    Product design and development of an aerodynamic hydration system for bicycling and triathlon

Several aerodynamic systems have been developed for use on bicycles but few have been designed to be truly aerodynamic or easy to use. This project focused on the design and development of a frame mounted hydration system for cycling and triathlon that offered improvements in usability and aerodynamic performance over the current market solutions. A wind tunnel test validated a viable location on to place the fluid reservoir. Product specifications were derived from consumer needs and concepts were generated to fulfill those needs. Several iterations of CAD models of the entire product were made. The resulting product, a top tube mounted reservoir with a drinking tube and tri-leaf check valve for re-filling the reservoir while riding will be prototyped by the author over the summer of 2007.by Mark Cote.S.B

    Who are CUIs Really For? Representation and Accessibility in the Conversational User Interface Literature

    The theme for CUI 2023 is 'designing for inclusive conversation', but who are CUIs really designed for? The field has its roots in computer science, which has a long acknowledged diversity problem. Inspired by studies mapping out the diversity of the CHI and voice assistant literature, we set out to investigate how these issues have (or have not) shaped the CUI literature. To do this we reviewed the 46 full-length research papers that have been published at CUI since its inception in 2019. After detailing the eight papers that engage with accessibility, social interaction, and performance of gender, we show that 90% of papers published at CUI with user studies recruit participants from Europe and North America (or do not specify). To complement existing work in the community towards diversity we discuss the factors that have contributed to the current status quo, and offer some initial suggestions as to how we as a CUI community can continue to improve.     A Systematic Review of Ethical Concerns with Voice Assistants

    Siri's introduction in 2011 marked the beginning of a wave of domestic voice assistant releases, and this technology has since become commonplace in consumer devices such as smartphones and TVs. But as their presence expands there have also been a range of ethical concerns identified around the use of voice assistants, such as the privacy implications of having devices that are always recording and the ways that these devices are integrated into the existing social order of the home. This has created a burgeoning area of research across a range of fields including computer science, social science, and psychology. This paper takes stock of the foundations and frontiers of this work through a systematic literature review of 117 papers on ethical concerns with voice assistants. In addition to analysis of nine specific areas of concern, the review measures the distribution of methods and participant demographics across the literature. We show how some concerns, such as privacy, are operationalized to a much greater extent than others like accessibility, and how study participants are overwhelmingly drawn from a small handful of Western nations. In so doing we hope to provide an outline of the rich tapestry of work around these concerns and highlight areas where current research efforts are lacking

    Extrinsic and intrinsic preanalytical variables affecting liquid biopsy in cancer

    Liquid biopsy, through isolation and analysis of disease-specific analytes, has evolved as a promising tool for safe and minimally invasive diagnosis and monitoring of tumors. It also has tremendous utility as a companion diagnostic allowing detection of biomarkers in a range of cancers (lung, breast, colon, ovarian, brain). However, clinical implementation and validation remains a challenge. Among other stages of development, preanalytical variables are critical in influencing the downstream cellular and molecular analysis of different analytes. Although considerable progress has been made to address these challenges, a comprehensive assessment of the impact on diagnostic parameters and consensus on standardized and optimized protocols is still lacking. Here, we summarize and critically evaluate key variables in the preanalytical stage, including study population selection, choice of biofluid, sample handling and collection, processing, and storage. There is an unmet need to develop and implement comprehensive preanalytical guidelines on the optimal practices and methodologies

    The Liquid Biopsy Consortium: Challenges and opportunities for early cancer detection and monitoring

    The emerging field of liquid biopsy stands at the forefront of novel diagnostic strategies for cancer and other diseases. Liquid biopsy allows minimally invasive molecular characterization of cancers for diagnosis, patient stratification to therapy, and longitudinal monitoring. Liquid biopsy strategies include detection and monitoring of circulating tumor cells, cell-free DNA, and extracellular vesicles. In this review, we address the current understanding and the role of existing liquid-biopsy-based modalities in cancer diagnostics and monitoring. We specifically focus on the technical and clinical challenges associated with liquid biopsy and biomarker development being addressed by the Liquid Biopsy Consortium, established through the National Cancer Institute. The Liquid Biopsy Consortium has developed new methods/assays and validated existing methods/technologies to capture and characterize tumor-derived circulating cargo, as well as addressed existing challenges and provided recommendations for advancing biomarker assays

    Arthroscopic partial meniscectomy versus physical therapy for degenerative meniscus lesions: how robust is the current evidence? A critical systematic review and qualitative synthesis

    The purpose of this systematic review was to investigate study quality and risk of bias for randomized trials comparing partial meniscectomy with physical therapy in middle-aged patients with degenerative meniscus tears.A systematic review of Medline, Embase, Scopus, and Google Scholar was performed from 1990 through 2017. The inclusion criteria were at least 1 validated outcome score, and middle-aged patients (40\ua0years and older) with a degenerative meniscus tear. Studies with a sham arm, and acute and concomitant injuries were excluded. Risk of bias was assessed with the Cochrane Risk of Bias Tool. The quality of studies was assessed with the Cochrane GRADE tool and quality assessment tool (Effective Public Health Practice Project). Publication bias was assessed by funnel plot and Egger's test. The I statistics was calculated a measure of statistical heterogeneity.Six studies were included, and all were assessed as having a high risk of bias. There was no publication bias (P\ua0= .23). All studies were downgraded (low, n\ua0= 5; very low, n\ua0= 1). The Effective Public Health Practice Project assessed 1 study as strong, 2 as moderate, and 3 as weak. The overall results demonstrated moderate to low quality of the included studies. The I statistic was 96.2%, demonstrating substantial heterogeneity between studies.The results of this systematic review strongly suggest that there is currently no compelling evidence to support arthroscopic partial meniscectomy versus physical therapy. The studies evaluated here exhibited a high risk of bias, and the weak to moderate quality of the available studies, the small sample sizes, and the diverse study characteristics do not allow any meaningful conclusions to be drawn. Therefore, the validity of the results and conclusions of prior systematic reviews and meta-analyses must be viewed with extreme caution. The quality of the available published literature is not robust enough at this time to support claims of superiority for either alternative, and both arthroscopic partial meniscectomy or physical therapy could be considered reasonable treatment options for this condition.Level II, systematic review of Level I and II studies

    Automatic detection of circulating tumor cells and cancer associated fibroblasts using deep learning

    Circulating tumor cells (CTCs) and cancer-associated fibroblasts (CAFs) from whole blood are emerging as important biomarkers that potentially aid in cancer diagnosis and prognosis. The microfilter technology provides an efficient capture platform for them but is confounded by two challenges. First, uneven microfilter surfaces makes it hard for commercial scanners to obtain images with all cells in-focus. Second, current analysis is labor-intensive with long turnaround time and user-to-user variability. Here we addressed the first challenge through developing a customized imaging system and data pre-processing algorithms. Utilizing cultured cancer and CAF cells captured by microfilters, we showed that images from our custom system are 99.3% in-focus compared to 89.9% from a top-of-the-line commercial scanner. Then we developed a deep-learning-based method to automatically identify tumor cells serving to mimic CTC (mCTC) and CAFs. Our deep learning method achieved precision and recall of 94% (± 0.2%) and 96% (± 0.2%) for mCTC detection, and 93% (± 1.7%) and 84% (± 3.1%) for CAF detection, significantly better than a conventional computer vision method, whose numbers are 92% (± 0.2%) and 78% (± 0.3%) for mCTC and 58% (± 3.9%) and 56% (± 3.5%) for CAF. Our custom imaging system combined with deep learning cell identification method represents an important advance on CTC and CAF analysis
