Untersuchung der immunstimulatorischen Eigenschaften des Helicobacter-cysteine-rich protein A (HcpA)
Helicobacter pylori (Hp) ist ein gramnegatives Bakterium, das den menschlichen Magen kolonisiert und dabei obligatorisch eine chronische Gastritis induziert. Trotz dieser Immunreaktion persistiert das Bakterium ohne antibiotische Behandlung im Normalfall lebenslang. Diese in der Regel asymptomatische Gastritis kann in Abhängigkeit von Wirtsfaktoren und Ernährungsgewohnheiten verschiedene klinische Manifestationen erreichen. Die Infektion mit Hp kann chronische aktive oder atrophische Gastritiden, peptidische Magengeschwüre und MALTome auslösen. Eine der Grundlagen der Entzündungsreaktion ist das sich im Magen ausbildende Zytokinmilieu, für dessen Polarisierung Interferon-gamma (IFN-gamma) als Leitzytokin der von T-Helfer-Zellen des Typs I (TH1) geprägten Immunantwort von großer Bedeutung ist.
Helicobacter-cysteine-rich-protein A (HcpA) ist ein von Hp aktiv sezerniertes Protein, das als neuer, immunstimulatorisch wirkender Virulenzfaktor beschrieben wurde. HcpA gehört einer nur in Campylobacterales vorkommenden Familie paraloger Proteine an. Die Stimulation von Splenozyten-Einzelzellsuspensionen aus naiven Mäusen mit in E. coli rekombinant exprimiertem HcpA konnte eine der Ko-Kultur mit lebendem Hp vergleichbare Zytokinfreisetzung, insbesondere des entzündungsfördernd wirkenden IFN-gamma, induzieren. Neben dieser direkten Aktivierung des angeborenen Immunsystems konnten durch serologische Untersuchungen von Hp infizierten Patienten hohe anti-HcpA Antikörpertiter nachgewiesen werden.
Zusammenfassend wurde in dieser Arbeit die Expression und Sekretion von HcpA unter physiologischen Bedingungen erstmals untersucht und beschrieben. Die erfolgreiche Etablierung einer stabil transfizierten Drosophila-Schneider-2 Zelllinie (DS-2) erlaubte die Expression großer Mengen von LPS-freiem rekombinantem HcpA, das im wesentlichen die gleichen immunstimulatorischen Eigenschaften wie in E. coli rekombinant exprimiertes HcpA aufwies. Die Aufklärung der durch HcpA induzierten Kaskade, die zur Aktivierung des angeborenen Immunsystems im Mausmodell als auch in humanen PBMC führt, konnte die Bedeutung von HcpA als immunologischem Virulenzfaktor weiter untermauern. Die Identifizierung von B- und T-Zellepitopen ist eine wichtige Vorraussetzung für sich jetzt anschließende Untersuchungen zur Entwicklung einer HcpA-basierten, protektiven Vakzinierungsstrategie. Der Befund, dass HcpA über den TLR4 seine Aktivierung des angeborenen Immunsystems direkt oder indirekt vermittelt, stellt ein weiteres zukunftsweisendes Ergebnis dieser Arbeit dar. Der Nachweis einer starken Immunaktivierung durch HcpA in humanen PBMCs zeigt erstmalig, dass die bisher nur im Maus-System beschriebenen immunmodulierenden Eigenschaften von HcpA voll auf den Menschen übertragbar zu sein scheinen. Insgesamt konnte die Arbeit damit einen wesentlichen Beitrag zum Verständnis eines neuen immunologisch wirkenden Virulenzfaktors von Hp leisten

Polymeric Nanoparticles Based on Tyrosine-Modified, Low Molecular Weight Polyethylenimines for siRNA Delivery
A major hurdle for exploring RNA interference (RNAi) in a therapeutic setting is still
the issue of in vivo delivery of small RNA molecules (siRNAs). The chemical modification of
polyethylenimines (PEIs) offers a particularly attractive avenue towards the development of more
efficient non-viral delivery systems. Here, we explore tyrosine-modified polyethylenimines with low
or very low molecular weight (P2Y, P5Y, P10Y) for siRNA delivery. In comparison to their respective
parent PEI, they reveal considerably increased knockdown efficacies and very low cytotoxicity upon
tyrosine modification, as determined in different reporter and wildtype cell lines. The delivery
of siRNAs targeting the anti-apoptotic oncogene survivin or the serine/threonine-protein kinase
PLK1 (polo-like kinase 1; PLK-1) oncogene reveals strong inhibitory effects in vitro. In a therapeutic
in vivo setting, profound anti-tumor effects in a prostate carcinoma xenograft mouse model are
observed upon systemic application of complexes for survivin or PLK1 knockdown, in the absence of
in vivo toxicity. We thus demonstrate the tyrosine-modification of (very) low molecular weight PEIs
for generating effcient nanocarriers for siRNA delivery in vitro and in vivo, present data on their
physicochemical and biological properties, and show their efficacy as siRNA therapeutic in vivo, in
the absence of adverse effects
Tyrosine-Modification of Polypropylenimine (PPI) and Polyethylenimine (PEI) Strongly Improves Efficacy of siRNA-Mediated Gene Knockdown
The delivery of small interfering RNAs (siRNA) is an efficient method for gene silencing through the induction of RNA interference (RNAi). It critically relies, however, on efficient vehicles for siRNA formulation, for transfection in vitro as well as for their potential use in vivo. While polyethylenimines (PEIs) are among the most studied cationic polymers for nucleic acid delivery including small RNA molecules, polypropylenimines (PPIs) have been explored to a lesser extent. Previous studies have shown the benefit of the modification of small PEIs by tyrosine grafting which are featured in this paper. Additionally, we have now extended this approach towards PPIs, presenting tyrosine-modified PPIs (named PPI-Y) for the first time. In this study, we describe the marked improvement of PPI upon its tyrosine modification, leading to enhanced siRNA complexation, complex stability, siRNA delivery, knockdown efficacy and biocompatibility. Results of PPI-Y/siRNA complexes are also compared with data based on tyrosine-modified linear or branched PEIs (LPxY or PxY). Taken together, this establishes tyrosine-modified PPIs or PEIs as particularly promising polymeric systems for siRNA formulation and delivery
A Framework for Data-Driven Explainability in Mathematical Optimization
Advancements in mathematical programming have made it possible to efficiently
tackle large-scale real-world problems that were deemed intractable just a few
decades ago. However, provably optimal solutions may not be accepted due to the
perception of optimization software as a black box. Although well understood by
scientists, this lacks easy accessibility for practitioners. Hence, we advocate
for introducing the explainability of a solution as another evaluation
criterion, next to its objective value, which enables us to find trade-off
solutions between these two criteria. Explainability is attained by comparing
against (not necessarily optimal) solutions that were implemented in similar
situations in the past. Thus, solutions are preferred that exhibit similar
features. Although we prove that already in simple cases the explainable model
is NP-hard, we characterize relevant polynomially solvable cases such as the
explainable shortest-path problem. Our numerical experiments on both artificial
as well as real-world road networks show the resulting Pareto front. It turns
out that the cost of enforcing explainability can be very small
MUDIL – Musikalisches Distance Learning: Erfahrungen, Auswirkungen, Perspektiven:Ein Forschungsbericht zu ausgewählten Ergebnissen einer Online-Befragung zum Musikunterricht während des ersten Corona-Lockdowns im Frühjahr 2020
Die kooperative Studie MUDIL beforscht die Arbeitsfelder von Musiklehrenden an Schulen und Musikschulen unter den Bedingungen von Distance Learning während der Covid-19-Krise in Österreich. Der vorliegende Forschungsbericht stellt ausgewählte Ergebnisse einer österreichweiten, quantitativen online-Befragung im Sommer 2020 dar, in der Rückmeldungen von 1158 Lehrenden aus Musikschulen und Regelschulen der Sekundarstufe in ganz Österreich und Südtirol ausgewertet werden konnten. Inhalt sind neben einer sozio-demografischen Einordnung der Befragungsergebnisse die technischen und rechtlichen Rahmenbedingungen, die pädagogischen Entwicklungen in Unterricht und Lernen sowie damit verbundene Probleme und Chancen, sowie Ausblicke auf zukünftige Entwicklungen mit Einschätzungen von Potentialen und Risiken

MicroRNAs are important cellular mediators of mRNA degradation and translation repression, which in turn can have an impact on various processes and, if their function is perturbed, can cause disease. Rep. 37, BSR20160578], which explores microRNA-146a and how it may play an indirect yet vital role in the proliferation of osteoarthritis (OA) chondrocytes.
