    Changes in zebrafish (Danio rerio) lens crystallin content during development.

    PurposeThe roles that crystallin proteins play during lens development are not well understood. Similarities in the adult crystallin composition of mammalian and zebrafish lenses have made the latter a valuable model for examining lens function. In this study, we describe the changing zebrafish lens proteome during development to identify ontogenetic shifts in crystallin expression that may provide insights into age-specific functions.MethodsTwo-dimensional gel electrophoresis and size exclusion chromatography were used to characterize the lens crystallin content of 4.5-day to 27-month-old zebrafish. Protein spots were identified with mass spectrometry and comparisons with previously published proteomic maps, and quantified with densitometry. Constituents of size exclusion chromatography elution peaks were identified with sodium dodecyl sulfate-polyacrylamide gel electrophoresis.ResultsZebrafish lens crystallins were expressed in three ontogenetic patterns, with some crystallins produced at relatively constant levels throughout development, others expressed primarily before 10 weeks of age (βB1-, βA1-, and γN2-crystallins), and a third group primarily after 10 weeks (α-, βB3-, and γS-crystallins). Alpha-crystallins comprised less than 1% of total lens protein in 4.5-day lenses and increased to less than 7% in adult lenses. The developmental period between 6 weeks and 4 months contained the most dramatic shifts in lens crystallin expression.ConclusionsThese data provide the first two-dimensional gel electrophoresis maps of the developing zebrafish lens, with quantification of changing crystallin abundance and visualization of post-translational modification. Results suggest that some crystallins may play stage specific roles during lens development. The low levels of zebrafish lens α-crystallin relative to mammals may be due to the high concentrations of γ-crystallins in this aquatic lens. Similarities with mammalian crystallin expression continue to support the use of the zebrafish as a model for lens crystallin function

    Comparison between RF and electrical signals from the partial discharge activity of twisted pair cables at reduced pressures

    A pressure-controlled test facility has been set up that allows the PD behaviour of polymer insulated twisted pair samples exposed to 50 Hz AC voltages in the range of 0 to 10 kV to be characterised. Resulting PD activity is quantified using the methods defined in IEC standard 60270 and by using a simple monopole antenna to detect the RF signals excited inside the pressure vessel by the discharges. This paper gives the results of preliminary tests performed on samples of wire insulated with Ethylenetetraflourethylene, Silicon Rubber and Polyvinylchloride in the pressure range between 103 and 105 Pa in atmospheric air. The dependence of PD inception voltage on the environmental pressure is reported. Changes in the behaviour of the PD activity; the correlations between the RF and electrical measurements and the frequency components of the RF signals as the applied voltage and pressure are varied are characterised and discussed

    This paper investigates the determinants of firms’ external skill shortages – that is, vacancies that are hard to fill for skill­related reasons. This paper utilises a specially­designed survey, the Business Strategy and Skills (BSS) module of the Business Operations Survey 2008 (BOS 2008). We estimate the determinants of firms reporting having vacancies and skill shortage in comparison to the non­skill­related vacancies using probit models and two­stage (Heckman) probit models with selection, to enable us to distinguish between those factors related with the firm posting a vacancy and those that make them difficult to fill. We consider a broad suite of variables, including firm’s size and industry, their market focus, R&D investment, innovation, previous performance (e.g. productivity), the degree of competition they are subject to

    3D-XY critical fluctuations of the thermal expansivity in detwinned YBa2Cu3O7-d single crystals near optimal doping

    The strong coupling of superconductivity to the orthorhombic distortion in YBa2Cu3O7-d makes possible an analysis of the superconducting fluctuations without the necessity of subtracting any background. The present high-resolution capacitance dilatometry data unambiguously demonstrate the existence of critical, instead of Gaussian, fluctuations over a wide temperature region (+/- 10 K) around Tc. The values of the amplitude ratio A+/A-=0.9-1.1 and the leading scaling exponent |alpha|<0.018, determined via a least-squares fit of the data, are consistent with the 3D-XY universality class.     Design and Construction of a Psychrometric Coil Testing Facility for Commercial Size Heat Exchanger Coils

    Heat exchanger coils are used to provide heating and cooling in HVAC&R equipment. Testing of coils is necessary to evaluate how design changes, such as refrigerant distributor, fin type, and circuitry, affect heat exchanger coil performance. A psychrometric coil testing facility has been designed to facilitate performance testing of commercial-sized coils at Oklahoma State University. The current status of the psychrometric coil testing facility will be discussed. The closed loop facility will contain various equipment to condition the air including heating/cooling coils, electric heaters, and a steam injection system. Heat exchanger coils of up to 20 nominal tons of cooling capacity can be tested in a 7 ft wide by 8 ft tall test section that can supply a maximum air flow rate of 8,000 CFM. With the initial chiller configuration, the facility will be able to operate over an air temperature range of 0°F (-17.8°C) to 140°F (60°C). It is planned to extend the operating envelope to -40°F (-40°C) with a high capacity low temperature chiller. The design phase of the structure is complete and the facility is currently under construction, while the coil conditioning loops are in the design phase. The design is compliant with multiple ASHRAE standards, including Standards 37 and 41.2 which aided in designing the airflow measurement apparatus and louvered air mixers, respectively. Future work includes completion of construction, selection of instrumentation, and development of a LabVIEW controls interface to be integrated upon tunnel completion

    Integration and instrument characterization of the cosmic infrared background experiment 2 (CIBER-2)

    The extragalactic background light (EBL) is the integrated emission from all objects outside of the Milky Way galaxy. Imprinted by the history of stellar emission, the EBL in the near infrared traces light back to the birth of the first stars in the Universe and can allow tight constraints on structure formation models. Recent studies using data from the Spitzer Space Telescope and the first Cosmic Infrared Background ExpeRiment (CIBER-1) find that there are excess fluctuations in the EBL on large scales which have been attributed to either high redshift galaxies and quasars, or to stars that were stripped from their host galaxies during merging events. To help disentangle these two models, multi-wavelength data can be used to trace their distinctive spectral features. Following the success of CIBER-1, CIBER-2 is designed to identify the sources of the EBL excess fluctuations using data in six wavebands covering the optical and near infrared. The experiment consists of a cryogenic payload and is scheduled to launch four times on a recoverable sounding rocket. CIBER-2 has a 28.5 cm telescope coupled with an optics system to obtain wide-field images in six broad spectral bands between 0.5 and 2.5 μm simultaneously. The experiment uses 2048 × 2048 HAWAII-2RG detector arrays and a cryogenic star tracker. A prototype of the cryogenic star tracker is under construction for a separate launch to verify its performance and star tracking algorithm. The mechanical, optical, and electrical components of the CIBER-2 experiment will have been integrated into the payload by mid-2018. Here we present the final design of CIBER-2 and our team’s instrument characterization efforts. The design and analysis of the optical focus tests will be discussed. We also report on the performance of CIBER-2 support systems, including the cooling mechanisms and deployable components. Finally, we outline the remaining tasks required to prepare the payload for launch
