    Socially defeated male rats display a blunted adrenocortical response to a low dose of 8-OH-DPAT

    The study examined in male Wistar rats the influence of social defeat on the neuroendocrine stress response system using injection of the 5-HT1A receptor agonist, 8-hydroxy-2-(di-n-propylamino)tetralin (8-OH-DPAT), as the pharmacological challenge. Social defeat was defined by the submissive postures displayed by the Wistar rats which were threatened and attacked by Tryon Maze Dull S3 rats for 10 min. 18-20 h after social defeat, the defeated rats were injected intravenously (i.v.) with a low and high dose of 8-OH-DPAT in their home cages. Blood samples were withdrawn from the freely moving cannulated rats for determination of plasma corticosterone and catecholamines. The corticosterone response to the low dose of 8-OH-DPAT (0.05 mg/kg, i.v.) was significantly diminished in the defeated rats as compared to the controls, but this dose failed to affect catecholamine concentrations. The high dose of 8-OH-DPAT (0.15 mg/kg, i.v.) significantly elevated corticosterone and adrenaline levels in defeated and control rats to the same extent, whereas no effect on noradrenaline was found. The present data thus indicate that social defeat blunts 5-HT1A receptor-mediated adrenocortical activation probably via a decrease in the sensitivity of a population of postsynaptic 5-HT receptors

    Ongunstige arbeidsomstandigheden veroorzaken 3,9% (onzekerheidsmarge 1,5%-7,2%) van de totale ziektelast in Nederland. De ziektelast is een maat om het verlies aan gezondheid uit te drukken. Het combineert vroegtijdige sterfte, de mate van vóórkomen van gezondheidsproblemen en de ernst van de gezondheidsproblemen. De ongunstige arbeidsomstandigheden die leiden tot de meeste ongezondheid zijn werkdruk, beeldschermwerk en blootstelling aan stoffen. Deze omstandigheden kunnen leiden tot burn-out, depressie, KANS (klachten van arm, nek en schouder), COPD (chronisch obstructieve longziekten) en longkanker. In het rapport is de positieve invloed van arbeid op de gezondheid niet meegenomen. Ook in 2020 veroorzaken burn-out, depressie en KANS veel ziektelast in de werkzame beroepsbevolking, bij ongewijzigde economische omstandigheden, een pensioengerechtigde leeftijd van 65 jaar en bij ongewijzigd (arbo)beleid. In 2007 heeft het RIVM voor het eerst laten zien welke arbeidsgerelateerde aandoeningen veel ziektelast in Nederland veroorzaken met gegevens uit 2003. Het huidige rapport biedt een hernieuwde versie met data uit 2007, evenals een toekomstverkenning en een verkenning van de ziektelast per sector. Deze schattingen geven beleidsmakers inzicht in de invloed van arbeidsrisico's op de gezondheid van werknemers. Deze benadering geeft ook aanknopingspunten voor maatregelen om de ziektelast door deze aandoeningen te verminderen.Occupational health risks cause 3.9% (uncertainty 1.5%-7.2%) of the total burden of disease in the Netherlands. The concept of burden of disease is a measure to express the loss of health. It combines the time lost due to premature mortality, prevalence and seriousness of the health problems. A high workload, working with a computer and exposure to harmful chemicals are the most unfavourable working conditions leading to health problems. They contribute most to the occupational burden of disease caused by: burn-out, depression, complaints of arm, neck and shoulder (CANS), chronic obstructive pulmonary disease and lung cancer. The health benefits of work were not included in this report. In 2020, burn-out, depression and CANS also cause a high burden of disease in the working population, considering unchanged economical conditions, a retirement age of 65 and unchanged health and safety policy. In 2007, the RIVM showed for the first time which occupational conditions contributed most to the burden of disease in 2003. The current report provides an update of the occupational burden of disease with data from 2007, as well as a forecast to 2020 and an exploration of the burden of disease per occupational sector. These estimates give policy makers insight in the influence of occupational risks on the health of employees. The data offer starting points for measures to reduce the burden of disease caused by these complaints.SZ

    Annual report on surveillance of respiratory infectious diseases 2013, the Netherlands

    Het griepseizoen (influenza) 2013/2014 was erg mild, na de uitzonderlijk langdurende epidemie in het seizoen 2012/2013. Ook was het een mild seizoen wat betreft het aantal mensen dat een longontsteking (pneumonie) opliep. In 2013 waren er geen grote uitbraken van de meldingsplichtige luchtweginfectieziekten legionellose (308 meldingen), papegaaienziekte (psittacose; 51 meldingen), Q-koorts (19 meldingen) en tuberculose (848 meldingen). Deze aantallen waren in het verslagjaar vergelijkbaar of lager dan het aantal meldingen in voorgaande jaren. Dit blijkt uit de jaarlijkse surveillance luchtweginfectieziekten 2013 van het RIVM. Griep en longontsteking leiden tot veel ziekenhuisopnames en sterfte in Nederland, waardoor het RIVM ze actief volgt. In vergelijking met griep komen de meldingsplichtige luchtweginfecties in Nederlands maar weinig voor. Ze zijn meldingsplichtig, omdat tijdige maatregelen, zoals de besmettingsbron opsporen, belangrijk kunnen zijn om uitbraken of verdere verspreiding van de ziekte te voorkomen. Het RIVM volgt ook potentieel gevaarlijke nieuwe luchtweginfecties die elders in de wereld voorkomen. In mei 2014 werden voor het eerst in Nederland twee patiënten gediagnostiseerd met het MERS coronavirus. In het seizoen 2013/2014 lag het aantal mensen dat met griepachtige klachten bij de huisarts kwam begin 2014 gedurende vier weken boven de grens waarmee een griepepidemie wordt geduid. Bij de patiënten met griepachtige klachten kwam naast influenzavirus vaak RSV (respiratoir syncytieel virus) en neusverkoudheid (rhinovirus) voor. Er kwamen minder mensen met een longontsteking bij de huisarts dan voorgaande seizoenen, maar het aantal longontstekingpatiënten in verpleeghuizen bleef gelijk.The 2013/2014 influenza season was extremely mild in the Netherlands, compared to the exceptionally long-lasting epidemic in 2012/2013. In addition, the number of pneumonia patients and overall mortality, possible complications of influenza, were low. In 2013, no major outbreaks of the notifiable respiratory infectious diseases legionellosis (308 notifications), psittacosis (51 notifications), Q-fever (19 notifications) and tuberculosis (848 notifications) occurred. These incidences are either comparable to or lower than preceding years. These are the outcomes of the annual report: 'Surveillance of respiratory infectious diseases 2013, the Netherlands', published by the Dutch National Institute for Public Health and the Environment (RIVM). Influenza and pneumonia are an important cause of hospital admissions and death in the Netherlands, a reason for the RIVM to actively monitor these diseases. In comparison to influenza, notifiable respiratory infectious diseases only rarely occur. These diseases are notifiable, as timely measures like source finding, are important for preventing outbreaks and/ or ongoing transmission of the disease. The RIVM also monitors potential threats to public health from new (worldwide) respiratory infections. In May 2014, the first two cases of MERS coronavirus were diagnosed in the Netherlands. During the 2013/2014 influenza-season, the number of patients with influenza-like illness (ILI) consulting a general practitioner, was above the threshold set for an influenza epidemic for four weeks at the beginning of 2014. In nose and throat samples of ILI-patients, RSV (respiratory syncytial virus) and rhinovirus were found in addition to the influenza virus. During the 2013/2014 influenza-season, fewer patients consulted the general practitioner for pneumonia than in previous years, however the number of pneumonia patients in nursing homes was similar.Ministerie van VW

    QT interval prolongation after sertraline overdose: a case report

    BACKGROUND: Selective serotonin reuptake inhibitors (SSRIs) are the most common antidepressants used in first-world countries and are generally well tolerated. Specifically, less cardiovascular toxicity has been reported in comparison with tricyclic antidepressants. Here we report QT interval prolongation after an overdose of the SSRI sertraline. CASE PRESENTATION: A previously healthy female patient presented with an attempted suicide with overdoses sertraline (2250 mg), diazepam (200 mg), and temazepam (400 mg). Routine laboratory studies were normal and her ECG upon admission showed a normal QT interval. The next day, her ECG showed prolongation of the QT(c )interval up to 525 ms. After discontinuation of sertraline the QT interval normalized. Echocardiography and exercise electrocardiography were normal. After hospitalization, the patient resumed sertraline in the normally recommended dose and QT interval remained within normal ranges. CONCLUSION: It seems that the SSRI sertraline in overdose may cause QT interval prolongation

In particular, methanofullereneshave been the preferred choice for solution-processed solar cells and photodiodes. The wide applicability of fullerenes as both ‘ntype’transport materials and electron acceptors is clear. However, what is still a matter of debate is whether the fullerenes can alsosupport efficient transport of holes, particularly in diode geometries.     Prevalence of Chlamydia trachomatis infection among women in a Middle Eastern community

    BACKGROUND: Common vaginal infections that manifest in women are usually easily diagnosed. However, Chlamydia infection is often asymptomatic, leading to infertility before it is detected. If it occurs in pregnancy, it could lead to significant neonatal morbidity. It may also play a role with other viral infections for e.g. Human Papilloma Virus in the development of cervical cancer. The objective of this study was to determine the prevalence of Chlamydia infection in women undergoing screening for cervical abnormalities as a part of a research project in primary and secondary care institutions in the United Arab Emirates. METHODS: In this cross sectional study married women attending primary and secondary care participating in a large nationwide cervical abnormalities screening survey were offered Chlamydia testing using a commercially available test kit. This kit uses a rapid immunoassay for the direct detection of Chlamydia trachomatis antigen in endocervical swab specimens. As this study was performed in a traditional Islamic country, unmarried women were excluded from testing, as the management of any positive cases would create legal and social problems. All married women consenting to take part in the study were included irrespective of age. RESULTS: Of 1039 women approached over a period of eight months 919 (88.5%) agreed to participate. The number of women in the 16 to 19 years was small (0.01%) and 30% were aged over 40 years. The prevalence of Chlamydia infection in this study was 2.6% (95% confidence interval 1.2–3.3%), which was marginally higher in women screened in secondary care (p = 0.05). CONCLUSION: This is one of the few reports on the prevalence of Chlamydia infection in women from the Middle East. Due to cultural and social constraints this study excluded a large proportion of women aged less than 19 years of age. Hence no direct comparisons on prevalence could be made with studies from the West, which all included younger women at high risk of Chlamydia. However this study emphasizes the importance of cultural factors while interpreting results of studies from different cultures and communities

    Single-Batch Production of Recombinant Human Polyclonal Antibodies

    We have previously described the development and implementation of a strategy for production of recombinant polyclonal antibodies (rpAb) in single batches employing CHO cells generated by site-specific integration, the SympressTM I technology. The SympressTM I technology is implemented at industrial scale, supporting a phase II clinical development program. Production of recombinant proteins by site-specific integration, which is based on incorporation of a single copy of the gene of interest, makes the SympressTM I technology best suited to support niche indications. To improve titers while maintaining a cost-efficient, highly reproducible single-batch manufacturing mode, we have evaluated a number of different approaches. The most successful results were obtained using random integration in a new producer cell termed ECHO, a CHO DG44 cell derivative engineered for improved productivity at Symphogen. This new expression process is termed the SympressTM II technology. Here we describe proof-of-principle data demonstrating the feasibility of the SympressTM II technology for single-batch rpAb manufacturing using two model systems each composed of six target-specific antibodies. The compositional stability and the batch-to-batch reproducibility of rpAb produced by the ECHO cells were at least as good as observed previously using site-specific integration technology.     Estimating Dynamic Gait Stability Using Data from Non-aligned Inertial Sensors

    Recently, two methods for quantifying the stability of a dynamical system have been applied to human locomotion: local stability (quantified by finite time maximum Lyapunov exponents, λs and λL) and orbital stability (quantified by maximum Floquet multipliers, MaxFm). In most studies published to date, data from optoelectronic measurement systems were used to calculate these measures. However, using wireless inertial sensors may be more practical as they are easier to use, also in ambulatory applications. While inertial sensors have been employed in some studies, it is unknown whether they lead to similar stability estimates as obtained with optoelectronic measurement systems. In the present study, we compared stability measures of human walking estimated from an optoelectronic measurement system with those calculated from an inertial sensor measurement system. Subjects walked on a treadmill at three different speeds while kinematics were recorded using both measurement systems. From the angular velocities and linear accelerations, λs, λL, and MaxFm were calculated. Both measurement systems showed the same effects of walking speed for all variables. Estimates from both measurement systems correlated high for λs and λL, (R > 0.85) but less strongly for MaxFm (R = 0.66). These results indicate that inertial sensors constitute a valid alternative for an optoelectronic measurement system when assessing dynamic stability in human locomotion, and may thus be used instead, which paves the way to studying gait stability during natural, everyday walking
