    Optimal distribution of phosphorus compounds in multi-layered natural fabric reinforced biocomposites

    Flame retardancy and mechanical performance of multi-layered biocomposites, consisting of polylactic acid (PLA) matrix films and plain-woven flax fabrics as reinforcement, were investigated. Full factorial design (32) was applied to evaluate the effects of the distribution of P and N containing compounds between the matrix and the fibrous carrier. Composition property correlations of the composite constituents (i.e. flax fabrics treated in aqueous solutions of diammonium phosphate and urea with differing ratio and concentrations and matrix films with 0 to 20 wt% ammonium polyphosphate based intumescent flame retardant content) were determined by thermogravimetric analyses and open flame tests. Positive interaction between the composite constituents was revealed for green composites consisting of various combinations of treated fabrics and intumescent PLA systems. The biocomposites flame retarded with a combined approach, i.e. with a balanced distribution of P containing additives between the phases, were found to gain improved mechanical performance and fire retardancy. It was confirmed by tensile testing and electron microscopy as well as by UL-94, limiting oxygen index and cone calorimeter tests. As a conclusion, interpretation is given for the optimum found

    Nanofibrous solid dosage form of living bacteria prepared by electrospinning

    The aim of this work was to investigate the suitability of electrospinning for biodrug delivery and to develop an electrospinning-based method to produce vaginal drug delivery systems. Lactobacillus acidophilus bacteria were encapsulated into nanofibers of three different polymers (polyvinyl alcohol and polyvinylpyrrolidone with two different molar masses). Shelf life of the bacteria could be enhanced by the exclusion of water and by preparing a solid dosage form, which is an advantageous and patient-friendly way of administration. The formulations were stored at –20, 7 and 25°C, respectively. Viability testing showed that the nanofibers can provide long term stability for huge amounts of living bacteria if they are kept at (or below) 7°C. Furthermore, all kinds of nanowebs prepared in this work dissolved instantly when they got in contact with water, thus the developed biohybrid nanowebs can provide new potential ways for curing bacterial vaginosis

    Research background. Protein A affinity chromatography is a well-established method currently used in the pharmaceutical industry. However, the high costs usually associated with chromatographic separation of protein A and the difficulties in continuous operation make the investigation of alternative purification methods very important. Experimental approach. In this study, extraction/back-extraction and precipitation/dissolution methods were developed and optimised. They were compared with protein A and cation exchange chromatography separations in terms of yield of monoclonal antibody (mAb) and amount of residual impurities, such as DNA and host cell proteins, and amount of mAb aggregates. For a comprehensive comparison of the different methods, experiments were carried out with the same cell-free fermentation broth containing adalimumab. Results and conclusions. Protein A and cation exchange chromatographic separations resulted in high yield and purity of adalimumab. The precipitation-based process resulted in high yield but with lower purity. The extraction-based purification resulted in low yield and purity. Thus, the precipitation-based method proved to be more promising than the extraction-based method for direct purification of adalimumab from harvested cell culture fluid. Novelty and scientific contribution. Although alternative purification methods may offer the advantages of simplicity and low-cost operation, further significant improvements are required to compete with the performance of chromatographic separations of adalimumab from true fermentation broth.Pozadina istraživanja. Afinitetna kromatografija pri kojoj se monoklonsko protutijelo veže za protein A je već dobro poznata metoda u farmaceutskoj industriji. Međutim, zbog visokih troškova izdvajanja proteina A pomoću kromatografije te poteškoća u provođenju kontinuiranog procesa važno je ispitati alternativne metode pročišćavanja. Eksperimentalni pristup. U ovome su radu razvijene i optimirane metode ekstrakcije sa ponovnom ekstrakcijom te taloženja i otapanja. Rezultati tih dvaju metoda, odnosno prinos monoklonskih protutijela, količina preostalih nečistoća, kao što su DNA i proteini stanica domaćina, te količina agregata monoklonskih protutijela, uspoređeni su s onima dobivenim afinitetnom kromatografijom na ionskom izmjenjivaču. Za sveobuhvatnu usporedbu različitih metoda, ispitivanja su provedena na identičnim hranjivim podlogama koje su sadržavale adalimumab. Rezultati i zaključci. Afinitetnom kromatografijom na ionskom izmjenjivaču dobiveni su veliki prinos i velika čistoća adalimumaba. Taloženjem su dobiveni veliki prinos, ali manja čistoća uzorka. Pročišćavanje pomoću ekstrakcije rezultiralo je manjim prinosom i manjom čistoćom uzorka. Zaključeno je da je metoda taloženja bolja od ekstrakcije za izravno pročišćavanje adalimumaba iz suspenzije stanične kulture. Novina i znanstveni doprinos. Iako alternativne metode imaju neke prednosti, kao što su jednostavna i jeftina primjena, neophodno ih je unaprijediti da bi dobiveni rezultati bili usporedivi s rezultatima dobivenim pročišćavanjem adalimumaba ionskom kromatografijom u hranjivoj podlozi

    ENABLING GENERIC DISTRIBUTED COMPUTING INFRASTRUCTURE COMPATIBILITY FOR WORKFLOW MANAGEMENT SYSTEMS

    Solving workflow management system’s Distributed Computing Infrastructure (DCI) incompatibility and their workflow interoperability issues are very challenging and complex tasks. Workflow management systems (and therefore their workflows, workflow developers and also their end-users) are bounded tightly to some limited number of supported DCIs, and efforts required to allow additional DCI support. In this paper we are specifying a concept how to enable generic DCI compatibility for grid workflow management systems (such as ASKALON, MOTEUR, gUSE/WS-PGRADE, etc.) on job and indirectly on workflow level. To enable DCI compatibility among the different workflow management systems we have developed the DCI Bridge software solution. In this paper we will describe its internal architecture, provide usage scenarios to show how the developed service resolve the DCI interoperability issues between various middleware types. The generic DCI Bridge service enables the execution of jobs onto the existing major DCI platforms (such as Service Grids (Globus Toolkit 2 and 4, gLite, ARC, UNICORE), Desktop Grids, Web services, or even cloud based DCIs)

    Characterisation of natural fibre reinforced PLA foams prepared by supercritical CO2 assisted extrusion

    Natural fibre reinforced polylactic acid (PLA) foams, as potential green replacements for petroleum-based polymer foams, were investigated. Highly porous (ε > 95%) microcellular PLA foams were manufactured by supercritical CO2 assisted extrusion process. To overcome the inherently low melt strength of PLA, epoxy-functionalized chain extender was applied, while talc was added to improve its crystallization kinetics. The combined application of chain extender and talc effectively promoted the formation of uniform cell structures. The effect of cellulose and basalt fibre reinforcement on the foamability, morphology, structure and mechanical properties of the PLA foams were investigated as well. The addition of 5 wt% natural fibres promoted the cell nucleation, but caused non-uniform distribution of cell size due to the microholes induced by local fibre-matrix debonding. The compression strength of the manufactured basalt fibre reinforced PLA foams reached 40 kPa

    Long-term exercise treatment reduces oxidative stress in the hippocampus of aging rats.

    Exercise can exert beneficial effects on cognitive functions of older subjects and it can also play an important role in the prevention of neurodegenerative diseases. At the same time it is perceivable that limited information is available on the nature of molecular pathways supporting the antioxidant effects of exercise in the brain. In this study 12-month old, middle-aged female Wistar rats were subjected to daily moderate intensity exercise on a rodent treadmill for a period of 15weeks which covered the early aging period unmasking already some aging-related molecular disturbances. The levels of reactive oxygen species (ROS), the amount of protein carbonyls, the levels of antioxidant intracellular enzymes superoxide dismutases (SOD-1, SOD-2) and glutathione peroxidase (GPx) were determined in the hippocampus. In addition, to identify the molecular pathways that may be involved in ROS metabolism and mitochondrial biogenesis, the activation of 5'-AMP-activated protein kinase (AMPK), the protein level of peroxisome proliferator-activated receptor gamma coactivator 1-alpha (PGC-1α), nuclear respiratory factor 1 (NRF-1) and mitochondrial transcription factor A (mtTFA) were measured. Our results revealed a lower level of ROS associated with a reduced amount of protein carbonyls in the hippocampus of physically trained rats compared to sedentary controls. Furthermore, exercise induced an up-regulation of SOD-1 and GPx enzymes, p-AMPK and PGC-1α, that can be related to an improved redox balance in the hippocampus. These results suggest that long-term physical exercise can comprises antioxidant properties and by this way protect neurons against oxidative stress at the early stage of aging

    Electrospinning scale-up and formulation development of PVA nanofibers aiming oral delivery of biopharmaceuticals

    Electrospinning is a promising drying technology providing a rapid and gentle drying at ambient temperature, thus electrospinning of polyvinyl alcohol aqueous solutions was investigated for the solid formulation of biopharmaceuticals. The commonly used single-needle electrospinning does not have adequate productivity to satisfy the industrial requirements, therefore our aim was to study the scale-up of the technology by using high-speed electrospinning. High molecular weight polyethylene oxide as a secondary polymer was applied to enhance the fiber formation of polyvinyl alcohol. While polyvinyl alcohol-polyethylene oxide formulations resulted in adequate fiber formation it was not possible to process them further as the friability of the fibers was too low. In order to increase the friability, the effect of adding various sugars (mannitol, glucose, lactose, saccharose, and trehalose) was investigated. The results showed that mannitol was the best friability enhancing excipient because of its crystallinity and low moisture content in the fibrous sample. In contrast, glucose, lactose, saccharose, and trehalose were amorphous with higher moisture content and fibers containing these were grindable only after post-drying

    Melt‐Blown and Electrospun Drug‐Loaded Polymer Fiber Mats for Dissolution Enhancement: A Comparative Study

    Melt blowing (MB) was investigated to prepare a fast dissolving fibrous drug-loaded solid dispersion and compared with solvent-based electrospinning (SES) and melt electrospinning (MES). As a conventional solvent-free technique coupled with melt extrusion and using a high-speed gas stream, MB can provide high-quality micro- and nanofibers at industrial throughput levels. Carvedilol, a weak-base model drug with poor water solubility, was processed using a common composition optimized for the fiber spinning and blowing methods based on a hydrophilic vinylpyrrolidone-vinyl acetate copolymer (PVPVA64) and PEG 3000 plasticizer. Scanning electron microscopy combined with fiber diameter analysis showed diameter distributions characteristic to each prepared fibrous fabrics (the mean value increased toward SES<MB<MES). Differential scanning calorimetry and X-ray diffraction studies revealed that the incorporated drug was in amorphous form regardless the preparation method. The HPLC studies demonstrated that all of the materials produced by the different techniques passed the regulatory purity requirements. The fibers exhibited ultrafast drug release tested under neutral pH conditions; the melt-blown sample dissolved within 2 min owing to its large specific surface area. The presented results confirm the applicability of MB as a novel formulation technique for polymer-based drug delivery systems
