective efficacy of epinastine hydrochloride for signs and symptoms caused by repeated nasal provocation with discs containing orchard grass pollen.MethodsA single-dose, placebo-controlled, double-blind, crossover clinical study was conducted in subjects with orchard grass pollinosis. The pollen challenge was conducted with the use of provocation discs containing orchard grass pollen.ResultsEpinastine hydrochloride suppressed nasal symptoms caused by nasal provocation tests using orchard grass pollen discs. Among the nasal symptoms, the number of sneezing was significantly inhibited 30 minutes and 60 minutes after the administration of epinastine hydrochloride, as compared with placebo. There were no adverse reactions to the study drugs.ConclusionsOur results suggest that nasal provocation tests with discs containing orchard grass pollen is a useful method for evaluating the onset of action of antiallergic drugs. As compared with placebo, epinastine hydrochloride decreased early-phase sneezing and the total nasal symptom score after repeated nasal provocations with orchard grass pollen discs

    Delay of Onset of Symptoms of Japanese Cedar Pollinosis by Treatment with a Leukotriene Receptor Antagonist

    ABSTRACTBackgroundLeukotriene receptor antagonists (LTRAs) are effective for prophylactic treatment of pollinosis based on studies showing that administration of LTRAs prior to or at the start of the pollen season reduces symptoms and QOL disturbance at the peak of pollen dispersal. Two goals of prophylactic treatment of pollinosis are use of fewer types of drugs and delay of onset of symptoms and impairement of QOL. Therefore, this study was performed to determine if pranlukast, a LTRA, met these goals in treatment of pollinosis.MethodsPranlukast or placebo was administered to patients who visited our hospital immediately before the start of Japanese cedar pollen dispersal. The study was performed for 4 weeks as a double blind randomized trial. Subsequently, all patients were given pranlukast for a further 4 weeks from the peak until the end of pollen dispersal. The incidence of symptoms and use of concomitant drugs were investigated from daily nasal allergy records kept by patients. QOL was evaluated using the JRQLQ questionnaire.ResultsIn the double blind period of the study, the percentage of patients who used concomitant drugs for nasal symptoms was significantly lower in the pranlukast group compared to the placebo group. Development of nasal symptoms (sneezing, runny nose and nasal congestion) and disturbance of daily activities were significantly delayed in the pranlukast group. No serious adverse reactions occurred in the pranlukast group and no patient withdrew from treatment with pranlukast.ConclusionsPranlukast is effective for prophylactic treatment of pollinosis

    Thermal and Mechanical Properties of Glass Cullet Mixed with Asphalt as Low-Exothermic Pavement Material

    The asphalt pavement contributes to the heat island phenomenon of the city. Thus in order to decrease the temperature of asphalt pavement and to utilize glass cullet as a recycling material as an aggregate, some tests were carried out. In this study the specimen in which the cullet was entrapped in cold-laid paving materials and non-mixing specimen are made respectively, and the surface temperature of each specimen is measured by the thermal video camera. A set of tests have been conducted and results indicated the clear effect of glass cullet in reducing the radiation temperature of asphalt pavement

    Inhibition of Liver Regeneration in Mice Following Extended Hepatectomy by Transfusion of Lymphokine Activated Killer Cells

    Lymphokine activated killer (LAK) cells can destroy not only tumor cells but also syngeneic liver cells. In this study, the effects of passive transfer of LAK cells on liver regeneration were examined by the 3H-thymidine uptake and bromodeoxyuridine (BrdU) labeling methods after resection of 70% of the volume of the liver. LAK cells were infused 12h after hepatectomy and the effects on regeneration of liver cells were examined 36 h later. The transfusion of LAK cells induced significant inhibition of liver regeneration at a dose of 5-10 x 10(7) cells. Neuraminidase treatment of lymphocytes is desirable to enhance the selective entrapment of LAK cells into the liver. When LAK cells were treated with neuraminidase (0.5 units/ml), and transfused into hepatectomized mice, more potent suppression of liver regeneration was induced in comparison with the same dose of LAK cells. The intraperitoneal injection of recombinant interleukin 2 (rIL-2) after partial hepatectomy also inhibited the regeneration of remnant liver. From these results, lymphocytes such as LAK cells appear to regulate liver regeneration.</p

    The extracellular hemoglobin (Hb) from the polychaete Perinereis aibuhitensis consists of four types of 144 globins and two types of 36 linker chains,having a molecular mass of about 3,500 kDa. There are two types of globin subunits: monomer chain a and disulfide-bondedt rimer AbB. The amino acid sequences of the four globin chains (a,A ,b ,B ) were already reported,previously (Yamanaka,M. et al. Natural Science Research of Tokushima University, 19, 63-92, 2005). The site of disulfide bonds in the globin subunits have been investigated. Each globin chain contains an intrachain disulfide bond between N-terminal and C-terminal Cys residues. In addition,the interchain disulfide bonds were found between chains A and b,and b and B. Therefore,it is elucidated that the chain b is situated at the center of disulfide-bonded trimer,such as A-b-B.The sites of disulfide-bonds determined all could be suitably fitted to the tree dimensional structure of each subunit in a model without stretching or twisting. It was also confirmed that there is no free Cys residue in Peinereis globins. The positions of Cys residues of Perinereis globin sequences were compared wIth those of other 27 chains derived from the homologous Hbs. Among 31 sequences,Cys residues were distributed in six sites. The sites 1 and 2 are located at the N-terminal region of amino acid sequences,the sItes 3 and 4 at the central region,and the sites 5 and 6 at C-terminal region. Furthermore,the Cys distribution was categorized into eight patterns. Perinereis Hb has four patterns I,IV,VII,VIII,be ing lack of the central sites 3 and 4. It should be noted that the pattern II includes the unique globins from Hbs of Lamellibrachia,Riftia and Oligobrachia that carry H2S to the symbiotic bacteria,suggesting that these globin chains might carry H2S in vivo. The phylogenetic tree of 31 globin chains derived from the giant Hbs is divided into two families A and B, as already poited out by us Previously.     Transient Receptor Potential 1 Regulates Capacitative Ca2+ Entry and Ca2+ Release from Endoplasmic Reticulum in B Lymphocytes〉

    Capacitative Ca2+ entry (CCE) activated by release/depletion of Ca2+ from internal stores represents a major Ca2+ influx mechanism in lymphocytes and other nonexcitable cells. Despite the importance of CCE in antigen-mediated lymphocyte activation, molecular components constituting this mechanism remain elusive. Here we demonstrate that genetic disruption of transient receptor potential (TRP)1 significantly attenuates both Ca2+ release-activated Ca2+ currents and inositol 1,4,5-trisphosphate (IP3)-mediated Ca2+ release from endoplasmic reticulum (ER) in DT40 B cells. As a consequence, B cell antigen receptor–mediated Ca2+ oscillations and NF-AT activation are reduced in TRP1-deficient cells. Thus, our results suggest that CCE channels, whose formation involves TRP1 as an important component, modulate IP3 receptor function, thereby enhancing functional coupling between the ER and plasma membrane in transduction of intracellular Ca2+ signaling in B lymphocytes

    Genetic analysis of typical wet-type age-related macular degeneration and polypoidal choroidal vasculopathy in Japanese population

    Age-related macular degeneration (AMD) is a common cause of blindness in the elderly. Caucasian patients are predominantly affected by the dry form of AMD, whereas Japanese patients have predominantly the wet form of AMD and/or polypoidal choroidal vasculopathy (PCV). Although genetic association in the 10q26 (ARMS2/HTRA1) region has been established in many ethnic groups for dry-type AMD, typical wet-type AMD, and PCV, the contribution of the 1q32 (CFH) region seem to differ among these groups.     Amino acid sequences of four types of globin chains a, A, b, and B of the giant Hb from the polychaete Perinereis aibuhitensis, which were collected at the delta of the Yangtze River in the People's Republic of China, were determined by the combination of Edman degradation and RT-PCR method. The primary structures of Perinereis globin chains are compared with those of globins isolated from the polychaete Tylorrhynchus heterochaetus which was collected at the delta of the River Yoshinogawa in Tokushima City. Perinereis globin chains appeared to have 140 amino acid residues for the chain a, 144 for A, 147 for b and B, respectively. These sequences were completely in agreement with the partial cDNA sequences by RT-PCR which covered 119~137 amino acid residues. Some heterogeneities were found for the chain A by protein sequencing. On the other hand, RT-PCR revealed a few polymorphisms in the sequences of chains a, A and b. According to the phylogenetic tree constructed by UPGMA method, Perinereis globin chains were appeared to be separated into two strains A and B at the time later than the separation of vertebrate Mb and Hb but earlier than the separation of the alpha and beta chains of vertebrate Hb. Perinereis globins b and B appeared to be separated from globin A after the time when the trimeric globin chains A, b, and B had been separated clearly from the derivative of globin a according to the phylogenetic tree constructed by NJ method. The homologies between the corresponding globin chains of Perinereis and Tylorrhynchus Hbs were estimated to be within the range of 48~84%
