    Induction of expressions of HLA class I and class II antigens by interferon on human renal cell carcinoma or bladder cancer cell lines

    Interferon-induced expressions of HLA class I and class II (DR, DP, DQ) antigens on human renal cell carcinoma (RCC) cell lines and bladder cancer cell lines were analyzed by flowcytometry. Class I antigen was present on all six different RCC cell lines and the expression was enhanced with IFN-γ treatment except one cell line, Caki-2. Although no RCC cell lines expressed class II antigens without IFN-γ treatment, the treatment induced the expression of DR antigen in all cell lines except Caki-2. DP antigen was induced on KPK-1 and KPK-13 with exposure to IFN-γ. On the other hand, bladder cancer cell lines studied showed surface expression of class I antigen and enhanced expression of class I antigen with IFN-γ treatment. However, IFN-γ didn't enhance the expression of class II antigens on any of those bladder cancer cell lines. The minimum concentrations of IFN-γ to increase the expression of class I and DR andtigens on both KPK-13 and ACHN were 4 IU/ml and 100 IU/ml, respectively. The incubation time with IFN-γ to get the maximal expression of class I and DR antigens on KPK-13 were 36 hr and 48 hr, respectively. Meanwhile, although the expression of class I antigen on RCC cell lines was enhanced by IFN-α, IFN-α could not give any effect in class II antigens. Our results demonstrate the obvious effect of IFN-γ to enhance the expression of not only HLA class I antigen but also class II antigens on RCC cell lines

    Identification of novel canonical strigolactones produced by tomato

    Canonical strigolactones (SLs), such as orobanchol, consist of a tricyclic lactone ring (ABC-ring) connected to a methylbutenolide (D-ring). Tomato plants have been reported to produce not only orobanchol but also various canonical SLs related to the orobanchol structure, including orobanchyl acetate, 7-hydroxyorobanchol isomers, 7-oxoorobanchol, and solanacol. In addition to these, structurally unidentified SL-like compounds known as didehydroorobanchol isomers (DDHs), whose molecular mass is 2 Da smaller than that of orobanchol, have been found. Although the SL biosynthetic pathway in tomato is partially characterized, structural elucidation of DDHs is required for a better understanding of the entire biosynthetic pathway. In this study, three novel canonical SLs with the same molecular mass as DDHs were identified in tomato root exudates. The first was 6,7-didehydroorobanchol, while the other two were not in the DDH category. These two SLs were designated phelipanchol and epiphelipanchol because they induced the germination of Phelipanche ramosa, a noxious root parasitic weed of tomato. We also proposed a putative biosynthetic pathway incorporating these novel SLs from orobanchol to solanacol

    Long-term monitoring of sediment runoff for an active sediment control in Joganji River

    There were huge sediment yielding and deposition due to debris flows by breaking natural landslide dams which were formed by earthquake in 1858 at upstream reach of Joganji River. Sediment transportation is still active by debris flow and flow with bedload due to rainfall, though a lot of erosion control dams have been constructed. Continuously measuring sediment runoff for long term along a main river is necessary to evaluate the propagation of sediment after the huge events for sediment management in the basin using well hydrological information. Appropriate tools are selected and applied to monitoring in the area managed by Tateyama Mountain Area Sabo Office along Joganji River, using a Reid-type bedload slot sampler, robust-type hydrophone and velocity meter on the bed for bedload and turbidity meter for washload. Monitored data is concentratedly collected at the office to apply risk management for sediment movement due to heavy rainfall and so on. Several typical data and problems to solved were shown because it passed around twenty years since sediment monitoring started, and those are reported in present study

    THYROID DYSFUNCTION FOLLOWING ALPHA-INTERFERON TREATMENT FOR CHRONIC HEPATITIS C

    In order to evaluate the influnces of IFNα on thyroid function, thyroid-stimulating hormone (TSH), total thyroxine (T4), free T4, tri-iodothyronine (T3), and thyroxine-binding globulin were examined in IFNα-treated 351 patients with chronic hepatitis C before and during therapy. As therapy, either 3 million units (MU) of human lymphoblastoid IFNα or 9MU of recombinant IFNα2a was administrated daily for the initial two weeks followed by three times a week for 22 weeks. There were nine patients showing thyroid dysfunction during IFNα therapy. They consist of one relapse of Graves' disease, one relapse of Hashimoto thyroiditis, one development of apparent thyroid insufficiency from subclinical hypothyroidism, five cases with transient hyperthyroidism and one case with transient hypothyroidism. T4 and T3 levels in most patients who transiently developed thyroid dysfunction were normalized spontaneously after the discontinuation of IFNα. Thyroid-related autoantibodies were positive in 4 patients before IFNα therapy and newly developed in one patient during therapy. Attention should be paid first to the previous histories of autoimmune thyroid diseases and the existence of thyroid-related autoantibodies for the prediction of development of thyroid dysfunction during IFNα therapy. In addition, serial examinations of TSH, T3 and T4 should be also necessary for early detection of transient thyroid dysfunction during IFNα therapy

    リポジトリ用データ管理システムの構築

    金沢大学総合メディア基盤センター近年のオープンサイエンスの活発化に伴い, 人間と機械の双方に可読性を持つリポジトリシステムの需要が高まっている. このような背景から, 本研究では, 国立情報学研究所が開発したWEKO を用いて, 金沢大学環日本海域環境研究センター及び同国際文化資源学研究センターのデータリポジトリの構築を行っている.従来, WEKO へのコンテンツの一括登録には, Windows でのみ動作するソフトウェアが必要であった. 我々は, ICT に精通しているとは限らないデータ所有者が, リポジトリ上のデータ管理を行えるように, OS に非依存なブラウザからファイルをアップロードするだけでコンテンツの一括登録に加え, メタデータやコンテンツの追加登録・更新等が可能なデータ管理システムを構築した. 特に更新機能では, 「更新フラグ」属性を用いて, コンテンツの世代管理とその公開方法の制御を可能にした. 本稿では両センターのデータリポジトリと構築したデータ管理システムの概要を述べる.Along with the recent activation of open science, the demand for a repository system that is readable to both humans and machines is increasing. In this context, we constructed data repositories for Institute of Nature and Environmental Technology and Center for Cultural Resource Studies in Kanazawa University using WEKO, which is a repository platform developed by the National Institute of Informatics. In the original design of the WEKO, it is necessary to utilize software for Windows for batch registration of data contents. In the present study, we developed a new user interface for data management for data owners who are not familiar with ICT. By using this system, it is possible for data owners to operate a batch processing for additional registration, update of the registered contents and modication of the metadata simply by uploading a le via browser without depending on the OS. Particularly, it is innovative that out software supports revision control and data access control of the contents by setting "update ag"attribute.
