    The national impact of Experience Based Career Education upon 1978-1982 graduates

    This study presents the findings of the nation-wide impact study of Experience Based Career Education upon the graduates of 1978-1982. Eight hundred sixty surveys were sent to an experimental sample of students who were involved in EBCE projects sponsored by all four of the National Institute of Education Research Laboratories. One hundred forty surveys were sent to a control group of students involved in Cooperative Vocational Education in the State of Iowa;Four research questions were investigated to determine the relationship of student characteristics (age, gender, community size, grade level, previous work experience and project group) and program characteristics (Model of EBCE, type of instructional support, length of time in program, number of sites explored, amount of time at site each week, and use of related instruction) to: (1) Student perception of personal-social skill development; (2) Student perception of world-of-work skill development;In addition, 12 descriptive questions were studied and the results presented in charts, tables and narration. From the information gained through the analysis and findings of this study, the following conclusions were drawn. (All four major hypotheses were rejected at the .01 level of significance). (1) Enrollment in EBCE, being a member of the female sex and having previous work experience were significant contributors toward positive personal-social skill development. (2) Enrollment in EBCE and previous work experience were significant contributors toward positive acquistion of world-of-work skills. (3) Enrollment in EBCE for more than one semester and involvement in experiential academics contributed significantly to personal-social skill development. (4) Enrollment in the Appalachia Educational Research or Northwest Regional models of EBCE, participation for two or more semesters and enrollment in related science contributed significantly to world-of-work skill acquisition

    Central neurogenic hyperventilation: pharmacologic intervention with morphine sulfate and correlative analysis of respiratory, sleep, and ocular motor dysfunction.

    Journal ArticleCentral neurogenic hyperventilation (CNH), for which there is no effective therapy, can eventually result in respiratory fatigue and death. This report describes a patient with CNH due to a brainstem anaplastic astrocytoma who also exhibited disturbances of sleep and ocular motor function. The CNH responded clinically to morphine sulfate and methadone. Analysis of ventilatory response to CO2 before and after morphine demonstrated a depression of ventilatory response (49 to 53% of baseline) and occlusion pressure response (35 to 50% of baseline) to CO2, with a requirement for high doses of naloxone (10 mg IV) to reverse the effect. Polysomnography revealed sustained hyperventilation, elevated O2 saturation, and low end-tidal CO2 throughout all stages of non-rapid eye movement (NREM) sleep, and absence of rapid eye movement (REM) sleep. Ocular motor evaluation disclosed absence of horizontal and reflexive saccades with compensatory head thrusts. Correlation of the clinical and physiologic data with the MRI abnormalities suggested that the lesion responsible for CNH in this patient might reside in the medial tegmental parapontine reticular formation. Since recurrent episodes of hyperventilation responded in a sustained fashion to IV and oral opiates, this treatment may warrant consideration in other patients with CNH

    Automatic calibration of modulated fractional-N frequency synthesizers

The certified thesis is available in the Institute Archives and Special Collections.The focus of this research has been the development of a low power, radio frequency transmitter architecture. Specifically, a technique for in service automatic calibration of a modulated phase locked loop (PLL) frequency synthesizer has been developed. Phase/frequency modulation is accomplished by modulating the feedback divide value in a phase locked loop frequency synthesizer. A digital precompensation filter is used to extend the modulation bandwidth by canceling the low-pass transfer function of the PLL. The automatic calibration circuit maintains accurate matching between the digital precompensation filter and the analog PLL transfer function across process and temperature variations. The automatic calibration circuit, which is the main contribution of this thesis, operates while the transmitter is in service. This online calibration eliminates the need for production calibration and periodic down time for calibration cycles.(cont.) In addition the calibration circuitry provides greater accuracy in the modulation than what is possible via offline methods of calibration. The calibration circuit works with M-ary GFSK as well as 2 level GFSK. The automatic calibration circuit has been implemented in two forms to prove its operation. The first version is a circuit board level implementation with a center frequency of around 60 MHz. The second implementation of the system is in a full custom 0.6 ,Lm BiCMOS integrated circuit. The integrated circuit contains the complete synthesizer with automatic calibration and operates in the 1.88 GHz frequency band used by the Digital European Cordless Telephone (DECT) standard. A data rate of 2.5 Mbps using 2 level GFSK and 5.0 Mbps using 4 level GFSK has been achieved with a power consumption of 78 mW.by Daniel R. McMahill.Ph.D

    Synthesis-Dependent Strand Annealing in Meiosis

    Recent studies led to the proposal that meiotic gene conversion can result after transient engagement of the donor chromatid and subsequent DNA synthesis-dependent strand annealing (SDSA). Double Holliday junction (dHJ) intermediates were previously proposed to form both reciprocal crossover recombinants (COs) and noncrossover recombinants (NCOs); however, dHJs are now thought to give rise mainly to COs, with SDSA forming most or all NCOs. To test this model in Saccharomyces cerevisiae, we constructed a random spore system in which it is possible to identify a subset of NCO recombinants that can readily be accounted for by SDSA, but not by dHJ-mediated recombination. The diagnostic class of recombinants is one in which two markers on opposite sides of a double-strand break site are converted, without conversion of an intervening heterologous insertion located on the donor chromatid. This diagnostic class represents 26% of selected NCO recombinants. Tetrad analysis using the same markers provided additional evidence that SDSA is a major pathway for NCO gene conversion in meiosis

    SOAT1 missense variant in two cats with sebaceous gland dysplasia.

    Spontaneously arisen hereditary diseases in domestic animals provide an excellent opportunity to study the physiological functions of the altered genes. We investigated two 4-month-old sibling domestic short haired kittens with dry dark debris around the eyes, nose, and ears, dark crusting on the legs and a thin poor hair coat. Skin biopsies revealed abnormal sebaceous gland morphology with lack of normal sebocyte arrangement and differentiation. Hair follicles had a distorted silhouette, interpreted as a change secondary to the observed sebaceous gland dysplasia. Whole genome sequencing on both affected kittens and 65 genetically diverse feline genomes was performed. Filtering for variants that were present in both kittens but absent from the control genomes revealed a homozygous missense variant in SOAT1, encoding sterol O-acyltransferase 1. The protein is localized in the endoplasmic reticulum and catalyzes the formation of cholesteryl esters, an essential component of sebum and meibum. The identified SOAT1:c.1531G > A variant is predicted to change a highly conserved glycine residue within the last transmembrane domain of SOAT1, p.Gly511Arg. In mice, variants in Soat1 or complete knockout of the gene lead to the "hair interior defect" (hid) or abnormal Meibomian glands, respectively. SOAT1:c.1531G > A represents a plausible candidate variant for the observed sebaceous gland dysplasia in both kittens of this study. The variant was not present in 10 additional cats with a similar clinical and histopathological phenotype suggesting genetic heterogeneity. SOAT1 variants should be considered as potential cause in hereditary sebaceous gland dysplasias of humans and domestic animals

    Diagnosed prevalence of Alzheimer’s disease and related dementias in Medicare Advantage plans

    IntroductionOne- third of Medicare beneficiaries are enrolled in Medicare Advantage (MA). Yet, little is known about MA beneficiaries diagnosed with Alzheimer’s disease (AD) and AD- related dementias (AD/ADRD).MethodsWe calculated the prevalence of AD/ADRD diagnoses in 2014 and 2016 in three MA plans. We determined the demographic characteristics of beneficiaries diagnosed with AD/ADRD, and whether they disenrolled from the MA plan for any reason within 364 days from the index date.ResultsIn 2014 and 2016, the overall prevalence of AD/ADRD diagnoses was 5.6% and 6.5%, respectively. In 2016, AD/ADRD beneficiaries were on average 82.4 (SD = 7.4) years of age, 61.8% female, and had multiple comorbidities. By 364 days post- index date, 32% of beneficiaries with AD/ADRD had disenrolled from their plan.     Smc5/6 coordinates formation and resolution of joint molecules with chromosome morphology to ensure meiotic divisions

    During meiosis, Structural Maintenance of Chromosome (SMC) complexes underpin two fundamental features of meiosis: homologous recombination and chromosome segregation. While meiotic functions of the cohesin and condensin complexes have been delineated, the role of the third SMC complex, Smc5/6, remains enigmatic. Here we identify specific, essential meiotic functions for the Smc5/6 complex in homologous recombination and the regulation of cohesin. We show that Smc5/6 is enriched at centromeres and cohesin-association sites where it regulates sister-chromatid cohesion and the timely removal of cohesin from chromosomal arms, respectively. Smc5/6 also localizes to recombination hotspots, where it promotes normal formation and resolution of a subset of joint-molecule intermediates. In this regard, Smc5/6 functions independently of the major crossover pathway defined by the MutLγ complex. Furthermore, we show that Smc5/6 is required for stable chromosomal localization of the XPF-family endonuclease, Mus81-Mms4Eme1. Our data suggest that the Smc5/6 complex is required for specific recombination and chromosomal processes throughout meiosis and that in its absence, attempts at cell division with unresolved joint molecules and residual cohesin lead to severe recombination-induced meiotic catastroph

    Pch2 Links Chromosome Axis Remodeling at Future Crossover Sites and Crossover Distribution during Yeast Meiosis

    Segregation of homologous chromosomes during meiosis I depends on appropriately positioned crossovers/chiasmata. Crossover assurance ensures at least one crossover per homolog pair, while interference reduces double crossovers. Here, we have investigated the interplay between chromosome axis morphogenesis and non-random crossover placement. We demonstrate that chromosome axes are structurally modified at future crossover sites as indicated by correspondence between crossover designation marker Zip3 and domains enriched for axis ensemble Hop1/Red1. This association is first detected at the zygotene stage, persists until double Holliday junction resolution, and is controlled by the conserved AAA+ ATPase Pch2. Pch2 further mediates crossover interference, although it is dispensable for crossover formation at normal levels. Thus, interference appears to be superimposed on underlying mechanisms of crossover formation. When recombination-initiating DSBs are reduced, Pch2 is also required for viable spore formation, consistent with further functions in chiasma formation. pch2Δ mutant defects in crossover interference and spore viability at reduced DSB levels are oppositely modulated by temperature, suggesting contributions of two separable pathways to crossover control. Roles of Pch2 in controlling both chromosome axis morphogenesis and crossover placement suggest linkage between these processes. Pch2 is proposed to reorganize chromosome axes into a tiling array of long-range crossover control modules, resulting in chiasma formation at minimum levels and with maximum spacing
