    Simultaneous determination of eight major bioactive compounds in Dachengqi Tang (DT) by high-performance liquid chromatography

    <p>Abstract</p> <p>Background</p> <p><it>Dachengqi Tang </it>(DT) is a common traditional Chinese medicine formula for expelling <it>neire </it>('internal heat') in the stomach and intestines. There was no reliable analytical method available for the quality control of DT.</p> <p>Methods</p> <p>A high-performance liquid chromatography (HPLC) method with a reverse phase C<sub>18 </sub>column (150 × 4.6 mm) was developed. The mobile phase was methanol with 0.2% acetic acid. Eight markers including naringin, hesperidin, aloe emodin, rhein, honokiol, magnolol, emodin and chrysophanol were determined.</p> <p>Results</p> <p>Regression analysis revealed a linear relationship between the concentrations of the markers and the peak area ratio of the standards and internal standard. The limit of detection (S/N = 3) and the limit of qualification (RSD < 20%) ranged from 0.21 to 0.43 ng/ÎŒl and 0.76 to 1.74 ng/ÎŒl respectively. The recovery was between 95.6% and 103.4%. The tests on the samples from three batches of DT showed that the profiles of the markers did not vary significantly among batches.</p> <p>Conclusion</p> <p>A reliable HPLC method for simultaneous determination of the eight markers in DT was developed.</p

    An Optimal Method For Product Selection By Using Online Ratings And Considering Search Costs

    With the collecting and publishing data about consumers purchasing and browsing products at the platform of online, this data prodives new ways to better understand the consumers search behavior before purchase. How to base on consumers online search behavior and simutaneously consider offline experience costs is worth studying. An optimal method based on the utility of the attribute of product is proposed. The proposed method follows steps below. Firstly, based on the multi-attribute utility theory, the overall utility of product is calculated by using ratings data. Secondly, the overall utility is combined into the original sequential search model to find the optimal selection strategy. Thirdly, the candidate product sets arranged in descending order of the reservation utilities are finally obtained. Finally, taking the online ratings data provided by a comprehensive automobile website as an example, lastly the proposed method is simulated and compared with other method. The result shows that the proposed method is feasible and effective

    Researching Dynamic Brand Competitiveness Based on Consumer Clicking Behavior

    Analyzing brand dynamic competition relationship by using consumer sequential online click data, which was collected from JD.com. It is found that the competition intensity of the products across categories is quite different. Owing to the purchasing time of durable-like goods is more flexible, that is, the purchasing probability of such products changes more obviously over time. Therefore, we use the Local Polynomial Regression Model to analyze the relationship between the brand competition of durable-like goods and the purchasing probability of the specific brand. Finding that when brands increase at a half of the total market share for consumers cognition preference, the brands’ competitiveness is peak and makes no significant different from one hundred percent for consumer to complete a transaction. The findings contribute to brand competitiveness for setting up marketing strategy from the dynamic and online consumer behavior’s perspective

    AoBck1 and AoMkk1 Are Necessary to Maintain Cell Wall Integrity, Vegetative Growth, Conidiation, Stress Resistance, and Pathogenicity in the Nematode-Trapping Fungus Arthrobotrys oligospora

    The cell wall integrity (CWI) pathway is composed of three mitogen-activated protein kinases (MAPKs), Bck1, Mkk1/2, and Slt2, and is one of the main signaling pathways for fungal pathogenesis, cell wall synthesis, and integrity maintenance. In this study, we characterized orthologs of Saccharomyces cerevisiae Bck1 and Mkk1 in the nematode-trapping (NT) fungus Arthrobotrys oligospora by multiple phenotypic comparison, and the regulation of conidiation and cell wall synthesis was analyzed using real-time PCR (RT-PCR). Both ΔAoBck1 and ΔAoMkk1 mutants showed severe defects in vegetative growth, cell nucleus number, and stress resistance. Both the mutants were unable to produce spores, and the transcription of several genes associated with sporulation and cell wall biosynthesis was markedly downregulated during the conidiation stage. Further, cell walls of the ΔAoBck1 and ΔAoMkk1 mutants were severely damaged, and the Woronin body failed to respond to cellular damage. In particular, the mutants lost the ability to produce mycelial traps for nematode predation. Taken together, AoBck1 and AoMkk1 play a conserved role in mycelial growth and development, CWI, conidiation, multi-stress tolerance, trap formation, and pathogenicity.     Risk prediction model establishment with tri-phasic CT image features for differential diagnosis of adrenal pheochromocytomas and lipid-poor adenomas: Grouping method

    ObjectivesThe purpose of this study was to establish a risk prediction model for differential diagnosis of pheochromocytomas (PCCs) from lipid-poor adenomas (LPAs) using a grouping method based on tri-phasic CT image features.MethodsIn this retrospective study, we enrolled patients that were assigned to a training set (136 PCCs and 183 LPAs) from two medical centers, along with an external independent validation set (30 PCCs and 54 LPAs) from another center. According to the attenuation values in unenhanced CT (CTu), the lesions were divided into three groups: group 1, 10 HU &lt; CTu ≀ 25 HU; group 2, 25 HU &lt; CTu ≀ 40 HU; and group 3, CTu &gt; 40 HU. Quantitative and qualitative CT imaging features were calculated and evaluated. Univariate, ROC, and binary logistic regression analyses were applied to compare these features.ResultsCystic degeneration, CTu, and the peak value of enhancement in the arterial and venous phase (DEpeak) were independent risk factors for differential diagnosis of adrenal PCCs from LPAs. In all subjects (groups 1, 2, and 3), the model formula for the differentiation of PCCs was as follows: Y = -7.709 + 3.617*(cystic degeneration) + 0.175*(CTu ≄ 35.55 HU) + 0.068*(DEpeak ≄ 51.35 HU). ROC curves were drawn with an AUC of 0.95 (95% CI: 0.927–0.973) in the training set and 0.91 (95% CI: 0.860–0.929) in the external validation set.ConclusionA reliable and practical prediction model for differential diagnosis of adrenal PCCs and LPAs was established using a grouping method

    Effects of different soil water holding capacities on vegetable residue return and its microbiological mechanism

    With the gradual expansion of the protected vegetable planting area, dense planting stubbles and increasing labor cost, the treatment of vegetable residues has become an urgent problem to be solved. Soil bacterial community structure plays an important role in vegetable residue return and is susceptible to environmental changes. Therefore, understanding the influences of different soil water holding capacities on plant residue decomposition and soil bacterial communities is important for biodegradation. During the whole incubation period, the weight loss ratio of plant residue with 100% water holding capacity was 69.60 to 75.27%, which was significantly higher than that with 60% water holding capacity in clay and sandy soil, indicating that high water holding capacity promoted the decomposition of plant residue. The degradation of lignin and cellulose was also promoted within 14 days. Furthermore, with the increase in soil water holding capacity, the contents of NH4+ increased to 5.36 and 4.54 times the initial value in the clay and sandy soil, respectively. The increase in napA and nrfA resulted in the conversion of NO3– into NH4+. The increase in water holding capacity made the bacterial network structure more compact and changed the keystone bacteria. The increase in water holding capacity also increased the relative abundance of Firmicutes at the phylum level and Symbiobacterium, Clostridium at the genus level, which are all involved in lignin and cellulose degradation and might promote their degradation. Overall, these findings provide new insight into the effects of different soil water holding capacities on the degradation of plant residues in situ and the corresponding bacterial mechanisms

    Finishing the euchromatic sequence of the human genome

    The sequence of the human genome encodes the genetic instructions for human physiology, as well as rich information about human evolution. In 2001, the International Human Genome Sequencing Consortium reported a draft sequence of the euchromatic portion of the human genome. Since then, the international collaboration has worked to convert this draft into a genome sequence with high accuracy and nearly complete coverage. Here, we report the result of this finishing process. The current genome sequence (Build 35) contains 2.85 billion nucleotides interrupted by only 341 gaps. It covers ∌99% of the euchromatic genome and is accurate to an error rate of ∌1 event per 100,000 bases. Many of the remaining euchromatic gaps are associated with segmental duplications and will require focused work with new methods. The near-complete sequence, the first for a vertebrate, greatly improves the precision of biological analyses of the human genome including studies of gene number, birth and death. Notably, the human enome seems to encode only 20,000-25,000 protein-coding genes. The genome sequence reported here should serve as a firm foundation for biomedical research in the decades ahead

    A 3D fractional-order nonlinear system with coexisting chaotic attractors is proposed in this paper. The necessary condition of the existence chaos is q≄0.8477. The fractional-order system exhibits chaotic attractors with the order as low as 2.5431. The largest Lyapunov exponent varying as fractional order q is given. Furthermore, there are the coexisting “positive attractor” and “negative attractor” in this fractional-order chaotic system, and the necessary condition for “positive attractor” and “negative attractor” is obtained. Meanwhile, a control scheme for the stabilization of the unstable equilibrium is suggested via a single state variable linear controller. Numerical results show that the control scheme is valid
