    A role for microbial selection in frescoes' deterioration in Tomba degli Scudi in Tarquinia, Italy

    Mural paintings in the hypogeal environment of the Tomba degli Scudi in Tarquinia, Italy, show a quite dramatic condition: the plaster mortar lost his cohesion and a white layer coating is spread over almost all the wall surfaces. The aim of this research is to verify if the activity of microorganisms could be one of the main causes of deterioration and if the adopted countermeasures (conventional biocide treatments) are sufficient to stop it. A biocide treatment of the whole environment has been carried out before the conservative intervention and the tomb has been closed for one month. When the tomb was opened again, we sampled the microorganisms present on the frescoes and we identified four Bacillus species and one mould survived to the biocide treatment. These organisms are able to produce spores, a highly resistant biological form, which has permitted the survival despite the biocide treatment. We show that these Bacillus strains are able to produce calcium carbonate and could be responsible for the white deposition that was damaging and covering the entire surface of the frescoes. Our results confirm that the sanitation intervention is non always resolutive and could even be deleterious in selecting harmful microbial communities

    Calcite moonmilk of microbial origin in the Etruscan Tomba degli Scudi in Tarquinia, Italy

    A white deposit covering the walls in the Stanza degli Scudi of the Tomba degli Scudi, Tarquinia, Italy, has been investigated. In this chamber, which is still preserved from any kind of intervention such as cleaning and sanitization, ancient Etruscans painted shields to celebrate the military power of the Velcha family. Scanning electron microscopy analysis has revealed the presence of characteristic nanostructures corresponding to a calcite secondary mineral deposit called moonmilk. Analysis of the microbial community identified Proteobacteria, Acidobacteria and Actinobacteria as the most common phyla in strong association with the moonmilk needle fibre calcite and nanofibers of calcium carbonate. Employing classical microbiological analysis, we isolated from moonmilk a Streptomyces strain able to deposit gypsum and calcium carbonate on plates, supporting the hypothesis of an essential contribution of microorganisms to the formation of moonmilk

    Motion artefacts in cone beam CT: an in vitro study about the effects on the images

    OBJECTIVE: In cone beam CT (CBCT), imperfect patient immobility, caused by involuntary movements, is one of the most important causes of artefacts and image quality degradation. Various works in literature address this topic, but seldom is the nature of the movement correlated with the type of artefact and the image degradation in a systematic manner, and the correlation analyzed and explained. METHODS: All three types of movements that can occur during a scan—nodding, tilting and rolling—were applied to a dry skull, in various manners from abrupt to gradual through the entire scan, at different times and angles, over a wide range of displacements. 84 scans were performed, with different skull movements, and the resulting images examined by two skilled radiologists, rated in a four-point scale and statistically analyzed. A commercial CBCT machine was used, featuring supine patient positioning. RESULTS: Different types of movements induce different artefacts, in different parts of the anatomy. In general, movement of short duration may lead to double contours (bilateral or monolateral depending upon the angle of the scan at which they occur), whereas gradual movements result into blurring. CONCLUSION: Not all movements cause motion artefacts that equally jeopardize the image. Rolling is the type of movement that most severely affects the image diagnostic value. ADVANCES IN KNOWLEDGE: These findings may help practitioners to identify the causes of motion artefacts and the resulting image degradation, and remediate them, and manufacturers to improve the patient-positioning devices

    Molecular changes underlying decay of sensory responses and enhanced seizure propensity in peritumoral neurons

    Background: Glioblastoma growth impacts on the structure and physiology of peritumoral neuronal networks, altering the activity of pyramidal neurons which drives further tumor progression. It is therefore of paramount importance to identify glioma-induced changes in pyramidal neurons, since they represent a key therapeutic target. Methods: We longitudinal monitored visual evoked potentials after the orthotopic implant of murine glioma cells into the mouse occipital cortex. With laser microdissection we analysed layer II-III pyramidal neurons molecular profile and with Local Field Potentials (LFP) recordings we evaluated the propensity to seizures in glioma-bearing animals with respect to control mice. Results: We determine the time course of neuronal dysfunction of glioma-bearing mice and we identify a symptomatic stage, based on the decay of visual response. At that time point, we microdissect layer II-III pyramidal neurons and evaluate the expression of a panel of genes involved in synaptic transmission and neuronal excitability. Compared to the control group, peritumoral neurons show a decrease in the expression of the SNARE complex gene SNAP-25 and the alpha1 subunit of the GABA-A receptor. No significant changes are detected in glutamatergic (i.e., AMPA or NMDA receptor subunit) markers. Further reduction of GABA-A signalling by delivery of a benzodiazepine inverse agonist, DMCM (methyl-6,7-dimethoxy-4-ethyl-beta-carboline-3-carboxylate) precipitates seizures in two mouse models of tumor-bearing mice. Conclusions: These studies reveal novel molecular changes that occur in the principal cells of the tumor-adjacent zone. These modifications may be therapeutically targeted to ameliorate patients' quality of life

    VADASE reliability and accuracy of real-time displacement estimation: Application to the Central Italy 2016 earthquakes

    The goal of this article is the illustration of the newfunctionalities of the VADASE (Variometric Approach for Displacements Analysis Stand-alone Engine) processing approach. VADASE was presented in previousworks as an approach able to estimate in real time the velocities and displacements in a global reference frame (ITRF), using high-rate (1 Hz or more) carrier phase observations and broadcast products (orbits, clocks) collected by a stand-alone GNSS receiver, achieving a displacements accuracy within 1-2 cm (usually better) over intervals up to a few minutes. It has been well known since the very first implementation and testing of VADASE that the estimated displacements might be impacted by two different effects: spurious spikes in the velocities due to outliers (consequently, displacements, obtained through velocities integration, are severely corrupted) and trends in the displacements time series, mainly due to broadcast orbit and clock errors. Two strategies are herein introduced, respectively based on Leave-One-Out cross-validation (VADASE-LOO) for a receiver autonomous outlier detection, and on a network augmentation strategy to filter common trends out (A-VADASE); they are combined (first, VADASE-LOO; second, A-VADASE) for a complete solution. Moreover, starting fromthis VADASE improved solution, an additional strategy is proposed to estimate in real time the overall coseismic displacement occurring at each GNSS receiver. New VADASE advances are successfully applied to the GPS data collected during the recent three strong earthquakes that occurred in Central Italy on 24 August and 26 and 30 October 2016, and the results are herein presented and discussed. The VADASE real-time estimated coseismic displacements are compared to the static ones derived from the daily solutions obtained within the standard post-processing procedure by the Istituto Nazionale di Geofisica e Vulcanologia
