    Dimethyl sulfoxide-respiring bacteria in Suribati Ike, a hypersaline lake, in Antarctica and the marine environment

    Dimethyl sulfoxide (DMSO) occurs worldwide, especially in marine environments as well as in lakes and rainwater. DMSO respiration by bacteria is assumed to play an important role in the sulfur cycle in Antarctica and on earth. We first studied whether DMSO-respiring bacteria existed in Antarctica. Eight strains were isolated that grew by DMSO respiration under anaerobic conditions from water of the halocline in a meromictic lake, Suribati Ike, near Syowa Station in Antarctica. All of them were related to known species belonging to the genus Marinobacter based on 16S rRNA gene sequences. Using a clone library analysis of 16S rRNA gene sequences, 38 of total 48 clones from water of the halocline were identified as Marinobacter. Studies on the various anaerobic respiration capabilities by bacteria in the halocline water found only DMSO respiration. Studies on bacteria with anaerobic respiration abilities in seawater from the Pacific Ocean and Seto Inland Sea, showed that either DMSO-respiring or nitrate-respiring bacteria were present and that all of isolates capable of DMSO respiration were closely related to Vibrio species

    Signet-Ring Cell-Like Amphicrine Cell Carcinoma of the Stomach

    A poorly differentiated adenocarcinoma containing diverse components, tubular, signet-ring cell and neuroendocrine elements, in a 79-year-old man is described. Endoscopy examination revealed a Borrmann type IV tumor in the antrum. Histologically, scattered foci of tubular and signet-ring cells and a greater number of neuroendocrine elements were observed. The tumor cells were positive for chromogranin A, neuron specific enolase (NSE) and carcinoembryonic antigen (CEA). Furthermore, about one third of the tumor cells were demonstrated as amphicrine differentiation by double staining with chromogranin A and periodic acid-Schiff (PAS). Amphicrine cell carcinomas with exocrine and endocrine differentiation in the stomach is rare and poor prognosis. The present case suggests that amphicrine cell carcinomas can occur in signet-ring cell morphogenesis, and that a multipotential stem cell is an origin of this unusual variant of neuroendocrine carcinoma

    Computed tomography (CT) has been widely used for long. In recently years, the useful application of CT would be the method of the reconstruction images. Historically speaking, reconstruction images were conducted so far by algebraic reconstruction technique (ART), filtered back projection (FBP), iterative reconstruction (IR) algorithms and so on. There are various effects of reconstruction image of CT in clinical practice. Two cases with such beneficial experiences were reported. Case 1 is 74-year-man with hypertension. He was diagnosed to have abdominal aortic aneurysm (AAA). By reconstruction images, the general shape and details were clearly observed including regulating background data. Case 2 is 48-year-female without history of low back pain. When she received abdominal CT, she was pointed out to have lumbar disc herniation between L4 and L5. The diagnosis was done from the lateral tomography of lumbar vertebrae. By reconstruction images with colored modification, the detail status of herniation was clearly observed. Consequently, reconstruction image method of CT would be useful and effective. Further evolution of CT apparatus and development of computer analysis would be expected for better clinical practice and research in the future

    Brown Adipose Tissue in Amyotrophic Lateral Sclerosis

    The prevalence of brown adipose tissue (BAT) in the periadrenal adipose tissue was studied in 20 autopsy cases of amyotrophic lateral sclerosis (ALS). In 19 of 20 cases, the periadrenal BAT in focal or diffuse distribution was observed. Histometry of BAT revealed average occupancy rates of 21.6% and 4.2% in ALS and age-matched control cases, respectively (p<0.01). Since emaciation and chronic hypoxia in those ALS patients were most likely related to severe muscle wasting and respiratory involvement, it is speculated that an increase in BAT occupancy rates may be secondary to altered thermogenesis which was augmented by chronic hypoxia

    AbstractTespa1 has been recently reported to be a critical molecule in T-cell development, however, the precise molecular mechanisms of Tespa1 remain elusive. Here, we demonstrate that Tespa1 shows amino-acid sequence homology to KRAS-induced actin-interacting protein (KRAP), an inositol 1,4,5-trisphosphate receptor (IP3R) binding protein, and that Tespa1 physically associates with IP3R in T and B lymphocytes. Two-consecutive phenylalanine residues (Phe185/Phe186) in Tespa1, which are conserved between Tespa1 and KRAP, are indispensable for the association between Tespa1 and IP3R. These findings suggest that Tespa1 plays critical roles in the immune system through the regulation of the IP3R

    SummaryObjectiveThe purpose of our study was to determine what variables were associated with ventricular fibrillation (VF) induced during electrophysiological stimulation test in patients without apparent organic heart disease.MethodsOur study evaluated 77 patients (51±15 years) who underwent electrophysiological stimulation test, signal averaging, and Na+ channel-blocker challenge test (pilsicainide test). The subjects were divided into two groups, the Brugada group and non-Brugada group. Further, the patients were divided into three subgroups on the base of symptoms (8, 7 symptomatic; 9, 13 syncope; 28, 12 asymptomatic group; in the Brugada and non-Brugada groups, respectively). Multivariate analyses evaluated the association between baseline clinical factors and the induction of VF.ResultsThe inducibility of VF was significantly (p<0.0001) higher in the Brugada group (n=33, 73%) than the non-Brugada group (n=4, 13%). The multivariate analysis demonstrated that symptoms (odds ratio (OR) 31.6; 95% confidence interval (CI): 2.3–430.6; p<0.01), type 1 electrocardiogram after pilsicainide test (OR 21.3; CI: 1.7–272.2; p<0.02), and syncope (OR 13.5; CI: 1.2–158.8; p<0.05) were strongly associated with the inducibility of VF, but not with family history, type 1 electrocardiogram in control, positive in late potential, maxΔST elevation (≧200μV) after pilsicainide test.ConclusionsThe symptoms, syncope, and type 1 electrocardiogram after pilsicainide test were independently associated with the electrophysiological substrate of VF in patients without apparent heart disease

    In Vitro Assessment of Factors Affecting the Apparent Diffusion Coefficient of Ramos Cells Using Bio-phantoms

    The roles of cell density, extracellular space, intracellular factors, and apoptosis induced by the molecularly targeted drug rituximab on the apparent diffusion coefficient (ADC) values were investigated using bio-phantoms. In these bio-phantoms, Ramos cells (a human Burkittセs lymphoma cell line) were encapsulated in gellan gum. The ADC values decreased linearly with the increase in cell density, and declined steeply when the extracellular space became less than 4 μm. The analysis of ADC values after destruction of the cellular membrane by sonication indicated that approximately 65% of the ADC values of normal cells originate from the cell structures made of membranes and that the remaining 35% originate from intracellular components. Microparticles, defined as particles smaller than the normal cells, increased in number after rituximab treatments, migrated to the extracellular space and significantly decreased the ADC values of bio-phantoms during apoptosis. An in vitro study using bio-phantoms was conducted to quantitatively clarify the roles of cellular factors and of extracellular space in determining the ADC values yielded by tumor cells and the mechanism by which apoptosis changes those values
