    Xanthogranulomatous Cholecystitis Masquerading as Gallbladder Cancer: Can It Be Diagnosed Preoperatively?

    Background. Xanthogranulomatous cholecystitis (XGC) is often misdiagnosed as gallbladder cancer (GBC). We aimed to determine the preoperative characteristics that could potentially aid in an accurate diagnosis of XGC masquerading as GBC. Methods. An analysis of patients operated upon with a preoperative diagnosis of GBC between January 2008 and December 2012 was conducted to determine the clinical and radiological features which could assist in a preoperative diagnosis of XGC. Results. Out of 77 patients who underwent radical cholecystectomy, 16 were reported as XGC on final histopathology (Group A), while 60 were GBC (Group B). The incidences of abdominal pain, cholelithiasis, choledocholithiasis, and acute cholecystitis were significantly higher in Group A, while anorexia and weight loss were higher in Group B. On CT, diffuse gallbladder wall thickening, continuous mucosal line enhancement, and submucosal hypoattenuated nodules were significant findings in Group A. CT findings on retrospect revealed at least one of these findings in 68.7% of the cases. Conclusion. Differentiating XGC from GBC is difficult, and a definitive diagnosis still necessitates a histopathological examination. An accurate preoperative diagnosis requires an integrated review of clinical and characteristic radiological features, the presence of which may help avoid radical resection and avoidable morbidity in selected cases

    Brzi lateks aglutinacijski test za serološki dokaz serotipa 4 ptičjeg adenovirusa upotrebom rekombinantnog antigena

    Hydropericardium hepatitis syndrome (HPS) is a newly emerging disease of poultry, which is caused by fowl adenovirus serotype 4. The virus was propagated in a primary chicken embryo liver culture. The cytopathic effect was observed from the third passage onwards. DNA was isolated from the infected culture and used as a template for amplification of a partial hexon gene (700 bp) using hexon gene specific primers. The amplified product was cloned into pProEX HT b vector and the ligated product was transformed into DH5α cells. The recombinant clones were analyzed by colony PCR and plasmid isolation, followed by restriction digestion to check the insert release. The positive clones were induced by IPTG. The induced culture fractions were checked at different hours and the induction was high at the 4th hour onwards. The expressed proteins were purified and confirmed by using hyperimmune serum against FAV4 by western blot analysis and the protein size of 50kda was obtained. The purified recombinant FAV4 protein was used as a serodiagnostic agent using enzyme linked immunosorbent assay and latex agglutination test.Sindrom hidroperikarda i hepatitisa nova je bolest peradi uzrokovana serotipom 4 ptičjeg adenovirusa. Virus je bio umnožen u primarnoj staničnoj kulturi podrijetlom od jetrenoga tkiva pilećega zametka. Citopatski učinak javio se nakon treće pasaže. DNK je bila izdvojena iz zaražene kulture i rabljena kao kalup za umnožavanje dijela gena heksona (700 bp) uz upotrebu specifičnih početnica. Umnoženi odsječak bio je kloniran u vektoru pProEX HT b, a proizašli proizvod prebačen u DH5α stanice. Rekombinantni klonovi bili su analizirani lančanom reakcijom polimerazom i izdvajanjem plazmida nakon čega je pomoću restrikcijske digestije provjerena uspješnost postupka. Pozitivni klonovi bili su inducirani pomoću IPTG-a. Frakcije induciranih stanica bile su provjeravane svakog sata, a indukcija je bila velika nakon četiri sata. Proizvedene bjelančevine bile su pročišćene i identificirane uporabom hiperimunoga seruma za FAV4 Western blot analizom te se pokazalo da je proizvedena bjelančevina veličine 50 kda. Pročišćena rekombinantna bjelančevina FAV4 bila je rabljena kao antigen u imunoenzimnom testu i lateks aglutinacijskom testu

    Oxidative stress has been identified as an underlying factor in the development of insulin resistance, β-cell dysfunction, impaired glucose tolerance and type 2 diabetes mellitus and it also play major role in kidney stone formation. The present study is aimed to elucidate the in vitro nephroprotective activity of two isoflavonoid glycosides, tectorigenin 7-O-β-D-glucosyl-(1→6)-β-D-glucoside (1) and tectorigenin 7-O-β-D-glucosyl-4'-O-β-D-glucoside (2) isolated from the n-BuOH fraction of Iris spuria L. (Zeal) rhizome MeOH extract against oxalate and high glucose-induced oxidative stress in NRK-49F cells. The results revealed that compounds 1 and 2 significantly increased the antioxidant enzyme activities and decreased MDA levels in both oxalate and high glucose stress. Treatment with these phytochemicals effectively down-regulated expression of crystal modulator genes and pro-fibrotic genes in oxalate and high glucose-mediated stress respectively. This study indicates cytoprotective, antioxidant, anti-urolithic and anti-diabetic effects of compounds 1 and 2 against oxalate and high glucose stress

GRG 2 (KT 235641) against selected extended spectrum beta lactamase (ESBL) producing Pseudomonas aeruginosa (P. aeruginosa) and Klebsiella pneumoniae (K. pneumoniae). Initially, the multi drug resistance (MDR) effect of selected uropathogens was confirmed by respective UTI panel of Hexa antibiotics disc methods. The zone of inhibition ≤22 mm for ceftazidime, ≤ 27 mm for cefotaxime and ≤8 mm zone of MIC stripe against both the uropathogens of phenotypic methods confirmed, the selected strains were ESBL producer. Among the various EA extracts, GRG 2 extract showed excellent antibacterial activity against both ESBL producing P. aeruginosa and K. pneumonia by agar well diffution method.     Molecular characterization and antibacterial effect of endophytic actinomycetes Nocardiopsis sp. GRG1 (KT235640) from brown algae against MDR strains of uropathogens

    Our study is to evaluate the potential bioactive compound of Nocardiopsis sp. GRG1 (KT235640) and its antibacterial activity against multi drug resistant strains (MDRS) on urinary tract infections (UTIs). Two brown algae samples were collected and were subjected to isolation of endophytic actinomycetes. 100 strains of actinomycetes were isolated from algal samples based on observation of morphology and physiological characters. 40 strains were active in antagonistic activity against various clinical pathogens. Among the strains, 10 showed better antimicrobial activity against MDRS on UTIs. The secondary metabolite of Nocardiopsis sp. GRG1 (KT235640) has showed tremendous antibacterial activity against UTI pathogens compared to other strains. Influence of various growth parameters were used for synthesis of secondary metabolites, such as optimum pH 7, incubation time 5–7 days, temperature (30 °C), salinity (5%), fructose and mannitol as the suitable carbon and nitrogen sources. At 100 μg/ml concentration MIC of Nocardiopsis sp. GRG1 (KT235640) showed highest percentage of inhibition against Proteus mirabilis (85%), and E.coli, Staphylococcus auerues, Psuedomonas aeroginasa, Enterobactor sp and Coagulinase negative staphylococci 78–85% respectively

    © 2018 Elsevier Ltd Copper oxide nanoparticles (CuO NPs) were synthesized biologically using leaf extract of Camilla japonica. The typical UV–visible spectral peak of CuO NPs was observed at a wavelength of ∼290 nm, which confirmed their successful synthesis. From scanning electron microscope (SEM) and transmission electron microscope (TEM) analyses, the synthesized CuO NPs were found to possess spherical shape. Energy dispersive X-ray analyzer (EDX) results revealed that the CuO NPs are almost pure with atomic percentages of 50.92 for Cu and 49.08 for O. Fourier transform infrared (FTIR) confirmed the presence of an absorption peak located at a wavenumber position of ∼480 cm −1 typical for highly pure CuO NPs. TEM images displayed that the particles are relatively uniform in size ∼15–25 nm. The P. aeruginosa and K. pneumonia showed complete resistance against Hexa 077 antibiotic discs.     Based on the recent research, the natural polysaccharide rich biopolymer of chitosan is an important carrier molecule for drug loading. In addition, the plant essential oils were another important material that frequently used to treat various infectious diseases. In addition, the chitosan loaded essential oils were heightened recent years due to the excellent biomedical properties. In this regard, the current study result was concentrated with encapsulation of plant essential oils into the chitosan nanoparticles for inhibit against A549 human lung cancer cells. In result, the unique and uniform size morphology of chitosan was clearly shown in scanning electron microscope (SEM) and transmission electron microscope (TEM). Further, the IC50 dose of the chitosan loaded essential oils at 1000 µg/mL concentration against A549 lung cancer cells was confirmed using cytotoxicity assay and this concentration was very effective than chitosan and essential oils alone. At 1000 µg/mL concentration, the high proliferation efficiency with unclear morphological evidence of A549 lung cancer cells was shown in phase contrast microscope. In addition, the AO/EB stain result was indicated more death cells with belbing formation of the treated cells were observed. At 1000 µg/mL of IC50 concentration of chitosan loaded essential oils was damaged the A549 lung cancer cells with high necrotic cells, chromatin condensation, and decreased cell division. Therefore, all the invitro experiments results were clearly stated that the chitosan loaded essential oils as more efficient alternative option to inhibit the cancer cells
