    EFFECTS OF DIFFERENT SALINITY LEVELS ON PHYSIOLOGICAL AND HEMATOLOGICAL RESPONSE OF ROCK BREAM Oplegnathus fasciatus

    Rock bream Oplegnathus fasciatus is one of economically important marine fish species in East Asia. However, lack of information about the salinity tolerance of rock bream related to its physiological response made this issue were needed to be studied. The present study was conducted to determine the effects of different salinity levels on physiological and hematological response of rock bream in order to obtain its salinity tolerance. Twelve rock breams (total length: 26.9 ± 0.6 cm, body weight: 477.3 ± 61.9 g) were used for experiments. Four experimental groups with three replications were conducted to measure the effects of salinity (5, 15, 25, and 35 practical salinity unit (psu)) on physiological and hematological response of rock bream. Fish were stocked into the chamber inside the closed recirculation system. At the end of each experiment, blood samples were collected. The study revealed that lower salinity exposure had tendency to decrease the physical and chemical properties of blood in rock bream. The value of Na+, Cl-, Ca, Mg, and osmolality showed tendency to decrease with lowering salinity, while cortisol and glucose showed tendency to increase from 35 psu to low salinity environment, indicating the enhancement of fish stress and resulted in fish mortality at 5 psu. The lowest cortisol value was 76.3 ng/mL in 25 psu, and the highest value was 188.8 ng/mL in 5 psu. Meanwhile, the lowest glucose value was 35.3 mg/dL and the highest value was 166.7 mg/dL (P<0.05).     EFFECTS OF SALINITY ON OXYGEN CONSUMPTION AND BLOOD PROPERTIES OF YOUNG GREY MULLETS Mugil cephalus

    Oxygen consumption (OC) is one of important factors in aquaculture activities, as the oxygen is a vital condition for all the organisms living in the water and having an aerobic type of respiration. OC is the preferred method for measuring and reporting the metabolic rate in fish. The aims of this study were to evaluate the effects of salinity on OC and blood properties of grey mullets. Five experimental groups were conducted to measure OC and blood properties of grey mullets Mugil cephalus (BW: 187.9 ± 45.8 g) according to salinity (30→0 psu, 0→30 psu) changes; SDS: fish reared in seawater (SW, 30 psu) directly shifted to SW, SGF: SW fish gradually shifted to freshwater (FW, 0 psu), SDF: SW fish directly shifted to FW, FDF: FW fish directly shifted to FW, and FDS: FW fish directly shifted to SW. The result showed that OC tended to decrease in the groups of SW fish shifted to FW showing 194.5 mg O2/kg/h at 25°C in SDS to 82.4 mg O2/kg/h at 15°C in SGF. On the contrary, OC increased in the groups of FW fish shifted to SW showing 80.5 mg O2/kg/h at 15°C in FDF to 184.0 mg O2/kg/h at 25°C in FDS. Cortisol levels at the end of experiments were rapidly increased with the lowering salinities in SW fish shifted to FW showing 20.6 ng/mL in SDS to 316.2 ng/mL in SDF, while those were decreased with the increasing salinities in FW fish shifted to SW showing 40.2 ng/mL in FDF to 10.3 ng/mL in FDS. However, glucose levels showed no significant differences among all experimental groups. Based on the information from this study, aquaculture of grey mullet might be applied or developed in freshwater due to its osmotic adaptation ability

    PMC-12, a Prescription of Traditional Korean Medicine, Improves Amyloid β

    PMC-12 is a prescription used in traditional Korean medicine that consists of a mixture of four herbal medicines, Polygonum multiflorum, Rehmannia glutinosa, Polygala tenuifolia, and Acorus gramineus, which have been reported to have various pharmacological effects on age-related neurological diseases. In the present study, we investigated whether PMC-12 improves cognitive deficits associated with decreased neuroinflammation in an amyloid-β-(Aβ-) induced mouse model and exerts the antineuroinflammatory effects in lipopolysaccharide-(LPS-) stimulated murine BV2 microglia. Intracerebroventricular injection of Aβ25-35 in mice resulted in impairment in learning and spatial memory, whereas this was reversed by oral administration of PMC-12 (100 and 500 mg/kg/day) in dose-dependent manners. Moreover, PMC-12 reduced the increase of Aβ expression and activation of microglia and astrocytes in the Aβ25-35-injected brain. Furthermore, quantitative PCR data showed that inflammatory mediators were significantly decreased by administration of PMC-12 in Aβ-injected brains. Consistent with the in vivo data, PMC-12 significantly reduced the inflammatory mediators in LPS-stimulated BV2 cells without cell toxicity. Moreover, PMC-12 exhibited anti-inflammatory properties via downregulation of ERK, JNK, and p38 MAPK pathways. These findings suggest that the protective effects of PMC-12 may be mediated by its antineuroinflammatory activities, resulting in the attenuation of memory impairment; accordingly, PMC-12 may be useful in the prevention and treatment of AD

    The Traditional Herbal Medicine, Dangkwisoo-San, Prevents Cerebral Ischemic Injury through Nitric Oxide-Dependent Mechanisms

    Dangkwisoo-San (DS) is an herbal extract that is widely used in traditional Korean medicine to treat traumatic ecchymosis and pain by promoting blood circulation and relieving blood stasis. However, the effect of DS in cerebrovascular disease has not been examined experimentally. The protective effects of DS on focal ischemic brain were investigated in a mouse model. DS stimulated nitric oxide (NO) production in human brain microvascular endothelial cells (HBMECs). DS (10–300 μg/mL) produced a concentration-dependent relaxation in mouse aorta, which was significantly attenuated by the nitric oxide synthase (NOS) inhibitor L-NAME, suggesting that DS causes vasodilation via a NO-dependent mechanism. DS increased resting cerebral blood flow (CBF), although it caused mild hypotension. To investigate the effect of DS on the acute cerebral injury, C57/BL6J mice received 90 min of middle cerebral artery occlusion followed by 22.5 h of reperfusion. DS administered 3 days before arterial occlusion significantly reduced cerebral infarct size by 53.7% compared with vehicle treatment. However, DS did not reduce brain infarction in mice treated with the relatively specific endothelial NOS (eNOS) inhibitor, N5-(1-iminoethyl)-L-ornithine, suggesting that the neuroprotective effect of DS is primarily endothelium-dependent. This correlated with increased phosphorylation of eNOS in the brains of DS-treated mice. DS acutely improves CBF in eNOS-dependent vasodilation and reduces infarct size in focal cerebral ischemia. These data provide causal evidence that DS is cerebroprotective via the eNOS-dependent production of NO, which ameliorates blood circulation

Ten percent of extra pulmonary tuberculosis localizes to the bones and joints, and 56% of such cases affect the spine. We treated a childhood case of spinal tuberculosis misdiagnosed as muscular dystrophy in a patient without specific constitutional symptoms.     Identifying genetic variants underlying medication-induced osteonecrosis of the jaw in cancer and osteoporosis: a case control study

    Background Bisphosphonate-induced osteonecrosis of the jaw (BRONJ) presents with a typical pattern of jaw necrosis in patients who have been prescribed bisphosphonates (BPs) and other antiangiogenetic drugs to treat osteoporosis or bone-related complications of cancer. Methods This study divided 38 patients with BRONJ into two groups according to the prescribing causes: cancer (n = 13) and osteoporosis (n = 25), and underwent whole exome sequencing and compared them with normal controls (n = 90). To identify candidate genes and variants, we conducted three analyses: a traditional genetic model, gene-wise variant score burden, and rare-variant analysis methods. Results The stop-gain mutation (rs117889746) of the PZP gene in the BRONJ cancer group was significantly identified in the additive trend model analysis. In the cancer group, ARIDS, HEBP1, LTBP1, and PLVAP were identified as candidate genes. In the osteoporosis group, VEGFA, DFFA, and FAM193A genes showed a significant association. No significant genes were identified in the rare-variant analysis pipeline. Biologically accountable functions related to BRONJ occurrence-angiogenesis-related signaling (VEGFA and PLVAP genes), TGF-β signaling (LTBP1 and PZP genes), heme toxicity (HEBP1) and osteoblast maturation (ARIDS)-were shown in candidate genes.
