    Spectrum-effect relationship between HPLC fingerprints and inhibitory activity in MUC5AC mucin of Pinelliae Rhizoma Praeparatum Cum Alumine

    Purpose: To investigate the spectrum-effect relationship between HPLC fingerprints and the inhibitory effect on MUC5AC mucin of Pinelliae Rhizoma Praeparatum Cum Alumine (PRPCA).Methods: The fingerprints of 20 PRPCA batches were established using HPLC and their similarities or differences were analyzed using hierarchical cluster analysis (HCA) and principal component analysis (PCA). The inhibitory effects of MUC5AC mucin were evaluated in LPS-treated NCI-H292 cells. The spectrum-effect relationship between common chromatographic peaks and MUC5AC inhibition was established using a partial least squares-discriminant analysis (PLS-DA).Results: Fifteen common chromatographic peaks were identified by analyzing HPLC fingerprints, with uridine, tyrosine, uracil, and inosine found as possible markers to distinguish the PRPCA from different sources. Spectrum-effect relationship analysis showed that the chromatographic peaks 5, 6, 10 (vernine), 12 (5-hydroxymethylfurfural), 14 (tryptophan) and 15 (adenosine) were closely associated with the inhibitory effect on MUC5AC mucin.Conclusion: The spectrum-effect relationship between HPLC fingerprints and the inhibitory effect on MUC5AC mucin of PRPCA was successfully established in the present study. Our findings further reveal the material basis of PRPCA and provide an effective method for its quality control

    Extraction optimization and characterization of polysaccharide antioxidants from Pinellia ternata (Thunb) Breit rhizome

    Purpose: To investigate the optimum extraction conditions of polysaccharides from Pinellia Rhizoma (PRP) and their antioxidant activities.Methods: Response surface methodology (RSM) was applied to optimize the water extraction conditions of PRP by Box-Benhnken design (BBD). A high performance liquid chromatography (HPLC) method was performed for determining the monosaccharide composition, while a high performance gel permeation chromatography (HPGPC) method was established for determining the molecular weight distribution of PRP. In addition, scavenging DPPH and superoxide anion radical scavenging assays were used to evaluate the antioxidant activities of PRP.Results: As a result, the optimum extraction conditions of PRP were as follows: extraction time, 103 min; solid-liquid ratio, 24 g/mL; and extraction no., 3. Under these conditions, the maximum extraction yield of PRP was 2.47 %, which matched the optimum value (2.55 %) predicted by RSM. The results of HPLC analysis suggest that the monosaccharide composition of PRP was mannose (Man), galactose acid (Gal acid), galactose (Gal), glucose (Glc), and arabinose (Ara) in molar ratios of 5.76:2.20:2.64:4.57:1, respectively, and the molecular weight ranged from 13592 to 445065. Furthermore, PRP showed marked antioxidant activities with the highest DPPH free radical scavenging rate of 71.5 % and O2- free radical scavenging rate of 87.5 % at a concentration of 10 mg/mL.Conclusion: RSM is a rapid and effective statistical technique for optimizing conditions for PRP extraction.     Spectrum-effect relationships between high performance liquid chromatography fingerprints and bioactivities of charred areca nut

    Purpose: To investigate the spectrum-effect relationships between high performance liquid chromatography (HPLC) fingerprints and duodenum contractility of charred areca nut (CAN) on rats.Methods: An HPLC method was used to establish the fingerprint of charred areca nut (CAN). The promoting effect on contractility of intestinal smooth was carried out to evaluate the duodenum contractility of CAN in vitro. In addition, the spectrum-effect relationships between HPLC fingerprints and bioactivities of CAN were investigated using multiple linear regression analysis (backward method).Results: Fourteen common peaks were detected and peak 3 (5-Hydroxymethyl-2-furfural, 5-HMF) was selected as the reference peak to calculate the relative retention time of 13 other common peaks. In addition, the equation of spectrum-effect relationships {Y = 3.818 - 1.126X1 + 0.817X2 - 0.045X4 - 0.504X5 + 0.728X6 - 0.056X8 + 1.122X9 - 0.247X13 - 0.978X14 (p < 0.05, R2 = 1)} was established in the present study by the multiple linear regression analysis (backward method). According to the equation, the absolute value of the coefficient before X1, X2, X4, X5, X6, X8, X9, X13, X14 was the coefficient between the component and the parameter.Conclusion: The model presented in this study successfully unraveled the spectrum-effect relationship of CAN, which provides a promising strategy for screening effective constituents of areca nut.Keywords: Charred areca nut, Spectrum-effect relationships, HPLC fingerprints, Duodenum contractilit

    No effect of occupational noise exposure on auditory brainstem response and speech perception in noise

    The primary aim of this study was to investigate whether auditory brainstem response (ABR) and speech perception in noise (SPiN) were associated with occupational noise exposure in normal hearing young factory workers. Forty young adults occupationally exposed to noise and 40 non-exposed young adults (control group) from Zhejiang province in China were selected. All participants presented with normal hearing thresholds and distortion product otoacoustic emissions. Participants were evaluated with the Mandarin Bamford-Kowal-Bench (BKB) test and ABR. The latter was obtained for click stimulus at 50, 60, 70, 80, and 90 dBnHL. Peak-to-trough amplitudes and latencies for waves I and V were obtained. The ABR wave I amplitude, the wave I/V amplitude ratio, the slope of the wave I amplitude growth as a function of stimulus intensity (AMP-ISlope), and the wave V latency shift with ipsilateral noise (LAT-VSlope) were used as ABR outcomes. Finally, equivalent continuous average sound pressure level normalized to 8 h (LAeq.8h) and cumulative noise exposure (CNE) were obtained for noise-exposed participants. No significant differences between groups were found for any ABR outcomes. Noise-exposed participants exhibited worse BKB scores than control group participants. A multivariate regression model showed that 23.3% of the variance in BKB scores was explained by group category (exposed vs. non-exposed) and hearing thresholds. However, since none of the ABR outcomes exploring cochlear synaptopathy were associated with noise exposure, we cannot conclude that cochlear synaptopathy was the contributing factor for the differences between groups for BKB scores. Factors that go beyond sensory processing may explain such results, especially given socio-economic differences between the noise-exposed and control groups. We conclude that in this sample of participants, occupational noise exposure was not associated with signs of cochlear synaptopathy as measured by ABR and BKB

    Assessment of radiation exposure and public health before and after the operation of Sanmen nuclear power plant

    IntroductionSanmen nuclear power plant (SNPP) operates the first advanced passive (AP1000) nuclear power unit in China.MethodsTo assess the radiological impacts of SNPP operation on the surrounding environment and the public health, annual effective dose (AED) and excess risk (ER) were estimated based on continuous radioactivity monitoring in drinking water and ambient dose before and after its operation during 2014–2021. In addition, the residents' cancer incidence was further analyzed through authorized health data collection.ResultsThe results showed that the gross α and gross β radioactivity in all types of drinking water were ranged from 0.008 to 0.017 Bq/L and 0.032 to 0.112 Bq/L, respectively. The cumulative ambient dose in Sanmen county ranged from 0.254 to 0.460 mSv/y, with an average of 0.354 ± 0.075 mSv/y. There is no statistical difference in drinking water radioactivity and ambient dose before and after the operation of SNPP according to Mann–Whitney U test. The Mann-Kendall test also indicates there is neither increasing nor decreasing trend during the period from 2014 to 2021. The age-dependent annual effective doses due to the ingestion of drinking water or exposure to the outdoor ambient environment are lower than the recommended threshold of 0.1 mSv/y. The incidence of cancer (include leukemia and thyroid cancer) in the population around SNPP is slightly higher than that in other areas, while it is still in a stable state characterized by annual percentage changes.DiscussionThe current comprehensive results show that the operation of SNPP has so far no evident radiological impact on the surrounding environment and public health, but continued monitoring is still needed in the future

    Traditional Uses, Origins, Chemistry and Pharmacology of Bombyx batryticatus: A Review

    Bombyx batryticatus (B. batryticatus), a well-known traditional animal Chinese medicine, has been commonly used in China for thousands of years. The present paper reviewed advances in traditional uses, origin, chemical constituents, pharmacology and toxicity studies of B. batryticatus. The aim of the paper is to provide more comprehensive references for modern B. batryticatus study and application. In Traditional Chinese Medicine (TCM) culture, drugs containing B. batryticatus have been used to treat convulsions, headaches, skin prurigo, scrofula, tonsillitis and fever. Many studies indicate B. batryticatus contains various compounds, including protein and peptides, fatty acids, flavonoids, nucleosides, steroids, coumarin, polysaccharide and others. Numerous investigations also have shown that extracts and compounds from B. batryticatus exert a wide spectrum of pharmacological effects both in vivo and in vitro, including effects on the nervous system, anticoagulant effects, antitumor effects, antibacterial and antifungal effects, antioxidant effects, hypoglycemic effects, as well as other effects. However, further studies should be undertaken to investigate bioactive compounds (especially proteins and peptides), toxic constituents, using forms and the quality evaluation and control of B. batryticatus. Furthermore, it will be interesting to study the mechanism of biological activities and structure-function relationships of bioactive constituents in B. batryticatus

    Childhood pesticide poisoning in Zhejiang, China: a retrospective analysis from 2006 to 2015

    Abstract Background Pesticide poisoning in children has been a serious public health issue around the world, especially in the developing countries where agriculture is still one of the largest economic sectors. The purpose of this study was to analyze epidemiological characteristics of acute pesticide poisoning in children from Zhejiang province, China. Methods The pesticide poisoning cases for children were retrieved from Occupational Disease Surveillance and Reporting System, Zhejiang Provincial Center for Disease Control and Prevention, China. The incident cases, deaths, and fatality rate of child pesticide poisoning from 2006 through 2015 were calculated. Results During the study period, totally 2952 children were poisoned by pesticides, with 66 deaths, resulting in a fatality rate of 2.24%. Among them, there were 1607 male cases with 28 deaths, and 1345 female cases with 38 deaths. Most of the cases occurred in preschool children (1349) and adolescent age group (1269). Organophosphate and carbamate insecticides were the cause of most poisonings (1130), leading to 34 deaths. The highest fatality rate (3.13%) was due to poisoning by herbicides and fungicides, causing 14 deaths out of 448 cases. Poisoning occurred mostly in rural areas (78%). And most pesticide poisoning occurred in the summer (896) and fall (811), while fewest poisoning cases in the winter (483) but with the highest fatality rate (3.52%). Conclusions This study shows that pesticide poisoning of children is a major health problem in Zhejiang, suggesting preventive strategies should be conducted to control childhood pesticide poisoning
