Determination of itraconazole and hydroxyitraconazole in plasma by use of liquid chromatography-tandem mass spectrometry with on-line solid-phase extraction

    Get PDF
In this paper a method for the simultaneous quantification of the anti-fungal drug itraconazole and its co-active metabolite hydroxyitraconazole in plasma employing liquid chromatography tandem-mass spectrometry and automated solid-phase extraction is described. The method proved rugged, enables short turn-around times and is highly specific. Since there is growing evidence for the importance of therapeutic drug monitoring of itraconazole in the prophylaxis and treatment of invasive fungal infections, the method described here is of interest for a large number of tertiary care hospital laboratories

Light-cone distribution amplitudes of the baryon octet

    Get PDF
We present results of the first ab initio lattice QCD calculation of the normalization constants and first moments of the leading twist distribution amplitudes of the full baryon octet, corresponding to the small transverse distance limit of the associated S-wave light-cone wave functions. The P-wave (higher twist) normalization constants are evaluated as well. The calculation is done using Nf=2+1N_f=2+1 flavors of dynamical (clover) fermions on lattices of different volumes and pion masses down to 222 MeV. Significant SU(3) flavor symmetry violation effects in the shape of the distribution amplitudes are observed.Comment: Update to the version published in JHE

A convenient and economic approach to achieve SI-traceable reference values to be used in drinking-water interlaboratory comparisons

    Get PDF
Abstract Metrologically traceable reference values add an essential benefit to interlaboratory comparisons: unlike consensus values, they can be used to establish national and international comparability. Furthermore, the participating laboratories obtain a reliable and unbiased benchmark to check their results for accuracy. Usually, metrologically traceable reference values are obtained by so-called primary methods which demand excessive efforts at great expense. Within the framework of two national drinkingwater interlaboratory comparisons (proficiency testing rounds), a new approach to provide metrologically traceable reference values was applied. It is solely based on existing data which were collected during the comparison itself. Lead (Pb) measurements serve as an example to show how metrologically traceable reference values were derived from the lead amount added during sample preparation and the amount of lead already present in the drinking-water matrix used to prepare these samples. Within this approach, the matrix content is calculated in a way similar to a standard addition experiment. An uncertainty budget for the reference value was set up which describes the link to the corresponding SI units. Isotope dilution mass spectrometry (IDMS) as a primary method was used to validate this approach in the case of cadmium, chromium, copper, lead, and nickel

The role of stakeholder involvement in the evolving EU HTA process:Insights generated through the European Access Academy’s multi-stakeholder pre-convention questionnaire

Involvement of all relevant stakeholders will be of utmost importance for the success of the developing EU HTA harmonization process. A multi-step procedure was applied to develop a survey across stakeholders/collaborators within the EU HTA framework to assess their current level of involvement, determine their suggested future role, identify challenges to contribution, and highlight efficient ways to fulfilling their role. The ‘key’ stakeholder groups identified and covered by this research included: patients‘, clinicians‘, regulatory, and Health Technology Developer representatives. The survey was circulated to a wide expert audience including all relevant stakeholder groups in order to determine self-perception by the ‘key’ stakeholders regarding involvement in the HTA process (self-rating), and in a second, slightly modified version of the questionnaire, to determine the perception of ‘key’ stakeholder involvement by HTA bodies, payers, and policymakers (external rating). Predefined analyses were conducted on the submitted responses. Fifty-four responses were received (patients 9; clinicians: 8; regulators: 4; HTDs 14; HTA bodies: 7; Payers: 5; policymakers 3; others 4). The mean self-perceived involvement score was consistently lower for each of the ‘key’ stakeholder groups than the respective external ratings. Based on the qualitative insights generated in the survey, a RACI Chart (Responsible/Accountable/Consulted/Informed) was developed for each of the stakeholder groups to determine their roles and involvement in the current EU HTA process. Our findings suggest extensive effort and a distinct research agenda are required to ensure adequate involvement of the key stakeholder groups in the evolving EU HTA process.</p

The Maintenance of Traditions in Marmosets: Individual Habit, Not Social Conformity? A Field Experiment

Social conformity is a cornerstone of human culture because it accelerates and maintains the spread of behaviour within a group. Few empirical studies have investigated the role of social conformity in the maintenance of traditions despite an increasing body of literature on the formation of behavioural patterns in non-human animals. The current report presents a field experiment with free-ranging marmosets (Callithrix jacchus) which investigated whether social conformity is necessary for the maintenance of behavioural patterns within groups or whether individual effects such as habit formation would suffice.Using a two-action apparatus, we established alternative behavioural patterns in six family groups composed of 36 individuals. These groups experienced only one technique during a training phase and were thereafter tested with two techniques available. The monkeys reliably maintained the trained method over a period of three weeks, despite discovering the alternative technique. Three additional groups were given the same number of sessions, but those 21 individuals could freely choose the method to obtain a reward. In these control groups, an overall bias towards one of the two methods was observed, but animals with a different preference did not adjust towards the group norm. Thirteen of the fifteen animals that discovered both techniques remained with the action with which they were initially successful, independent of the group preference and the type of action (Binomial test: exp. proportion: 0.5, p<0.01).The results indicate that the maintenance of behavioural patterns within groups 1) could be explained by the first rewarded manipulation and subsequent habit formation and 2) do not require social conformity as a mechanism. After an initial spread of a behaviour throughout a group, this mechanism may lead to a superficial appearance of conformity without the involvement of such a socially and cognitively complex mechanism. This is the first time that such an experiment has been conducted with free-ranging primates

    Hybrid Cultures in Medieval Europe

Color poster with text and graphs.Bachiochi et al. (2011) developed an article analysis activity to assess psychology students' mastery of American Psychological Association student learning outcomes (SLOs) for research methods (Goal 2; APA 2007). Students read a research report and answered factual and analysis questions about it. Bachiochi et al. used the activity to assess their department's success at teaching research methods over a two-course sequence, finding that it both documented students' learning and identified ways in which they could improve their program. This study applied the Baciochi et al. approach to assess student attainment of the UWEC Psychology Department's SLOs for research methods.University of Wisconsin--Eau Claire Office of Research and Sponsored Programs
