    Cloning, expression, and biochemical characterization of β-etherase LigF from Altererythrobacter sp. B11

    Lignin, a complex heteropolymer present in plant cell walls, is now recognized as a valuable renewable resource with potential applications in various industries. The lignin biorefinery concept, which aims to convert lignin into value-added products, has gained significant attention in recent years. β-etherases, enzymes that selectively cleave β-O-4 aryl ether bonds in lignin, have shown promise in lignin depolymerization. In this study, the β-etherase LigF from Altererythrobacter sp. B11 was cloned, expressed, purified, and biochemically characterized. The LigF-AB11 enzyme exhibited optimal activity at 32 °C and pH 8.5 when catalyzing the substrate PNP-AV. The enzyme displayed mesophilic behavior and demonstrated higher activity at moderate temperatures. Stability analysis revealed that LigF-AB11 was not thermostable, with a complete loss of activity at 60 °C within an hour. Moreover, LigF-AB11 exhibited excellent pH stability, retaining over 50 % of its activity after 1 h under pH conditions ranging from 3.0 to 11.0. Metal ions and surface impregnation agents were found to affect the enzyme's activity, highlighting the importance of considering these factors in enzymatic processes for lignin depolymerization. This study provides valuable insights into the biochemical properties of LigF-AB11 and contributes to the development of efficient enzymatic processes for lignin biorefineries. Further optimization and understanding of β-etherases will facilitate their practical application in the valorization of lignin

    Es un texto que posiciona el campo de la Comunicación-Educación a partir de interrogantes críticos, sociales y culturales a los que los investigadores están convocados para el cambio de realidades y producciones de subjetividad en un mundo altamente competitivo e individualizado; esto se enlaza con la herencia colonial y el imaginario eurocéntrico, tan presentes en los modos de representación social. La reivindicación del campo que propone el texto manifiesta la necesidad de interrumpir el exceso del capitalismo con su intromisión biográfica y con la (in)visibilización de lo otro y de los otros. Por esto, un efecto político queda para la escuela y para la configuración de pedagogías insurrectas que logren reconocer saberes de construcción cotidiana, significación de niños y jóvenes con los medios tecnológicos, narrativas feministas y subjetivaciones contrahegemónicas en las que un sujeto insiste por una emancipación decolonial y con una apuesta desde el Sur. Es un buen texto, muy académico y propositivo.It is a text that positions the field of Communication-Education from critical, social and cultural questions to which researchers are summoned for the change of realities and productions of subjectivity in a highly competitive and individualized world; this is linked to the colonial heritage and the Eurocentric imaginary, so present in modes of social representation. The vindication of the field that the text proposes manifests the need to interrupt the excess of capitalism with its biographical interference and with the (in)visibility of the other and of the others. For this reason, a political effect remains for the school and for the configuration of insurgent pedagogies that manage to recognize knowledge of daily construction, meaning of children and young people with technological means, feminist narratives and counter-hegemonic subjectivations in which a subject insists on a decolonial emancipation. and with a bet from - the South.
