Transcriptional profiling of ParA and ParB mutants in actively dividing cells of an opportunistic human pathogen Pseudomonas aeruginosa.

Accurate chromosome segregation to progeny cells is a fundamental process ensuring proper inheritance of genetic material. In bacteria with simple cell cycle, chromosome segregation follows replication initiation since duplicated oriC domains start segregating to opposite halves of the cell soon after they are made. ParA and ParB proteins together with specific DNA sequences are parts of the segregation machinery. ParA and ParB proteins in Pseudomonas aeruginosa are important for optimal growth, nucleoid segregation, cell division and motility. Comparative transcriptome analysis of parA null and parB null mutants versus parental P. aeruginosa PAO1161 strain demonstrated global changes in gene expression pattern in logarithmically growing planktonic cultures. The set of genes similarly affected in both mutant strains is designated Par regulon and comprises 536 genes. The Par regulon includes genes controlled by two sigma factors (RpoN and PvdS) as well as known and putative transcriptional regulators. In the absence of Par proteins, a large number of genes from RpoS regulon is induced, reflecting the need for slowing down the cell growth rate and decelerating the metabolic processes. Changes in the expression profiles of genes involved in c-di-GMP turnover point out the role of this effector in such signal transmission. Microarray data for chosen genes were confirmed by RT-qPCR analysis. The promoter regions of selected genes were cloned upstream of the promoter-less lacZ gene and analyzed in the heterologous host E. coliΔlac. Regulation by ParA and ParB of P. aeruginosa was confirmed for some of the tested promoters. Our data demonstrate that ParA and ParB besides their role in accurate chromosome segregation may act as modulators of genes expression. Directly or indirectly, Par proteins are part of the wider regulatory network in P. aeruginosa linking the process of chromosome segregation with the cell growth, division and motility

Dokonano również analizy oferty pakietów pobytowych, których głównym elementem są usługi SPA & Wellness na przykładzie trzech wybranych obiektów: Hotelu Farmona Business & SPA, Hotelu Mercure Racławice Dosłońce Conference & Spa**** oraz Hotelu**** Medical Spa Malinowy Zdrój. W pracy przedstawiono rys historyczny branży SPA & Wellness, szczegółową charakterystykę obiektów należących do tej branży, sylwetki gości SPA oraz różnice pomiędzy hotelem SPA, a hotelem ze SPA.The issue of this master thesis concerns of the perception of SPA & Wellness services as a tourism product in hotels SPA. The paper is an attempt to analyze the meaning of SPA & Wellness in the market of tourist services, hotel services in particular. It presents a theoretical approach to the construction of the tourist product and the characteristics of hotel services with the division into basic services and additional services. Based on the tourist motives of the hotel’s guests were unable to prove the great importance of SPA & Wellness in the hospitality business, including treating these services as essential services and the main theme of arrival guests to the hotel spa facilities. On the example of three selected objects: Hotel Farmona Business & SPA Hotel ****Mercure Racławice Dosłońce Conference & Spa and Hotel**** Medical Spa Malinowy Zdrój were analyzed stay packages offers whose main element are SPA & Wellness services. The paper presents historical industry SPA & Wellness, a detailed characterization of objects belonging to the SPA industry, profiles of SPA visitors and differences between the SPA hotel and the hotel with SPA

Problematyka niniejszej pracy magisterskiej dotyczy koncepcji zarządzania talentami we współczesnych organizacjach oraz porusza kwestię metod szkoleniowych stosowanych podczas programów rozwojowych dla absolwentów uczelni wyższych. W pracy przedstawione zostało teoretyczne podejście do zarządzania talentami, główne elementy procesu zarządzania talentami takie jak: identyfikacja i pozyskanie talentu, doskonalenie i rozwój oraz wyjście z organizacji, a także modelowe ujęcia tego procesu. Praca zawiera opis przebiegu programu rozwojowego Ekstraklasa realizowanego przez firmę Tesco. Opisane zostały metody identyfikacji i rekrutacji talentów do programu oraz kluczowe kompetencje, które stanowią o atrakcyjności kandydata. Ukazano również przebieg doskonalenia, metody szkolenia i rozwoju potencjału utalentowanego uczestnika programu, które prowadzą go do objęcia stanowiska kierowniczego.The objective of this master’s thesis is to provide a description of the concept of talent management applied to contemporary organizations. It also addresses the issue of training methods used during the course of development programs aimed at university graduates. Throughout this thesis the reader will gain an understanding of a theoretical approach to talent management, the main elements of the talent management process such as both talent identification and acquisition, but also development and improvement, resignation from work and the model approach to this process. By way of example, this thesis further contains a thorough description of the ‘Ekstraklasa’ development program implemented by Tesco. The methods of talent identification and recruitment are also accounted for as well as the key competencies responsible for the determining the candidate’s attractiveness for the company. Finally, the last but not the least important part of the thesis concerns the process of improvement, training methods and development of talented participants of the program which would eventually lead such individuals to managerial positions

Sickle cell anemia

Anemia sierpowata jest jedną z najczęstszych i najważniejszych ludzkich chorób genetycznych wywołanych mutacją punktową, a hemoglobina S (HbS) pełniąca kluczową role w rozwoju tej choroby jest jednym z najpowszechniejszych wariantów nieprawidłowych hemoglobin występujących u ludzi. Osoby cierpiące na tę przypadłość dziedziczą dwa nieprawidłowe geny hemoglobiny, po jednym od każdego rodzica. Przebieg kliniczny niedokrwistości sierpowatej jest ciężki i uciążliwy, ze względu na obecność wielu ostrych oraz przewlekłych powikłań. Liczne badania dowiodły, że obecnie najskuteczniejszymi formami łagodzenia tej choroby jest transfuzja krwi oraz zastosowanie w leczeniu hydroksymocznika, jednak takie zabiegi nie mogą być praktykowane przez dłuższy okres czasu.Celem pracy jest ogólne zobrazowanie omawianego problemu. W szczególności analizie poddane zostały kluczowe aspekty choroby: zasięg występowania, dziedziczenie, patofizjologia oraz możliwości leczenia za pomocą modyfikacji genetycznych. Potrzebne są dalsze badania w celu opracowania efektywnych metod łagodzenia objawów klinicznych, które bezpośrednio będą wpływać na konkretne zmiany patofizjologiczne oraz powikłania anemii sierpowatej.Sickle-cell anemia is the most common and most important human genetic disease caused by point mutation. Hemoglobin S (HbS) plays a key role in the development of this disease and is one of the most common variant of abnormal hemoglobin in humans. People who suffer from this ailment inherit two genes coding abnormal hemoglobin, one from each parent. Clinical symptoms of sickle cell disease are heavy and burdensome, due to a variety of acute and chronic complications. Several studies have shown that currently the most effective form of alleviating this disease is a blood transfusion and treatment with hydroxyurea. However, this treatment can not be practiced for a long period of time.The main aim of this work is a general presentation of the problem. The analysis covered key aspects of the disease: extent of occurrence, inheritance, pathophysiology and emerging options of treatment by genetic modification. Further studies are needed to develop effective curative methods and alleviate clinical symptoms, which will affect the specific pathophysiological changes and complications of sickle cell anemia

The paper is an attempt to analyze the practical aspects of Human Resource Management, especially the process of employees recruitment. It describes not only a theoretical approach to workforce planning, recruitment procedures and methods of selection of candidates but also a practical use. Practical use of these methods and procedures is presented based on analysis of the process of employees in one of the Spa& Wellness companies- Hotel**** Malinowy Zdrój in Solec Zdrój. The second section presents a history of Spa & Wellness from antiquity to modern times and gives information about kinds of the objects belonging to the Spa & Wellness industry

The effect of different solvents and number of extraction steps on the polyphenol content and antioxidant capacity of basil leaves (Ocimum basilicum L.) extracts

The objectives of this study were to determine best conditions for the extraction of phenolic compounds from fresh, frozen and lyophilized basil leaves. The acetone mixtures with the highest addition of acetic acid extracted most of the phenolic compounds when fresh and freeze-dried material have been used. The three times procedure was more effective than once shaking procedure in most of the extracts obtained from fresh basil leaves – unlike the extracts derived from frozen material. Surprisingly, there were not any significant differences in the content of phenolics between the two used procedures in the case of lyophilized basil leaves used for extraction. Additionally, the positive correlation between the phenolic compounds content and antioxidant activity of the studied extracts has been noted. It is concluded that the acetone mixtures were more effective than the methanol ones for polyphenol extraction. The number of extraction steps in most of the cases was also a statistically significant factor affecting the yield of phenolic extraction as well as antioxidant potential of basil leaf extracts

We give sufficient conditions, as well as some necessary conditions, for the Orlicz-Lorentz space Δθ,ω to have the weak-star uniform Kadec-Klee property. These results generalize the characterization of the weak-star uniform Kadec-Klee property in the Lorentz space Δω = Lω,1 due to Dilworth and Hsu. © 2007 Cambridge Philosophical Society
