Znaczenie prognostyczne VEGF i jego receptorów w raku endometrium typu endometrioidalnego
Abstract Background: Angiogenesis is of crucial importance for endometrial tumor growth and Vascular Endothelial Growth Factor (VEGF) is the key mediator of angiogenesis. Objective: The purpose of our study was to assess the prognostic value of VEGF and its receptors in relation to endometrioid endometrial carcinomas. Material and methods: In this study, we conducted an immunohistochemical evaluation of VEGF and VEGFRs expression in 84 tissue samples obtained from endometrioid endometrial cancer patients undergoing curative surgical treatment. Results: Out of 84 cancers, strong positive expression of VEGF was seen in 35 (42%) tumors. The overall strong positive rates were 33% for VEGFR-1 and for 15% for VEGFR-2. There was a significant correlation between clinical stage and VEGF and VEGFR-1 overexpression (p=0.027 and p=0.004, respectively). A mutant Escherichia coli that attaches peptidoglycan to lipopolysaccharide and displays cell wall on its surface
The lipopolysaccharide (LPS) forms the surface-exposed leaflet of the outer membrane (OM) of Gram-negative bacteria, an organelle that shields the underlying peptidoglycan (PG) cell wall. Both LPS and PG are essential cell envelope components that are synthesized independently and assembled by dedicated transenvelope multiprotein complexes. We have identified a point-mutation in the gene for O-antigen ligase (WaaL) in Escherichia coli that causes LPS to be modified with PG subunits, intersecting these two pathways. Synthesis of the PG-modified LPS (LPS*) requires ready access to the small PG precursor pool but does not weaken cell wall integrity, challenging models of precursor sequestration at PG assembly machinery. LPS* is efficiently transported to the cell surface without impairing OM function. Because LPS* contains the canonical vancomycin binding site, these surface-exposed molecules confer increased vancomycin-resistance by functioning as molecular decoys that titrate the antibiotic away from its intracellular target. Morfologia zatoki wieńcowej - badania z zastosowaniem echokardiografii wewnątrzsercowej
Wstęp: Ujście zatoki wieńcowej (CSo) stanowi podstawę tzw. trójkąta Kocha, w którego
obrębie przebiega droga wolna węzła przedsionkowo-komorowego, będąca celem aplikacji prądu
o częstotliwości radiowej w trakcie ablacji. Celem pracy było porównanie wielkości CSo przy zastosowaniu echokardiografii wewnątrzsercowej (ICE) u pacjentów z AVNRT, chorych
z AVRT oraz u osób bez wywiadu tachyarytmii.
Materiał i metody: Badaniem objęto 14 chorych z AVNRT (w wieku śr. 41,2 ± 11,3 roku)
i 11 pacjentów z AVRT (w wieku śr. 45,8 ± 13,2 roku). Grupę kontrolną stanowiło
16 osób poddanych zabiegowi implantacji układu stymulującego z użyciem ICE (w wieku śr.
57,8 ± 17,8 roku). Pole powierzchni oraz wymiar podłużny CSo określano, stosując ICE.
Wyniki: Odpowiednio u chorych z AVNRT, AVRT i w grupie kontrolnej oceniono: pole powierzchni
CSo (Me: 126,5 vs. 100,5 vs. 104 mm2) i wymiar poprzeczny CSo (śr. ± SD: 20,7 ± 7,3 vs. 16,3 ± 4,4 vs. 17,1 ± 5,2 mm). U pacjentów z AVNRT CSo miało istotnie większe
pole powierzchni niż u osób z AVRT (p = 0,01), a także w porównaniu z pacjentami z grupy
kontrolnej (p = 0,04). Wymiar podłużny CSo był istotnie dłuższy u pacjentów z AVNRT niż
u chorych z AVRT (p = 0,03). Nie stwierdzono korelacji (τKendall) między polem powierzchni CSo
a wiekiem, płcią, powierzchnią ciała oraz maksymalnym odstępem AH (atrium-His).
Wnioski: Pacjenci z AVNRT charakteryzują się większym CSo w porównaniu z osóbami bez

Biogenesis of Shigella flexneri IcsA protein.
The IcsA autotransporter is a vital virulence factor for Shigella flexneri, a humanspecific causative agent of bacillary dysentery that accounts for over a million global deaths annually. Ingested Shigellae invade and spread throughout the colonic epithelium. IcsA confers motility to intracellular bacteria by engaging host actin regulatory proteins to polymerise filaments of actin in a processes termed actin-based motility. This IcsA-dependent motility potentiates the intercellular spreading.
IcsA is displayed at one pole of the bacterium, thereby providing a functional focus for actin polymerisation that generated propulsive force. This work investigated the biogenesis of IcsA, seeking to identify factors that direct the cytoplasmic deliver of the nascent protein towards the pole. A refined polar targeting region, IcsA₅₃₂₋₅₇₀ has been identified. Additionally, insertion mutant, i532 and i563, within the recognised targeting sequence IcsA₅₀₆₋₆₂₀ that have been identified that are defective, though not entirely deficient, in polar targeting. GFP+ fusions to these mutated targeting sequences revealed rapid motion of fluorescent foci throughout the cytoplasm, a process that likely precedes polar targeting in the wild-type. The delivery of the polarly targeted IcsA₅₀₆₋₆₂₀ region was shown to occur contemporaneously with segregating origins of chromosomal replication (oriC) and likely shares a common cell cycle cue.
The diffusive properties of exported IcsA in outer-membrane have also been investigated, addressing whether the protein diffuses in the outer membrane or is masked by LPS. To directly observe the behaviour of IcsA soon after it appears at the cell surface, a strategy exploiting metabolic biotinylation was developed and used to rapidly and specifically label nascent IcsA in the outer membrane. In further investigation of the IcsA-LPS interplay, the profile of polar LPS was shown to be uniform in comparison to the lateral cell body in S. flexneri.

Envelope Stress Responses: An Interconnected Safety Net
The Escherichia coli cell envelope is a protective barrier at the frontline of interaction with the
environment. Fidelity of envelope biogenesis must be monitored to establish and maintain a
contiguous barrier. Indeed, the envelope must also be repaired and modified in response to
environmental assaults. Envelope stress responses (ESRs) sense envelope damage or defects and
alter the transcriptome to mitigate stress. We will review recent insights into stress sensing
mechanisms of the σE and Cpx systems and the interaction of these ESRs. Small RNAs (sRNAs)
are increasingly prominent regulators of the transcriptional response to stress. These fast-acting
regulators also provide avenues for inter-ESR regulation that could be important when cells face
multiple contemporaneous stresses, as is the case during infection

The CpxQ sRNA Negatively Regulates Skp To Prevent Mistargeting of β-Barrel Outer Membrane Proteins into the Cytoplasmic Membrane
The promoter most strongly induced upon activation of the Cpx two-component envelope stress response is the cpxP promoter. The 3′ untranscribed region (UTR) of the cpxP transcript is shown to produce a small RNA (sRNA), CpxQ. We investigated the role of CpxQ in combating envelope stress. Remarkably, the two effectors specified by the transcript are deployed to combat distinct stresses in different cellular compartments. CpxP acts in both a regulatory negative-feedback loop and as an effector that combats periplasmic protein misfolding. We find that CpxQ combats toxicity at the inner membrane (IM) by downregulating the synthesis of the periplasmic chaperone Skp. Our data indicate that this regulation prevents Skp from inserting β-barrel outer membrane proteins (OMPs) into the IM, a lethal event that likely collapses the proton motive force. Our findings suggest that Skp can fold and directly insert OMPs into a lipid bilayer in vivo without the aid of the Bam complex
