    Evaluation of total phenols and antibacterial activity of certain drug plants against some bacterial species

    The present paper deals with antimicrobial activity of Glycyrrhiza glabra, Tinospora cordifolia, Tribulus terrestris, Thevetia peruviana, Adhatoda vasica, Vitex negungo, Apium graveolens and, Annona squamosa. It was studied against Bacillus cereus, Staphylococcus aureus, Salmonella typhy B (STB), Shigella dysenteriae using solvent (methanol and petroleum ether) extraction method. Methanol extract of all eight plants showed prominent inhibitory activity against S. dysenteriae and STB while G. glabra, V. negundo, and T. peruviana showed maximum zone of inhibition against four studied bacterial species. These results suggest that methanol extract is more effective than petroleum ether for the antimicrobial activity. Further total phenols were estimated and antibacterial activity of these plants in relation to total phenolic content is discussed

    MOLECULAR DOCKING STUDY ON 1H-(3,4d) PYRAZOLO-PYRIMIDINES AS CYCLIN DEPENDANT KINASE (CDK2) INHIBITORS

    Objective: CDK2 inhibitors are implicated in several carcinomas viz. Carcinoma of lung, bladder, sarcomas and retinoblastoma. Pyrazolopyrimidines, being purine bioisosters inhibit more than one type of kinase. In this study, we are studying some novel derivatives of 1H-pyrazolo [3,4d] pyrimidines not reported earlier. The objective of the present study is an attempt towards design and development of 1H-[3,4-] pyrazolo-pyrimidines as CDK2 inhibitors through rational drug design.Methods: The present study has been done on CDK2 structure, PDB ID, 3WBL, co-crystallized with ligand PDY from RCSB protein data bank. A series of seventeen 1H-Pyrazolo [3,4-d] pyrimidines feasible for synthesis was docked on the said CDK2 receptor using Auto Dock 4 version, 1.5.6. Outputs were exported to discovery studio 3.5 client for visual inspection of the binding modes and interactions of the compounds with amino acid residues in the active sites.Results: The results of docking studies revealed that the present series of 1H-Pyrazolo[3,4-d] pyrimidines is showing significant binding through hydrogen bonding, hydrophobic, pi and Van der waals interactions, similar to the ligand PDY. Some conserved H-bond interactions comparable to bioisosters and compounds presently under human trials were noted. Ki values predicted in silico also suggest that the series will show promising CDK2 inhibitory activity.Conclusion: The series designed and docked can be further developed by synthesis and in vitro and in vivo activity. The receptor inhibitory activity can also be checked by specific receptor assays

    A study of genital pruritus in female patients attending the dermatology OPD at a tertiary care center in South Rajasthan

    Background: Genital pruritus is a common complaint among women, the cause of which could be multitude. Identification of these causes can lead to prompt resolution of pruritus with appropriate therapies. The objective of our study was to determine the clinical profile of genital pruritus and its impact on quality of life amongst all female patients attending Dermatology OPD at our tertiary care centre. Methods: An observational, descriptive, cross-sectional study conducted from June 2020-May 2021on 196 female patients attending the dermatology OPD with primary complaints of genital pruritus. Diagnosis was established by history and clinical examination and confirmed by bed side tests and laboratory investigations as and when required. Results: The mean age of the patients was 36.87±15.24 years with 41.8% in the age group of 18-35 years. In majority (47.4%) of patients, itch was of moderate severity according to the Numerical rating scale. Infectious diseases (45.9%) were the most common cause followed by inflammatory (33.2%), idiopathic (19.4%) and hormonal (1.5%). Vulvovaginal candidiasis was by far the commonest (36.2%). The most common inflammatory dermatoses were lichen simplex chronicus (12.2%) followed by lichen sclerosus atrophicus (10.2%) and contact dermatitis (6.1%). 7 patients with idiopathic itch were diagnosed to have an underlying psychiatric illness. Conclusions: As genital pruritus has multiple causes, proper categorization and diagnostic evaluation would be appropriate to achieve optimal treatment to meet the diverse needs of women who suffer from it
