    Video Game Cultivation: Sowing the Seeds of Consumer Behavior

    George Gerbner (1998) dedicated a significant amount of this life attempting to understand the creation and impact of mass media messages on viewers. Out of this research sprang cultivation theory, which holds viewers\u27 realities can be impacted over time by the media environment they inhabit and its overarching messages. While often applied to television, cultivation theory has largely ignored other mediums, specifically video games. Video games have evolved as a medium since the public arcades of the 1980\u27s and now run on high-powered, internet-enabled consoles. These consoles allow producers into the home of the consumer, saturating them with messages. This thesis advocates video game producer messages are encoded in such a manner as to encourage the consumption of video game content and instill in the consumer a reality constructed by the producer. To interrogate these messages, I conduct a case study of a video game developer, Tacit Games (a pseudonym), and examine how the producer attempts to cultivate consumption in the consumer through approaches like downloadable content and framing the video game experience for the video game consumer. Specifically, I examine messages surrounding the developer\u27s franchise, City Mayhem (also a pseudonym). To do this, I conduct qualitative interviews with five senior employees of the company to ascertain what messages are produced for the consumer in relation to the goals of this thesis. The data collected from these interviews is analyzed through the lens of cultural Marxism, which entails the examination of the limitations and pressures exerted on the formation of culture

    Clues to Neuro-Degeneration in Niemann-Pick Type C Disease from Global Gene Expression Profiling

    BACKGROUND: Niemann-Pick Type C (NPC) disease is a neurodegenerative disease that is characterized by the accumulation of cholesterol and glycosphingolipids in the late endocytic pathway. The majority of NPC cases are due to mutations in the NPC1 gene. The precise function of this gene is not yet known. METHODOLOGY/PRINCIPAL FINDINGS: Using cDNA microarrays, we analyzed the genome-wide expression patterns of human fibroblasts homozygous for the I1061T NPC1 mutation that is characterized by a severe defect in the intracellular processing of low density lipoprotein-derived cholesterol. A distinct gene expression profile was identified in NPC fibroblasts from different individuals when compared with fibroblasts isolated from normal subjects. As expected, NPC1 mutant cells displayed an inappropriate homeostatic response to accumulated intracellular cholesterol. In addition, a number of striking parallels were observed between NPC disease and Alzheimer's disease. CONCLUSIONS/SIGNIFICANCE: Many genes involved in the trafficking and processing of amyloid precursor protein and the microtubule binding protein, tau, were more highly expressed. Numerous genes important for membrane traffic and the cellular regulation of calcium, metals and other ions were upregulated. Finally, NPC fibroblasts exhibited a gene expression profile indicative of oxidative stress. These changes are likely contributors to the pathophysiology of Niemann-Pick Type C disease

    Dementia Revealed: Novel Chromosome 6 Locus for Late-Onset Alzheimer Disease Provides Genetic Evidence for Folate-Pathway Abnormalities

    Genome-wide association studies (GWAS) of late-onset Alzheimer disease (LOAD) have consistently observed strong evidence of association with polymorphisms in APOE. However, until recently, variants at few other loci with statistically significant associations have replicated across studies. The present study combines data on 483,399 single nucleotide polymorphisms (SNPs) from a previously reported GWAS of 492 LOAD cases and 496 controls and from an independent set of 439 LOAD cases and 608 controls to strengthen power to identify novel genetic association signals. Associations exceeding the experiment-wide significance threshold () were replicated in an additional 1,338 cases and 2,003 controls. As expected, these analyses unequivocally confirmed APOE's risk effect (rs2075650, ). Additionally, the SNP rs11754661 at 151.2 Mb of chromosome 6q25.1 in the gene MTHFD1L (which encodes the methylenetetrahydrofolate dehydrogenase (NADP+ dependent) 1-like protein) was significantly associated with LOAD (; Bonferroni-corrected P = 0.022). Subsequent genotyping of SNPs in high linkage disequilibrium () with rs11754661 identified statistically significant associations in multiple SNPs (rs803424, P = 0.016; rs2073067, P = 0.03; rs2072064, P = 0.035), reducing the likelihood of association due to genotyping error. In the replication case-control set, we observed an association of rs11754661 in the same direction as the previous association at P = 0.002 ( in combined analysis of discovery and replication sets), with associations of similar statistical significance at several adjacent SNPs (rs17349743, P = 0.005; rs803422, P = 0.004). In summary, we observed and replicated a novel statistically significant association in MTHFD1L, a gene involved in the tetrahydrofolate synthesis pathway. This finding is noteworthy, as MTHFD1L may play a role in the generation of methionine from homocysteine and influence homocysteine-related pathways and as levels of homocysteine are a significant risk factor for LOAD development

    Scalable synthesis of an architectural library of well-defined poly(acrylic acid) derivatives: role of structure on dispersant performance

    The synthesis and systematic comparison of a comprehensive library of well-defined polymer architectures based on poly(acrylic acid) is reported. Through the development of new synthetic methodologies, linear, single branched, precision-branched comb, and star polymers were prepared and their performance as dispersants was evaluated. The ability to accurately control chain lengths and branch points allows the subtle interplay between structure and dispersant performance to be defined and affords critical insights into the design of improved polymeric additives for coating formulations. The general industrial relevance of ionic polymers and branched macromolecular architectures supports these design rules for a wide range of other applications and materials, including as additives for personal care products and in water treatment.
