    Architectural Urbanism: Melbourne/Seoul

    Architectural Urbanism is an ambition and sensibility for propositions that address the context of the city within the operative scale of the small architectural project. Architectural urbanism represents a tailoring of projects to the local; to the materiality and specificity of the everyday; and to the grain and substance of the location above all else. Architectural urbanism is less about erasure and more about insertion; infill; the weaving of old and new and the dynamics that evolve from subtle and careful manipulation of the city in detail. The exhibition explores commonality in the apparently different contexts of both cities ā€“ speculating on these as forms of ā€˜architectural urbanism' in the contemporary city of the Asia-Pacific at its northern and southern extremes. Architectural projects from five Melbourne architectural practitioners have been selected to exhibit in Seoul. The practices are: Muir Mendes, Baracco + Wright, Iredale Pederson Hook, NMBW Architecture Studio and Kerstin Thompson Architects - all of whom have strong links to the RMIT Architecture design research and teaching community.     Whole genome sequencing-based mapping and candidate identification of mutations from fixed zebrafish tissue

    As forward genetic screens in zebrafish become more common, the number of mutants that cannot be identified by gross morphology or through transgenic approaches, such as many nervous system defects, has also increased. Screening for these difficult-to-visualize phenotypes demands techniques such as whole-mount in situ hybridization (WISH) or antibody staining, which require tissue fixation. To date, fixed tissue has not been amenable for generating libraries for whole genome sequencing (WGS). Here, we describe a method for using genomic DNA from fixed tissue and a bioinformatics suite for WGS-based mapping of zebrafish mutants. We tested our protocol using two known zebrafish mutant alleles, gpr126st49 and egr2bfh227, both of which cause myelin defects. As further proof of concept we mapped a novel mutation, stl64, identified in a zebrafish WISH screen for myelination defects. We linked stl64 to chromosome 1 and identified a candidate nonsense mutation in the F-box and WD repeat domain containing 7 (fbxw7) gene. Importantly, stl64 mutants phenocopy previously described fbxw7vu56 mutants, and knockdown of fbxw7 in wild-type animals produced similar defects, demonstrating that stl64 disrupts fbxw7. Together, these data show that our mapping protocol can map and identify causative lesions in mutant screens that require tissue fixation for phenotypic analysis

    See Me Smoke-Free: Protocol for a Research Study to Develop and Test the Feasibility of an mHealth App for Women to Address Smoking, Diet, and Physical Activity

    Background: This paper presents the protocol for an ongoing research study to develop and test the feasibility of a multi-behavioral mHealth app. Approximately 27 million women smoke in the US, and more than 180,000 women die of illnesses linked to smoking annually. Women report greater difficulties quitting smoking. Concerns about weight gain, negative body image, and low self-efficacy may be key factors affecting smoking cessation among women. Recent studies suggest that a multi-behavioral approach, including diet and physical activity, may be more effective at helping women quit. Guided imagery has been successfully used to address body image concerns and self-efficacy in our 3 target behaviorsā€”exercise, diet and smoking cessation. However, it has not been used simultaneously for smoking, diet, and exercise behavior in a single intervention. While imagery is an effective therapeutic tool for behavior change, the mode of delivery has generally been in person, which limits reach. mHealth apps delivered via smart phones offer a unique channel through which to distribute imagery-based interventions. Objective: The objective of our study is to evaluate the feasibility of an mHealth app for women designed to simultaneously address smoking, diet, and physical activity behaviors. The objectives are supported by three specific aims: (1) develop guided imagery content, user interface, and resources to reduce weight concern, and increase body image and self-efficacy for behavior change among women smokers, (2) program a prototype of the app that contains all the necessary elements of text, graphics, multimedia and interactive features, and (3) evaluate the feasibility, acceptability, and preliminary efficacy of the app with women smokers. Methods: We created the program content and designed the prototype application for use on the Android platform in collaboration with 9 participants in multiple focus groups and in-depth interviews. We programmed and tested the applicationā€™s usability with 6 participants in preparation for an open, pre- and posttest trial. Currently, we are testing the feasibility and acceptability of the application, evaluating the relationship of program use to tobacco cessation, dietary behaviors, and physical activity, and assessing consumer satisfaction with approximately 70 women smokers with Android-based smart phones. Results: The study was started January 1, 2014. The app was launched and feasibility testing began in April 1, 2015. Participants were enrolled from April 1-June 30, 2015. During that time, the app was downloaded over 350 times using no paid advertising. Participants were required to use the app ā€œmost daysā€ for 30 days or they would be dropped from the study. We enrolled 151 participants. Of those, 78 were dropped or withdrew from the study, leaving 73 participants. We have completed the 30-day assessment, with a 92% response rate. The 90-day assessment is ongoing. During the final phase of the study, we will be conducting data analyses and disseminating study findings via presentations and publications. Feasibility will be demonstrated by successful participant retention and a high level of app use. We will examine individual metrics (eg, duration of use, number of screens viewed, change in usage patterns over time) and engagement with interactive activities (eg, activity tracking). Conclusions: We will aggregate these data into composite exposure scores that combine number of visits and overall duration to calculate correlations between outcome and measures of program exposure and engagement. Finally, we will compare app use between participants and non-participants using Google Analytics

We focus on the application of frameworks from social psychology to education, one of the 5 Esā€”engineering, education, enforcement, encouragement, and evaluation. We use the structure of the 5 Es to organize information with particular attention to engineering and education in the literature review. Engineering is essential because the infrastructure is vital to protecting cyclists. Education is emphasized since the central focus of the report is safety

    Isotopic temperatures from the early and mid-pliocene of the US Middle Atlantic coastal plain, and their implications for the cause of regional marine climate change

    Mean seasonal extreme temperatures on the seafloor calculated from the shell Ī“18O of the scallop Placopecten clintonius from the basal part of the early Pliocene Sunken Meadow Member (Yorktown Formation) in Virginia are very similar to those from the same horizon at the latitude of Cape Hatteras in North Carolina (~210 km to the south). The lowest and highest temperatures calculated from each shell (using Ī“18Oseawater = +0.7ā€°) give mean values for winter and summer of 8.4 Ā± 1.1 Ā°C (Ā± 1Ļƒ) and 18.2 Ā± 0.6 Ā°C in Virginia, and 8.6 Ā± 0.4 Ā°C and 16.5 Ā± 1.1 Ā°C in North Carolina (respective median temperatures: 13.3 Ā°C and 12.6 Ā°C). Patterns of ontogenetic variation in Ī“18O, Ī“13C and microgrowth increment size indicate summer water-column stratification in both areas, with summer surface temperatures perhaps 6 Ā°C higher than on the seafloor. The low winter paleotemperatures in both areas are most simply explained by the greater southward penetration of cool northern waters in the absence of a feature equivalent to Cape Hatteras. The same current configuration but a warmer general climate can account for the high benthic seasonal range (over 15.0 Ā°C in some cases) but warmer median temperatures (15.7-21.3 Ā°C) derived from existing Ī“18O data from scallops of the higher Yorktown Formation (using Ī“18Oseawater = +0.7ā€° for the upper Sunken Meadow Member and Ī“18Oseawater = +1.1ā€° for the mid-Pliocene Rushmere, Morgarts Beach and Moore House members). Existing Ī“18O data from the infaunal bivalve Mercenaria of the Rushmere Member yields a similarly high median temperature (21.6 Ā°C) but a low seasonal range (9.2 Ā°C), pointing to the periodic influence of warm currents, possibly at times when the Gulf Stream was exceptionally vigorous.British Geological Survey (BUFI S157 Studentship); UoD Research-Inspired Curriculum Fun

    An Assessment of the Model of Concentration Addition for Predicting the Estrogenic Activity of Chemical Mixtures in Wastewater Treatment Works Effluents

    The effects of simple mixtures of chemicals, with similar mechanisms of action, can be predicted using the concentration addition model (CA). The ability of this model to predict the estrogenic effects of more complex mixtures such as effluent discharges, however, has yet to be established. Effluents from 43 U.K. wastewater treatment works were analyzed for the presence of the principal estrogenic chemical contaminants, estradiol, estrone, ethinylestradiol, and nonylphenol. The measured concentrations were used to predict the estrogenic activity of each effluent, employing the model of CA, based on the relative potencies of the individual chemicals in an in vitro recombinant yeast estrogen screen (rYES) and a short-term (14-day) in vivo rainbow trout vitellogenin induction assay. Based on the measured concentrations of the four chemicals in the effluents and their relative potencies in each assay, the calculated in vitro and in vivo responses compared well and ranged between 3.5 and 87 ng/L of estradiol equivalents (E2 EQ) for the different effluents. In the rYES, however, the measured E2 EQ concentrations in the effluents ranged between 0.65 and 43 ng E2 EQ/L, and they varied against those predicted by the CA model. Deviations in the estimation of the estrogenic potency of the effluents by the CA model, compared with the measured responses in the rYES, are likely to have resulted from inaccuracies associated with the measurement of the chemicals in the extracts derived from the complex effluents. Such deviations could also result as a consequence of interactions between chemicals present in the extracts that disrupted the activation of the estrogen response elements in the rYES. E2 EQ concentrations derived from the vitellogenic response in fathead minnows exposed to a series of effluent dilutions were highly comparable with the E2 EQ concentrations derived from assessments of the estrogenic potency of these dilutions in the rYES. Together these data support the use of bioassays for determining the estrogenic potency of WwTW effluents, and they highlight the associated problems for modeling approaches that are reliant on measured concentrations of estrogenic chemicals

    Key considerations when involving children in health intervention design: reflections on working in partnership with South Asian children in the UK on a tailored Management and Intervention for Asthma (MIA) study

    Participatory research is an empowering process through which individuals can increase control over their lives, and allows researchers/clinicians to gain a clearer understanding of a childā€™s needs. However, involving children in participatory research is still relatively novel, despite national and international mandates to engage children in decision making. This paper draws on the learnings from designing the Management and Intervention for Asthma (MIA) study, which used a collaborative participatory method to develop an intervention-planning framework for South Asian children with asthma. There are currently 1 million children in the UK receiving treatment for asthma, making it one of the most prevalent chronic childhood illnesses. Symptoms of asthma are often underrecognized in children from South Asian communities in the UK, contributing to increased disease severity and increased attendance at the emergency department compared to White British children. Despite this, ethnic minorities are often excluded from research and thus absent from the ā€˜evidence baseā€™, making it essential to hear their perspectives if health inequalities are to be successfully addressed. We worked alongside healthcare professionals, community facilitators, parents, and children to identify the key concerns and priorities they had and then designed the framework around their needs. Reflecting on the process, we identified several key considerations that need to be addressed when co-developing interventions with children. These include the power dynamics between the parent/researcher and child; navigating the consent/assent process; how parental involvement might affect the research; establishing a convenient time and location; how to keep children engaged throughout the process; tailoring activities to different levels of ability; and accounting for cultural differences. These factors were considered by the researchers when designing the study, however, implementing them was not without its challenges and highlighted the need for researchers to develop expertise in this field. Tailoring existing research methods allowed us to explore childrenā€™s perceptions, priorities, and experiences of illness more effectively. However, involving children in participatory research is a complex undertaking, and researchers need to ensure that they have the expertise, time, and resources necessary to be able to fully support the needs of child participants before deciding to commit to this approach

    Artemin, a Novel Member of the GDNF Ligand Family, Supports Peripheral and Central Neurons and Signals through the GFRĪ±3ā€“RET Receptor Complex

    AbstractThe glial cell lineā€“derived neurotrophic factor (GDNF) ligands (GDNF, Neurturin [NTN], and Persephin [PSP]) signal through a multicomponent receptor system composed of a high-affinity binding component (GFRĪ±1ā€“GFRĪ±4) and a common signaling component (RET). Here, we report the identification of Artemin, a novel member of the GDNF family, and demonstrate that it is the ligand for the former orphan receptor GFRĪ±3ā€“RET. Artemin is a survival factor for sensory and sympathetic neurons in culture, and its expression pattern suggests that it also influences these neurons in vivo. Artemin can also activate the GFRĪ±1ā€“RET complex and supports the survival of dopaminergic midbrain neurons in culture, indicating that like GDNF (GFRĪ±1ā€“RET) and NTN (GFRĪ±2ā€“RET), Artemin has a preferred receptor (GFRĪ±3ā€“RET) but that alternative receptor interactions also occur

    Biotechnologies as catalysts for driving net zero

    R&D impact delivered by this work extends to policy development and to the benefits derived from delivering circularity, green growth and reducing carbon emissions by anaerobic digestion that (1) recovers a variety of organic wastes and low value biomass and (2) produces bioenergy and fertiliser. Other biotechnologies being developed can recover resources for the production of fuels (CH4, H2 and NH3), chemicals e.g. volatile fatty acids, biopolymers e.g. polyhydroxyalkanoates and single-cell proteins that can be used for animal feed. Biotechnologies delivering solutions for Power to X, for energy storage and for the capture and use of carbon have also been a focus of our research. Monitoring and control methodologies for the biotechnologies have been developed, including the use of analytical technologies such as FTNIR, GC-IMS and qPCR. Work continues on the valorisation of digestates as microbial and algae growth media, and the recovery of nutrients (NPK). Evaluations of the fate of polymers in the environment, their biochemical recycling and the production of biostimulants for soil and crop improvements, nitrogen fixing and emissionsā€™ reduction are all in progress. Technologies are currently across the TRL 3-6 range and require further R&D to progress them to commercialisation. Deploying industrial biotechnologies is essential to act as sustainable catalysts for change and for delivering net zero, circular economy and green growth. Biotechnologies can impact beneficially on the sustainability of cities and benefit their relationship and integration with surrounding rural areas
