    Epigenetic inheritance based evolution of antibiotic resistance in bacteria

    BACKGROUND: The evolution of antibiotic resistance in bacteria is a topic of major medical importance. Evolution is the result of natural selection acting on variant phenotypes. Both the rigid base sequence of DNA and the more plastic expression patterns of the genes present define phenotype. RESULTS: We investigated the evolution of resistant E. coli when exposed to low concentrations of antibiotic. We show that within an isogenic population there are heritable variations in gene expression patterns, providing phenotypic diversity for antibiotic selection to act on. We studied resistance to three different antibiotics, ampicillin, tetracycline and nalidixic acid, which act by inhibiting cell wall synthesis, protein synthesis and DNA synthesis, respectively. In each case survival rates were too high to be accounted for by spontaneous DNA mutation. In addition, resistance levels could be ramped higher by successive exposures to increasing antibiotic concentrations. Furthermore, reversion rates to antibiotic sensitivity were extremely high, generally over 50%, consistent with an epigenetic inheritance mode of resistance. The gene expression patterns of the antibiotic resistant E. coli were characterized with microarrays. Candidate genes, whose altered expression might confer survival, were tested by driving constitutive overexpression and determining antibiotic resistance. Three categories of resistance genes were identified. The endogenous β-lactamase gene represented a cryptic gene, normally inactive, but when by chance expressed capable of providing potent ampicillin resistance. The glutamate decarboxylase gene, in contrast, is normally expressed, but when overexpressed has the incidental capacity to give an increase in ampicillin resistance. And the DAM methylase gene is capable of regulating the expression of other genes, including multidrug efflux pumps. CONCLUSION: In this report we describe the evolution of antibiotic resistance in bacteria mediated by the epigenetic inheritance of variant gene expression patterns. This provides proof in principle that epigenetic inheritance, as well as DNA mutation, can drive evolution

    Effects of environmental impact and nutrition labelling on food purchasing: An experimental online supermarket study

    Nutrition labels and ecolabels can support consumers to make healthier and more sustainable choices, and the former is now widespread. But there is no information on the impact of ecolabels in the presence of nutrition labels. The aims of this study were primarily to examine whether (1) ecolabels are effective at promoting sustainable purchasing behaviour if presented alongside nutrition labels; (2) and secondarily, whether nutrition labels are effective at promoting healthier purchasing if presented alongside ecolabels. Participants (N = 2730) visited an experimental online supermarket platform, and were randomised to see products with (1) environmental impact labels only; (2) nutrition (NutriScore) labels only; (3) both environmental and nutrition labels; (4) no labels. Linear regressions compared the mean environmental impact scores (EIS; primary outcome) and health scores of products in participants’ shopping baskets across each condition. Compared to control (no labels) there were significant reductions in the EIS when environmental impact labels were presented: Alone (−1.3, 95%CI: −2.3 to −0.4) or With nutrition labels (−2.0, 95%CI: −2.9 to −1.0), with no evidence of differences in effectiveness between these two conditions. There was no evidence of an impact of nutrition labels on either the EIS or the healthiness of purchases, both when nutrition labels were shown alone and when ecolabels were also present. Environmental impact labels may be effective at encouraging more sustainable purchases alone or when used alongside nutrition labels. This adds to the evidence base on the feasibility and effectiveness of environmental impact labelling as an important measure to change dietary behaviour to improve planetary health

    Speckle Suppression with the Project 1640 Integral Field Spectrograph

    Project 1640 is a high-contrast imaging instrument recently commissioned at Palomar observatory. A combination of a coronagraph with an integral field spectrograph (IFS), Project 1640 is designed to detect and characterize extrasolar planets, brown dwarfs, and circumstellar material orbiting nearby stars. In this paper, we present our data processing techniques for improving upon instrument raw sensitivity via the removal of quasi-static speckles. Our approach utilizes the chromatic image diversity provided by the IFS in combination with the locally-optimized combination of images (LOCI) algorithm to suppress the intensity of residual contaminating light in close angular proximity to target stars. We describe the Project 1640 speckle suppression pipeline (PSSP) and demonstrate the ability to detect companions with brightness comparable to and below that of initial speckle intensities using on-sky commissioning data. Our preliminary results indicate that suppression factors of at least one order of magnitude are consistently possible, reaching 5σ5\sigma contrast levels of 2.1×10−52.1\times10^{-5} at 1\arcsec in the H-band in 20 minutes of on-source integration time when non-common-path errors are reasonably well-calibrated. These results suggest that near-infrared contrast levels of order ≈10−7\approx10^{-7} at subarcsecond separations will soon be possible for Project 1640 and similarly designed instruments that receive a diffraction-limited beam corrected by adaptive optics (AO) systems employing deformable mirrors with high actuator-density.Comment: accepted to Ap
