    De novo acute megakaryoblastic leukemia with p210 BCR/ABL and t(1;16) translocation but not t(9;22) Ph chromosome

    Acute megakaryoblastic leukemia (AMKL) is a type of acute myeloid leukemia (AML), in which majority of the blasts are megakaryoblastic. De novo AMKL in adulthood is rare, and carries very poor prognosis. We here report a 45-year-old woman with de novo AMKL with BCR/ABL rearrangement and der(16)t(1;16)(q21;q23) translocation but negative for t(9;22) Ph chromosome. Upon induction chemotherapy consisting of homoharringtonine, cytarabine and daunorubicin, the patient achieved partial hematological remission. The patient was then switched to imatinib plus one cycle of CAG regimen (low-dose cytarabine and aclarubicin in combination with granulocyte colony-stimulating factor), and achieved complete remission (CR). The disease recurred after 40 days and the patient eventually died of infection. To the best of our knowledge, this is the first report of de novo AMKL with p210 BCR/ABL and der(16)t(1;16)(q21;q23) translocation but not t(9;22) Ph chromosome

    A strange star scenario for the formation of isolated millisecond pulsars

    According to the recycling model, neutron stars in low-mass X-ray binaries were spun up to millisecond pulsars (MSPs), which indicates that all MSPs in the Galactic plane ought to be harbored in binaries. However, about 20%20\% Galactic field MSPs are found to be solitary. To interpret this problem, we assume that the accreting neutron star in binaries may collapse and become a strange star when it reaches some critical mass limit. Mass loss and a weak kick induced by asymmetric collapse during the phase transition (PT) from neutron star to strange star can result in isolated MSPs. In this work, we use a population-synthesis code to examine the PT model. The simulated results show that a kick velocity of ∼60 km s−1\sim60~{\rm km~s}^{-1} can produce ∼6×103\sim6\times10^3 isolated MSPs and birth rate of ∼6.6×10−7 yr−1\sim6.6\times10^{-7} {\rm ~yr}^{-1} in the Galaxy, which is approximately in agreement with predictions from observations.     Meta-analysis of effects of long-term exposure to PM2.5 on C-reactive protein levels

    BackgroundFine particulate matter (PM2.5) is a serious air pollutant associated with elevated levels of C-reactive protein (CRP), an inflammatory indicator.ObjectiveTo assess the potential impacts of long-term exposure to PM2.5 on CRP levels based on previous epidemiological studies.MethodsPubMed, Embase, Web of Science, CNKI, and Wanfang databases were searched to screen the cohort studies published from January 1, 2000 to January 1, 2022 on the effects of long-term exposure to PM2.5 on CRP levels. "Fine Particulate Matter", "PM2.5", "Particulate Air Pollutants", "Ambient Particulate Matter", "CRP", "C-reactive Protein", and "High Sensitivity C-reactive Protein" in English or Chinese were the key words used in the search. The percentage change in CRP level per 10 μg·m−3 increase in PM2.5 concentration in each study was extracted, followed by meta-analysis, subgroup analysis, and sensitivity analysis.ResultsA total of 1241 articles were retrieved, and 7 articles were included. Random-effects models were used to merge the included data, and it was found that the percentage of CRP level increased by 10.41% (95%CI: 2.24%-18.57%, P<0.05), when PM2.5 concentration increased by 10 μg·m−3, І2=84.2%. The subgroup analysis conducted with grouping based on the annual mean concentration of PM2.5 long-term exposure showed that the intra-group heterogeneity was significantly reduced in the <15 μg·m−3 and the 15- μg·m−3 groups, and the subgroup forest analysis showed differences between the two groups. The results of sensitivity analysis showed that there was a high degree of heterogeneity among the 7 studies, and the 2 papers with the highest annual average PM2.5 concentration were the sources of heterogeneity. The Egger test and the funnel plot indicated that no obvious publication bias was found.ConclusionLong-term exposure to PM2.5 can raise levels of CRP in human body

    Research Progress on Ferroptosis as a Therapeutic Strategy in Renal Cell Carcinoma

    Renal cell carcinoma (RCC) is a common type of kidney cancer in adults and constitutes approximately 90% of all renal malignancies. Although advancements have been made in the treatment of RCC, the 5 -year survival rate is still low, and new treatment modalities are still required. Ferroptosis is an iron-dependent programmed cell death caused by the accumulation of lipid peroxide products. Recent studies revealed the involvement of ferroptosis metabolism, lipid peroxidation, and System XC-GSH-GPX4 shafts as major mechanisms closely related to RCC progression. Nanoparticles in combination with small molecular ferroptosis induction agents have the advantages of solubility, targeted enhancement, low systemic toxicity, controllable drug control, and synergy advantage in emerging combination therapies. In the future, it is possible to be used in nano treatment. The relationship between ferroptosis-related mechanisms and RCC progression and its role in the treatment could provide novel treatment strategies for patients with advance-stage RCC

    Three-Dimensional Reconstruction of Thoracic Structures: Based on Chinese Visible Human

    We managed to establish three-dimensional digitized visible model of human thoracic structures and to provide morphological data for imaging diagnosis and thoracic and cardiovascular surgery. With Photoshop software, the contour line of lungs and mediastinal structures including heart, aorta and its ramus, azygos vein, superior vena cava, inferior vena cava, thymus, esophagus, diaphragm, phrenic nerve, vagus nerve, sympathetic trunk, thoracic vertebrae, sternum, thoracic duct, and so forth were segmented from the Chinese Visible Human (CVH)-1 data set. The contour data set of segmented thoracic structures was imported to Amira software and 3D thorax models were reconstructed via surface rendering and volume rendering. With Amira software, surface rendering reconstructed model of thoracic organs and its volume rendering reconstructed model were 3D reconstructed and can be displayed together clearly and accurately. It provides a learning tool of interpreting human thoracic anatomy and virtual thoracic and cardiovascular surgery for medical students and junior surgeons

    Protective immunity against Trichinella spiralis infection induced by TsNd vaccine in mice

    BACKGROUND: We have previously reported that Trichinella spiralis Nudix hydrolase (TsNd) bound to intestinal epithelial cells (IECs), and vaccination of mice with recombinant TsNd protein (rTsNd) produced a partial protective immunity. The aim of this study was to investigate the immune protection induced by TsNd DNA vaccine. METHODS: The full-length cDNA sequence of TsNd gene was cloned into pcDNA3.1 and used to immunize BALB/c mice by intramuscular injection. Transcription and expression of TsNd were detected by RT-PCR and IFT. The levels of specific IgA, IgG, IgG1 and IgG2a, and cytokines were assayed by ELISA at weeks 0, 6 and 8 post-immunization. The immune protection of TsNd DNA vaccine against challenge infection was investigated. RESULTS: Immunization of mice with TsNd DNA elicited a systemic Th1/Th2 immune response and a local mucosal IgA response. The in vitro transcription and expression of TsNd gene was observed at all developmental stages of T. spiralis (ML, IIL, AW and NBL). Anti-rTsNd IgG levels were increased after immunization and levels of IgG1 were obviously higher than that of IgG2a. Intestinal specific IgA levels of immunized mice were significantly higher than those of vector and PBS control mice. Cytokine profiling also showed a significant increase in Th1 (IFN-γ, IL-2) and Th2 (IL-4, 10) responses in splenocytes of immunized mice on stimulation with rTsNd. Vaccination of mice with pcDNA3.1-TsNd displayed a 40.44% reduction in adult worms and a 53.9% reduction in larval burden. CONCLUSIONS: TsNd DNA induced a mixed Th1/Th2 immune response and partial protection against T. spiralis infection in mice

    B cell receptor signaling pathway involved in benign lymphoepithelial lesions of the lacrimal gland

    AIM: To detect the expression of B cell receptor signaling pathway (BCRSP) in lacrimal gland benign lymphoepithelial lesions (LGBLEL). METHODS: Gene microarray was used to compare whole-genome expression in lacrimal gland tissues from LGBLEL patients to tissues from orbital cavernous hemangioma (control tissues). Expression of BCRSP was confirmed by polymerase chain reaction (PCR) and immunohistochemistry. RESULTS: The expression of 22 genes of the BCRSP increased significantly in LGBLEL patients. PCR analysis showed that CD22, CR2, and BTK were all highly expressed in LGBLEL tissues. Immunohistochemical analysis showed that CR2 protein was present in LGBLEL, but CD22 and BTK proteins were negative. CR2, CD22, and BTK were not observed in the orbital cavernous hemangiomas with either PCR or immunohistochemistry. CONCLUSION: BCRSP might be involved in the pathogenesis of LGBLEL

    This is the first of a two-part series on the watershed hydrological experimental system (WHES). Since the foundational stage and developmental stage of hydrological basin study with a duration of more than ca. one century, facing with the changing environment and, the declined risk of field study while the catchment hydrology is trapped in a theoretical impasse, a third phase of renovation on hydrological experiments seems ready to come out inevitably. Learned from Chinese decades’ experiences on the field basin study for the question of what is wrong with the status quo, our exploratory idea is reported in this part. From the viewpoint of general system theory based on the paralleled concepts of the ancient Chinese and the Western, it is considered that the adequate method should face the characters of the complex dynamic system instead of previous static, linear system. From the viewpoint of another philosophical paralleled concept of the Middle Way, it should also face the operation and organizing of the mesoscopic systems for the organized complexity. Then, a framework of WHES is suggested with its organization based on the strategy of constrain complexity and add complexity and on the strategy of manipulation including the artificial-natural and controlled-natural objects. Such a trial framework, the Chuzhou WHES, is reported including the suggested critical zone experimental block (CZEB) instead of the experimental basin (EB) in the last decades

    This is the second of a two-part series on the watershed hydrological experimental system (WHES) aimed at practice and test of it at Chuzhou Hydrology Laboratory. It constitutes both natural and artificial entities of different scales, within which two typical main subjects are reviewed here. First is a natural watershed Nandadish, which is subjected to be a Critical Zone Experimental Block, under manipulation strategy of constrain complexity to compare with the pure natural watersheds, it is the controlled-natural as we termed. Second is an artificial catchment Hydrohill, under the strategy of add complexity to compare with the simple artificial lysimeters, it is the artificial-natural as we termed. The constructions and instrumentations of these experimental catchments are reviewed, especially their renovation version during recent years after a long abandonment. Some results get during the operation of Chuzhou WHES are outlined here as well
