    Bulk Water Exports: Environmental Concerns and Business Realities

    Hydrologists expect the demand for water will continue to increase with the world’s growing population. As a result, some have predicted that by the year 2025, as many as 3.5 billion people will be living in water stressed or water scarce countries, compared to 500 million people in 2002. Water scarcity is also exacerbated as peoples’ lifestyles become more industrialized and thus more consumptive on a per capita basis. The aim of this paper is to concentrate on one aspect of the complex issue of water exports, namely, the feasibility and practicality of a water export business and its various impacts. The issue of water exports will be put in perspective, because if such exports are not attractive from a business point of view, it is unlikely that such exports will ever take place. The term water exports will be used in this paper to refer to bulk transfers of water from one location to another using tankers or water bags. This kind of transfer is generally considered a type of medium scale export, as opposed to sales of bottled water (small scale) and watershed diversions (large-scale)

    The Future City: A Selection of Views on the Reorganization of Government in Greater Winnipeg

    Book: pp.54; digital file.This is a collection of short essays on the potential workings and outcomes of the Unicity scheme then proposed by the Manitoba provincial government, and attempts to give careful consideration and critique of the scheme. Essays are written by researchers and academics from the University of Winnipeg, members of the local media and the private sector. The paper argues that amalgamation into a regional government will have both positive and negative effects. Generally, the provision of equitable levels of services and effective planning for the entire region are viewed positively, while the potential for Unicity to become too unwieldy and too under-representative of marginalized communities is seen as a cause for concern. An additional criticism that arises from these essays, is that partisan politics will become more firmly rooted at the municipal level in Winnipeg, on account of Unicity’s proposed parliamentary-style city council. The test of Unicity, therefore, is the degree to which the proposed community committees and ward representation are able to flourish. The editor concludes by saying that the Unicity has the potential to be a framework for a new, more democratic style of politics through greater citizen engagement. This end must be actively pursued, he writes, otherwise the new framework will be governed by the same old politics that characterized Greater Winnipeg in recent years

    The Moyjil site, south-west Victoria, Australia : Stratigraphic and geomorphic context

    Shelly deposits at Moyjil (Point Ritchie, Warrnambool), Victoria, together with ages determined from a variety of techniques, have long excited interest in the possibility of a preserved early human influence in far south-eastern Australia. This paper presents a detailed analysis of the stratigraphy of the host Bridgewater Formation (Pleistocene) at Moyjil and provides the context to the shelly deposits, evidence of fire and geochronological sampling. We have identified five superposed calcarenite–palaeosol units in the Bridgewater Formation, together with two prominent erosional surfaces that may have hosted intensive human activity. Part of the sequence is overlain by the Tower Hill Tuff, previously dated as 35 ka. Coastal marine erosion during the Last Interglacial highstand created a horizontal surface on which deposits of stones and shells subsequently accumulated. Parts of the erosional surface and some of the stones are blackened, perhaps by fire. The main shell deposit was formed by probable mass flow, and additional shelly remains are dispersed in the calcareous sand that buried the surface

    Dynamics of Dynamin during Clathrin Mediated Endocytosis in PC12 Cells

    Members of the dynamin super-family of GTPases are involved in disparate cellular pathways. Dynamin1 and dynamin2 have been implicated in clathrin-mediated endocytosis. While some models suggest that dynamin functions specifically at the point of vesicle fission, evidence also exists for a role prior to fission during vesicle formation and it is unknown if there is a role for dynamin after vesicle fission. Although dynamin2 is ubiquitously expressed, dynamin1 is restricted to the nervous system. These two structurally similar endocytic accessory proteins have not been studied in cells that endogenously express both.The present study quantitatively assesses the dynamics of dynamin1 and dynamin2 during clathrin-mediated endocytosis in PC12 cells, which endogenously express both proteins. Both dynamin isoforms co-localized with clathrin and showed sharp increases in fluorescence intensity immediately prior to internalization of the nascent clathrin-coated vesicle. The fluorescence intensity of both proteins then decreased with two time constants. The slower time constant closely matched the time constant for the decrease of clathrin intensity and likely represents vesicle movement away from the membrane. The faster rate may reflect release of dynamin at the neck of nascent vesicle following GTP hydrolysis.This study analyses the role of dynamin in clathrin-mediated endocytosis in a model for cellular neuroscience and these results may provide direct evidence for the existence of two populations of dynamin associated with nascent clathrin-coated vesicles

    The phocein homologue SmMOB3 is essential for vegetative cell fusion and sexual development in the filamentous ascomycete Sordaria macrospora

    Members of the striatin family and their highly conserved interacting protein phocein/Mob3 are key components in the regulation of cell differentiation in multicellular eukaryotes. The striatin homologue PRO11 of the filamentous ascomycete Sordaria macrospora has a crucial role in fruiting body development. Here, we functionally characterized the phocein/Mob3 orthologue SmMOB3 of S. macrospora. We isolated the gene and showed that both, pro11 and Smmob3 are expressed during early and late developmental stages. Deletion of Smmob3 resulted in a sexually sterile strain, similar to the previously characterized pro11 mutant. Fusion assays revealed that ∆Smmob3 was unable to undergo self-fusion and fusion with the pro11 strain. The essential function of the SmMOB3 N-terminus containing the conserved mob domain was demonstrated by complementation analysis of the sterile S. macrospora ∆Smmob3 strain. Downregulation of either pro11 in ∆Smmob3, or Smmob3 in pro11 mutants by means of RNA interference (RNAi) resulted in synthetic sexual defects, demonstrating for the first time the importance of a putative PRO11/SmMOB3 complex in fruiting body development
