Peer reviewe

AbstractThe joint European research project ELVA brings in line technology options and customer expectations for third generation electric vehicles. The objective is to develop innovative vehicle architectures fully exploiting freedoms in design that result from the electric drivetrain. This paper summarises the results of the first phase of the project which comprised deep technology and market forecasting as well as an open design contest for future electric vehicles. In the end, an outlook is given for the next phase in which three detailed vehicle concepts will be virtually developed and assessed with regard to several key requirements such as energy efficiency, material application and safety

Severe Early-Onset Obesity Due to Bioinactive Leptin Caused by a p.N103K Mutation in the Leptin Gene.

CONTEXT: Congenital leptin deficiency is a very rare cause of severe early-onset obesity. We recently characterized a mutation in the leptin gene (p.D100Y), which was associated with detectable leptin levels and bioinactivity of the hormone. CASE DESCRIPTION: We now describe two siblings, a 9-year-old girl and a 6-year-old boy with severe early-onset obesity and hyperphagia, both homozygous for a c.309C>A substitution in the leptin gene leading to a p.N103K amino acid exchange in the protein and detectable circulating levels of leptin. In vitro experiments in a heterologous cell system demonstrated that the mutated protein was biologically inactive. Treatment with sc recombinant human leptin led to rapid improvement of eating behavior and weight loss. CONCLUSIONS: Sequencing of the leptin gene may need to be considered in hyperphagic, severely obese children with detectable levels of circulating leptin. This work was supported by Grant BMBF 01GI1120A from the Federal Ministry of Education and Research. CD90 is dispensable for white and beige/brown adipocyte differentiation

Brown adipose tissue (BAT) is a thermogenic organ in rodents and humans. In mice, the transplantation of BAT has been successfully used to combat obesity and its comorbidities. While such beneficial properties of BAT are now evident, the developmental and cellular origins of brown, beige, and white adipocytes have remained only poorly understood, especially in humans. We recently discovered that CD90 is highly expressed in stromal cells isolated from human white adipose tissue (WAT) compared to BAT. Here, we studied whether CD90 interferes with brown or white adipogenesis or white adipocyte beiging. We applied flow cytometric sorting of human adipose tissue stromal cells (ASCs), a CRISPR/Cas9 knockout strategy in the human Simpson-Golabi-Behmel syndrome (SGBS) adipocyte model system, as well as a siRNA approach in human approaches supports the hypothesis that CD90 affects brown or white adipogenesis or white adipocyte beiging in humans. Taken together, our findings call the conclusions drawn from previous studies, which claimed a central role of CD90 in adipocyte differentiation, into question

The HERAB target is operated to produce protonnucleon interactions at the HERA proton storage ring. Therefore eight wire targets are arranged around the HERA proton beam. The distance of these wire targets to the center of the proton beam is continously steered in such a way that the interaction rate remains as constant as possible and that each of the targets contributes equally to the total rate. For the reconstruction of ebvents in the HERAB detector it is important that no interactions arise in the time slots between the proton bunches rotating in HERA and that there are no large fluctuations of the interaction rate. This work describes the mechanics of the HERAB target system, which is essential for the precise steering of the interaction rate, and summarizes the accomplished improvements of a newly constructed mechanics in comparison to the previously used target mechanics. While operating the target system not desired interactions in the time slots between the proton bunches of the beam were observed at the target. These coasting beam interactions are examined in detail in this work. The examinati ons show that the observed coasting beam consists of several components, which can be classified by their energy deviation relative to the nominal energy and their lifetime. Possible production mechanisms are discussed for the individual components. Furthermore the principle of modulating the storage ring tune was examined for the reduction of fluctuations of the interaction rate. With the help of a single particle simulation the principle of the tune modulation is described in detail. It is shown that the modulation increases the diffusion of the protons in the working area of the target. The increased diffusion leads to smaller fluctuations of the interaction rate, which is shown using measurements of the described target system
