    An ovarian mucinous cystadenoma with adnexal tuberculosis: a case report

    We report the occurrence of a case of a benign ovarian tumour- mucinous cystadenoma ovary with adnexal tuberculosis. Our case was a middle-aged Indian woman who presented with abdominal distension and discomfort at the gynaecology clinic of M.Y. hospital, Indore. The data were collected by history-taking, clinical examination, laboratory investigations, transabdominal ultrasonographic examination, and by histopathological study of the excised surgical specimen. It was reported as ovarian mucinous cystadenoma with adnexal TB. This case report emphasizes the significance of thorough evaluation of all women presenting with vague abdominal pains and thorough search of any other pathology in the specimen, like in our case it was tuberculosis. With the increasing awareness of such conditions, more and more cases could be detected and reported.

    Cytopathological study of salivary gland lesion in patients at a tertiary care centre, Indore: a one-year study

    Background: Fine needle aspiration cytology (FNAC) is being increasingly used in the diagnosis of salivary gland lesions. However, the diagnosis is still difficult sometimes. Salivary gland lesions account for 2-6.5% of all the neoplasms of the head and neck. The objective of this study was to evaluate the diagnostic accuracy and the sensitivity and specificity of FNAC in various salivary gland lesions and their histological correlation wherever possible.Methods: A total of 123 FNACs were done on salivary gland swellings in the Department of Pathology, M.G.M. Medical College, Indore, Madhya Pradesh, India. The PAP stained cytological and H and E stained histopathological slides were studied and correlated.Results: On FNAC, slides were studied and analyzed and the following lesions were observed: Inflammatory lesion (69), Inflammatory cystic lesion (1), Pleomorphic adenoma (29), monomorphic adenoma (1), Warthin’s tumour (2), Benign Cystic lesion (4), Benign lesion (not specified) (4), Adenoid cystic carcinoma (3), Mucoepidermoid carcinoma (2), Metastatic carcinoma (3), and malignant tumour (unspecified) (5). Histopathological correlation was available in 41 cases and studied respectively.Conclusions: The overall sensitivity, specificity and the diagnostic accuracy were 90.2%, 97.6%, and 90.2%, respectively. Hence, the appropriate therapeutic management could be planned earlier. This study documents that FNAC of the salivary gland tumours is accurate, simple, rapid, inexpensive, well tolerated and harmless procedure for the patient

    Primary intracranial squamous cell carcinoma is extremely rare, with most cases arising from malignant transformation of dysembryogenetic lesions such as epidermoid and dermoid cysts. Intracranial squamous cell neoplasm arising de novo is even rarer and has been reported in only four patients to date. We herein describe a case of primary intracranial squamous cell carcinoma arising de novo in the right frontal lobe in a 35-year-old woman treated with a combination of surgery and postoperative conformal radiation. We have also shed light on the biology and the therapeutic options of this enigmatic tumour.Pierwotny śródczaszkowy rak płaskonabłonkowy jest wyjątkową rzadkością i w większości przypadków rozwija się w wyniku zezłośliwienia zmian o charakterze dysembriogenetycznym, np. torbieli naskórkowej lub skórzastej. Śródczaszkowy rak płaskonabłonkowy powstały de novo jest jeszcze rzadszy – dotąd opisano 4 takie przypadki. W niniejszej pracy przedstawiono przypadek nowotworu powstałego de novo w prawym płacie czołowym u 35-letniej chorej, którą z tego powodu poddano leczeniu chirurgicznemu i pooperacyjnej radioterapii konformalnej. Podano również informacje na temat biologii i możliwości leczenia tego zagadkowego guza

    Genome wide expression profiling of two accession of G. herbaceum L. in response to drought

    <p>Abstract</p> <p>Background</p> <p>Genome-wide gene expression profiling and detailed physiological investigation were used for understanding the molecular mechanism and physiological response of <it>Gossypium herbaceum</it>, which governs the adaptability of plants in drought conditions. Recently, microarray-based gene expression analysis is commonly used to decipher genes and genetic networks controlling the traits of interest. However, the results of such an analysis are often plagued due to a limited number of genes (probe sets) on microarrays. On the other hand, pyrosequencing of a transcriptome has the potential to detect rare as well as a large number of transcripts in the samples quantitatively. We used Affymetrix microarray as well as Roche's GS-FLX transcriptome sequencing for a comparative analysis of cotton transcriptome in leaf tissues under drought conditions.</p> <p>Results</p> <p>Fourteen accessions of <it>Gossypium herbaceum </it>were subjected to mannitol stress for preliminary screening; two accessions, namely Vagad and RAHS-14, were selected as being the most tolerant and most sensitive to osmotic stress, respectively. Affymetrix cotton arrays containing 24,045 probe sets and Roche's GS-FLX transcriptome sequencing of leaf tissue were used to analyze the gene expression profiling of Vagad and RAHS-14 under drought conditions. The analysis of physiological measurements and gene expression profiling showed that Vagad has the inherent ability to sense drought at a much earlier stage and to respond to it in a much more efficient manner than does RAHS-14. Gene Ontology (GO) studies showed that the phenyl propanoid pathway, pigment biosynthesis, polyketide biosynthesis, and other secondary metabolite pathways were enriched in Vagad under control and drought conditions as compared with RAHS-14. Similarly, GO analysis of transcriptome sequencing showed that the GO terms <it>responses to various abiotic stresses </it>were significantly higher in Vagad. Among the classes of transcription factors (TFs) uniquely expressed in both accessions, RAHS-14 showed the expression of ERF and WRKY families. The unique expression of ERFs in response to drought conditions reveals that RAHS-14 responds to drought by inducing senescence. This was further supported by transcriptome analysis which revealed that RAHS-14 responds to drought by inducing many transcripts related to senescence and cell death.</p> <p>Conclusion</p> <p>The comparative genome-wide gene expression profiling study of two accessions of <it>G.herbaceum </it>under drought stress deciphers the differential patterns of gene expression, including TFs and physiologically relevant processes. Our results indicate that drought tolerance observed in Vagad is not because of a single molecular reason but is rather due to several unique mechanisms which Vagad has developed as an adaptation strategy.</p

    Transcriptomic and metabolomic shifts in rice roots in response to Cr (VI) stress

    <p>Abstract</p> <p>Background</p> <p>Widespread use of chromium (Cr) contaminated fields due to careless and inappropriate management practices of effluent discharge, mostly from industries related to metallurgy, electroplating, production of paints and pigments, tanning, and wood preservation elevates its concentration in surface soil and eventually into rice plants and grains. In spite of many previous studies having been conducted on the effects of chromium stress, the precise molecular mechanisms related to both the effects of chromium phytotoxicity, the defense reactions of plants against chromium exposure as well as translocation and accumulation in rice remain poorly understood.</p> <p>Results</p> <p>Detailed analysis of genome-wide transcriptome profiling in rice root is reported here, following Cr-plant interaction. Such studies are important for the identification of genes responsible for tolerance, accumulation and defense response in plants with respect to Cr stress. Rice root metabolome analysis was also carried out to relate differential transcriptome data to biological processes affected by Cr (VI) stress in rice. To check whether the Cr-specific motifs were indeed significantly over represented in the promoter regions of Cr-responsive genes, occurrence of these motifs in whole genome sequence was carried out. In the background of whole genome, the lift value for these 14 and 13 motifs was significantly high in the test dataset. Though no functional role has been assigned to any of the motifs, but all of these are present as promoter motifs in the Database of orthologus promoters.</p> <p>Conclusion</p> <p>These findings clearly suggest that a complex network of regulatory pathways modulates Cr-response of rice. The integrated matrix of both transcriptome and metabolome data after suitable normalization and initial calculations provided us a visual picture of the correlations between components. Predominance of different motifs in the subsets of genes suggests the involvement of motif-specific transcription modulating proteins in Cr stress response of rice.</p

    A cross section of skin diseases in rural Allahabad

    <b>Background: </b> The pattern of skin diseases varies form one country to another and across different parts within the same country. A two-day multispecialty medical camp was held among the local population at the district of Kausambi, Allahabad, UP, in October 2005. A cross section of pattern of skin diseases observed at the camp is reported and compared with similar studies in literature. <b> Materials and Methods: </b> All cases attending the medical camp were included in the study. All those with dermatological complaints were examined in detail, brief relevant history was elicited and clinical diagnosis was made. <b> Results: </b> Skin diseases comprised 7.86&#x0025; of all those who attended the camp. The 11-20 year age group was the most common age group involved with 164 (31.4&#x0025;) cases. Infective disorders were found in 59.1&#x0025; and noninfective disorders in 40.9&#x0025; of all the skin cases. Among the infective disorders, fungal infections were most common (54.52&#x0025;), and among the noninfective dermatoses, eczemas were most common (39.2&#x0025;) cutaneous disorders. <b> Conclusion: </b> Our study brought out a higher prevalence of infective dermatoses and a relatively higher, but statistically insignificant, prevalence of fungal infections, scabies and eczemas, thereby reflecting minor regional variance in our study group

    A T9G Mutation in the Prototype TATA-Box TCACTATATATAG Determines Nucleosome Formation and Synergy with Upstream Activator Sequences in Plant Promoters1[W]

    We had earlier reported that mutations to G and C at the seventh and eighth positions in the prototype TATA-box TCACTATATATAG inhibited light-dependent activation of transcription from the promoter. In this study, we characterized mutations at the ninth position of the prototype TATA-box. Substitution of T at the ninth position with G or C enhanced transcription from the promoter in transgenic tobacco (Nicotiana tabacum) plants. The effect of T9G/C mutations was not light dependent, although the 9G/C TATA-box showed synergy with the light-responsive element (lre). However, the 9G/C mutants in the presence of lre failed to respond to phytochromes, sugar, and calcium signaling, in contrast to the prototype TATA-box with lre. The 9G/C mutation shifted the point of initiation of transcription, and transcription activation was dependent upon the type of activating element present upstream. The synergy in activation was noticed with lre and legumin activators but not with rbcS, Pcec, and PR-1a activators. The 9G mutation resulted in a micrococcal nuclease-sensitive region over the TATA-box, suggesting a nucleosome-free region, in contrast to the prototype promoter, which had a distinct nucleosome on the TATA-box. Thus, the transcriptional augmentation with mutation at the ninth position might be because of the loss of a repressive nucleosomal structure on the TATA-box. In agreement with our findings, the promoters containing TATAGATA as identified by genome-wide analysis of Arabidopsis (Arabidopsis thaliana) are not tightly repressed

    Angiomyomatous hamartoma of lymph nodes: Clinicopathological study of 6 cases with review of literature

    Angiomyomatous hamartoma (AMH) is a rare disease with predisposition for inguinal and femoral lymph nodes. Histologically, it is characterized by replacement of lymph nodal parenchyma with irregularly distributed thick walled blood vessels, haphazardly arranged smooth muscle cells, variable amount of fat and fibrous tissue in a sclerotic lymphatic stroma. Few cases have also been reported in popliteal and sub - mandibular location. The exact pathogenesis is still not known. Although this entity is very rare, its recognition is important in discriminating it from other benign and malignant vascular lesions of lymph nodes
