Microleakage of Three Cement Bases
The purpose of this study was to evaluate the ability of a glass-ionomer cement-base material to prevent bacterial penetration along the dentin interface and to compare it with two conventional cement-base materials. A total of 107 Class 5 restorations was placed in Rhesus monkey teeth by means of three test materials [zinc oxide-eugenol (ZOE), copalite varnish + zinc phosphate cement base (V + ZP), and a glass-ionomer lining cement (GI)], with unetched and unbonded resin composite used alone as a control material and as a final restoration over the test base materials. Following disinfection, Class 5 cavities were prepared on the buccal surfaces of the teeth to the inner one-half of dentin. A sterile filter-paper disk was then placed on the axial wall and covered with a Teflon disk. Next, the cavities were based to the dento-enamel junction with one of the test base materials and finally restored with unetched and unbonded resin composite. After five and 16 weeks, the filter-paper disks were retrieved and cultivated for the presence and type of bacteria. The five-week results showed positive growth in two groups: the composite-only controls and the V + ZP group. The 16-week results showed growth in all of the test groups, but only one of nine teeth showed growth in the zinc oxide-eugenol group and one of 16 teeth in the glass-ionomer group. Photon Shot Noise Limits on Optical Detection of Neuronal Spikes and Estimation of Spike Timing
AbstractOptical approaches for tracking neural dynamics are of widespread interest, but a theoretical framework quantifying the physical limits of these techniques has been lacking. We formulate such a framework by using signal detection and estimation theory to obtain physical bounds on the detection of neural spikes and the estimation of their occurrence times as set by photon counting statistics (shot noise). These bounds are succinctly expressed via a discriminability index that depends on the kinetics of the optical indicator and the relative fluxes of signal and background photons. This approach facilitates quantitative evaluations of different indicators, detector technologies, and data analyses. Our treatment also provides optimal filtering techniques for optical detection of spikes. We compare various types of Ca2+ indicators and show that background photons are a chief impediment to voltage sensing. Thus, voltage indicators that change color in response to membrane depolarization may offer a key advantage over those that change intensity. We also examine fluorescence resonance energy transfer indicators and identify the regimes in which the widely used ratiometric analysis of signals is substantially suboptimal. Overall, by showing how different optical factors interact to affect signal quality, our treatment offers a valuable guide to experimental design and provides measures of confidence to assess optically extracted traces of neural activity
First scattered light images of debris disks around HD 53143 and HD 139664
We present the first scattered light images of debris disks around a K star
(HD 53143) and an F star (HD 139664) using the coronagraphic mode of the
Advanced Camera for Surveys (ACS) aboard the Hubble Space Telescope (HST). With
ages 0.3 - 1 Gyr, these are among the oldest optically detected debris disks.
HD 53143, viewed ~45 degrees from edge-on, does not show radial variation in
disk structure and has width >55 AU. HD 139664 is seen close to edge-on and has
belt-like morphology with a dust peak 83 AU from the star and a distinct outer
boundary at 109 AU. We discuss evidence for significant diversity in the radial
architecture of debris disks that appears unconnected to stellar spectral type
or age. HD 139664 and possibly the solar system belong in a category of narrow
belts 20-30 AU wide. HD 53143 represents a class of wide disk architecture with
characteristic width >50 AU.Comment: 7 pages, 3 figure
Effects of depleting ionic strength on 31P nuclear magnetic resonance spectra of micellar casein during membrane separation and diafiltration of skim milk
peer-reviewedMembrane separation processes used in the concentration and isolation of micellar casein-based milk proteins from skim milk rely on extensive permeation of its soluble serum constituents, especially lactose and minerals. Whereas extensive literature exists on how these processes influence the gross composition of milk proteins, we have little understanding of the effects of such ionic depletion on the core structural unit of micellar casein [i.e., the casein phosphate nanocluster (CPN)]. The 31P nuclear magnetic resonance (NMR) is an analytical technique that is capable of identifying soluble and organic forms of phosphate in milk. Thus, our objective was to investigate changes to the 31P NMR spectra of skim milk during microfiltration (MF) and diafiltration (DF) by tracking movements in different species of phosphate. In particular, we examined the peak at 1.11 ppm corresponding to inorganic phosphate in the serum, as well as the low-intensity broad signal between 1.5 and 3.0 ppm attributed to casein-associated phosphate in the retentate. The MF concentration and DF using water caused a shift in the relevant 31P NMR peak that could be minimized if orthophosphate was added to the DF water. However, this did not resolve the simultaneous change in retentate pH and increased solubilization of micellar casein protein. The addition of calcium in combination with orthophosphate prevented micellar casein solubilization and simultaneously contributed to preservation of the CPN structure, except for overcorrection of retentate pH in the acidic direction. A more complex DF solution, involving a combination of phosphate, calcium, and citrate, succeeded in both CPN and micellar casein structure preservation while maintaining retentate pH in the region of the original milk pH. The combination of 31P NMR as an analytical technique and experimental probe during MF/DF processes provided useful insights into changes occurring to CPN while retaining the micellar state of casein
Asthma exacerbations and worsenings in patients aged 1-75 years with add-on tiotropium treatment
This review explores the effect of tiotropium Respimat® add-on therapy on asthma exacerbations and worsenings, adverse events (AEs) related to exacerbations and symptoms and any effects on seasonality across the 10 UniTinA-asthma® clinical trials comprising over 6000 patients. When added on to inhaled corticosteroids ± additional therapies, tiotropium significantly reduced the risk of exacerbations and worsenings in adults with symptomatic severe asthma and provided a non-significant improvement in worsenings in adults with symptomatic moderate and mild asthma, which was significant for patients with moderate asthma receiving tiotropium 2.5 µg once daily vs. placebo. Trials in paediatric patients were not powered to assess exacerbations or worsenings, but when AEs related to asthma exacerbations and symptoms were grouped into a composite endpoint and pooled, tiotropium improved outcomes vs. placebo (rate ratio 0.76; 95% confidence interval 0.63, 0.93). The reduction in exacerbations with tiotropium is apparent across all patients during the observed seasonal peaks of these events

Innovating Pedagogy 2015: Open University Innovation Report 4
This series of reports explores new forms of teaching, learning and assessment for an interactive world, to guide teachers and policy makers in productive innovation. This fourth report proposes ten innovations that are already in currency but have not yet had a profound influence on education. To produce it, a group of academics at the Institute of Educational Technology in The Open University collaborated with researchers from the Center for Technology in Learning at SRI International. We proposed a long list of new educational terms, theories, and practices. We then pared these down to ten that have the potential to provoke major shifts in educational practice, particularly in post-school education. Lastly, we drew on published and unpublished writings to compile the ten sketches of new pedagogies that might transform education. These are summarised below in an approximate order of immediacy and timescale to widespread implementation
