    Appalachia is a region of contrasts. Rich history, abundant natural beauty, and hard-working, good-hearted people co-exist alongside enduring poverty and unemployment, polluted abandoned mine lands, chronic poor health, and widespread opioid abuse. Appalachia is historically under-resourced and underinvested. A 2017 report showed granting foundations spent only 43.00perpersoninAppalachiaversusthenationalaverageof43.00 per person in Appalachia versus the national average of 451.00. This means the average Appalachian resident had access to less than 10% of the charitable giving funds available to most Americans; yet, the need for investment and charitable giving in Appalachia is greater now than ever due to higher rates of poverty and unemployment, struggles with educational achievement, and poor health outcomes.These current facts do not have to be the final story of Appalachia. Geoff Marietta, an Eastern Kentucky investor and founder of Invest 606, eastern Kentucky's business accelerator and pitch competition, said in a recent interview that "if you want to be successful in investing, you have to know a secret...you have to know something no one else knows, because if everyone else knows, it becomes price competitive." The secret Geoff knows is simple: Appalachia is one of the best places in America to invest. There is a movement across the region with local residents leading the charge to rewrite the story of Appalachia as people are currently investing in their communities like never before. After over a century of relying on coal, a single extractive industry, locals are spurring an entrepreneurship renaissance and making their hometowns places where they want to continue living and others want to start visiting.This playbook builds on the momentum of the Appalachian Investment Ecosystem Initiative, Invest Appalachia, Central Appalachian Network, Appalachian Funders Network, and countless other efforts to foster sustainable economic development across the region, and provides communities with tools and resources to stimulate downtown revitalization.

    Development of the MyCyberSkills™ iPad App: A Scenarios-Based, Hands-On Measure of Non-IT Professionals’ Cybersecurity Skills

    Although advances in Information Technology (IT) have been significant over the past several decades when it comes to protection of corporate information systems (IS), human errors and social engineering appear to prevail in circumventing such IT protections. While most employees may have the best of intentions, without cybersecurity skills they represent the weakest link in an organization’s IS security. Skills are defined as the combination of knowledge, experience, and ability to do something well. Cybersecurity skills correspond to the skills surrounding the hardware and software required to execute IS security to mitigate cyber-attacks. However, the current measures of end-user cybersecurity skills are based on self-reported surveys. This study is the second phase of a larger research project that is aimed to develop a scenario-based iPad application to measure cybersecurity skills based on actual scenarios with hands-on tasks that the participants complete in demonstrating their skills. To design a measure that has both high validity and reliability, subject matter experts’ (SMEs) opinion of the top nine cybersecurity skills and their skill importance weight were identified in the first phase of the study following the Delphi method. This phase of the research in progress involves the design and development of the MyCyberSkills™ iPad application (app) using scenario-based, hands-on tasks related to each of the nine SMEs identified cybersecurity skills

    Human Trafficking by the Numbers: The Initial Benchmark of Prevalence and Economic Impact for Texas

Traffickers exploit approximately 600 million from victims of labor trafficking in Texas.BackgroundThough human trafficking is widespread in geographically large states with large urban centers like Texas, the true scope of this hidden crime is largely unconfirmed as data on human trafficking are difficult to ascertain. Existing data gathered in anti-trafficking efforts focus almost exclusively on identified victims, shedding light on only a fraction of the problem. The first phase of the Statewide Human Trafficking Mapping Project of Texas focused on providing empiricallygrounded data as a benchmark about the extent of human trafficking across the state. The following three primary research questions guided our data collection efforts, which included queries of existing databases, interviews, focus groups, and web-based surveys.1.What is the prevalence of human trafficking in Texas?2.What is the economic impact of human trafficking in Texas?3.What is our understanding of human trafficking in Texas?MethodsThe findings in this report were derived using a multi-methods approach to quantify the prevalence and economic impact of human trafficking in Texas. Higher-than-average risk industry and community segments were chosen for sex and labor markets. We defined community segments asgroups of people considered to be at higher-than-average risk of trafficking because of risk indicators found in trafficking cases (e.g. homelessness). More specifically, rather than attempting to establish prevalence of trafficking among the 27.4 million people living in Texas, for the purposes of demonstrating our methodology, establishing some benchmarks on human trafficking prevalence and economic impact estimates, and providing a concrete example of our planned activities moving forward, victimization rates were applied to a select few community segments that are at higher-than-average risk of trafficking.The methodology has addressed the critical industry and community segments to accurately estimate prevalencewhile reducing overlap between the chosen segments

    Using Physiologically-Based Pharmacokinetic Models to Incorporate Chemical and Non-Chemical Stressors into Cumulative Risk Assessment: A Case Study of Pesticide Exposures

    Cumulative risk assessment has been proposed as an approach to evaluate the health risks associated with simultaneous exposure to multiple chemical and non-chemical stressors. Physiologically based pharmacokinetic/pharmacodynamic (PBPK/PD) models can allow for the inclusion and evaluation of multiple stressors, including non-chemical stressors, but studies have not leveraged PBPK/PD models to jointly consider these disparate exposures in a cumulative risk context. In this study, we focused on exposures to organophosphate (OP) pesticides for children in urban low-income environments, where these children would be simultaneously exposed to other pesticides (including pyrethroids) and non-chemical stressors that may modify the effects of these exposures (including diet). We developed a methodological framework to evaluate chemical and non-chemical stressor impacts on OPs, utilizing an existing PBPK/PD model for chlorpyrifos. We evaluated population-specific stressors that would influence OP doses or acetylcholinesterase (AChE) inhibition, the relevant PD outcome. We incorporated the impact of simultaneous exposure to pyrethroids and dietary factors on OP dose through the compartments of metabolism and PD outcome within the PBPK model, and simulated combinations of stressors across multiple exposure ranges and potential body weights. Our analyses demonstrated that both chemical and non-chemical stressors can influence the health implications of OP exposures, with up to 5-fold variability in AChE inhibition across combinations of stressor values for a given OP dose. We demonstrate an approach for modeling OP risks in the presence of other population-specific environmental stressors, providing insight about co-exposures and variability factors that most impact OP health risks and contribute to children’s cumulative health risk from pesticides. More generally, this framework can be used to inform cumulative risk assessment for any compound impacted by chemical and non-chemical stressors through metabolism or PD outcomes

    A Pilot Study of the Effects of Mycoplasma ovipneumoniae Exposure on Domestic Lamb Growth and Performance

    Mycoplasma ovipneumoniae is a globally distributed pathogen that has been associated with pneumonia in both domestic and wild Caprinae. It is closely related to M. hyopneumoniae, a respiratory pathogen of swine that is associated with decreased growth rates of pigs as well as clinical respiratory disease. In order to assess the effects of M. ovipneumoniae on lamb performance, we generated a cohort of lambs free of M. ovipneumoniae by segregation of test negative ewes after lambing, then compared the growth and carcass quality traits of M. ovipneumoniae-free and -colonized lambs from weaning to harvest. Some signs of respiratory disease were observed during the feeding trial in both lamb groups, but the M. ovipneumoniae-exposed group included more affected lambs and higher average disease scores. At harvest, lungs of lambs in both groups showed few grossly visible lesions, although the M. ovipneumoniae-exposed group did exhibit increased microscopic lung lesions (P\u3c0.05). In addition, M. ovipneumoniae exposed lambs produced lower average daily gains (P\u3c0.05), and lower yield grade carcasses (P\u3c0.05) compared to those of non-exposed lambs. The results demonstrated the feasibility of test and segregation for elimination of M. ovipneumoniae from groups of sheep and suggested that this pathogen may impair lamb growth and productivity even in the absence of overt respiratory disease

    Haptoglobin genotype predicts development of coronary artery calcification in a prospective cohort of patients with type 1 diabetes

    <p>Abstract</p> <p>Background</p> <p>Coronary artery disease has been linked with genotypes for haptoglobin (Hp) which modulates extracorpuscular hemoglobin. We hypothesized that the Hp genotype would predict progression of coronary artery calcification (CAC), a marker of subclinical atherosclerosis.</p> <p>Methods</p> <p>CAC was measured three times in six years among 436 subjects with type 1 diabetes and 526 control subjects participating in the Coronary Artery Calcification in Type 1 Diabetes (CACTI) study. Hp typing was performed on plasma samples by polyacrylamide gel electrophoresis.</p> <p>Results</p> <p>The Hp 2-2 genotype predicted development of significant CAC only in subjects with diabetes who were free of CAC at baseline (OR: 1.95, 95% CI: 1.07-3.56, p = 0.03), compared to those without the Hp 2-2 genotype, controlling for age, sex, blood pressure and HDL-cholesterol. Hp 2 appeared to have an allele-dose effect on development of CAC. Hp genotype did not predict CAC progression in individuals without diabetes.</p> <p>Conclusions</p> <p>Hp genotype may aid prediction of accelerated coronary atherosclerosis in subjects with type 1 diabetes.</p

    Comparison of greenhouse gas fluxes from tropical forests and oil palm plantations on mineral soil

    In Southeast Asia, oil palm (OP) plantations have largely replaced tropical forests. The impact of this shift in land use on greenhouse gas (GHG) fluxes remains highly uncertain, mainly due to a relatively small pool of available data. The aim of this study is to quantify differences of nitrous oxide (N2O) and methane (CH4) fluxes as well as soil carbon dioxide (CO2) respiration rates from logged forests, oil palm plantations of different ages, and an adjacent small riparian area. Nitrous oxide fluxes are the focus of this study, as these emissions are expected to increase significantly due to the nitrogen (N) fertilizer application in the plantations. This study was conducted in the SAFE (Stability of Altered Forest Ecosystems) landscape in Malaysian Borneo (Sabah) with measurements every 2 months over a 2-year period. GHG fluxes were measured by static chambers together with key soil physicochemical parameters and microbial biodiversity. At all sites, N2O fluxes were spatially and temporally highly variable. On average the largest fluxes (incl. 95 % CI) were measured from OP plantations (45.1 (24.0–78.5) µg m−2 h−1 N2O-N), slightly smaller fluxes from the riparian area (29.4 (2.8–84.7) µg m−2 h−1 N2O-N), and the smallest fluxes from logged forests (16.0 (4.0–36.3) µg m−2 h−1 N2O-N). Methane fluxes were generally small (mean ± SD): −2.6 ± 17.2 µg CH4-C m−2 h−1 for OP and 1.3 ± 12.6 µg CH4-C m−2 h−1 for riparian, with the range of measured CH4 fluxes being largest in logged forests (2.2 ± 48.3 µg CH4-C m−2 h−1). Soil respiration rates were larger from riparian areas (157.7 ± 106 mg m−2 h−1 CO2-C) and logged forests (137.4 ± 95 mg m−2 h−1 CO2-C) than OP plantations (93.3 ± 70 mg m−2 h−1 CO2-C) as a result of larger amounts of decomposing leaf litter. Microbial communities were distinctly different between the different land-use types and sites. Bacterial communities were linked to soil pH, and fungal and eukaryotic communities were linked to land use. Despite measuring a large number of environmental parameters, mixed models could only explain up to 17 % of the variance of measured fluxes for N2O, 3 % of CH4, and 25 % of soil respiration. Scaling up measured N2O fluxes to Sabah using land areas for forest and OP resulted in emissions increasing from 7.6 Mt (95 % confidence interval, −3.0–22.3 Mt) yr−1 in 1973 to 11.4 Mt (0.2–28.6 Mt) yr−1 in 2015 due to the increasing area of forest converted to OP plantations over the last ∼ 40 years
