    In situ data collection and structure refinement from microcapillary protein crystallization

    In situ X-ray data collection has the potential to eliminate the challenging task of mounting and cryocooling often fragile protein crystals, reducing a major bottleneck in the structure determination process. An apparatus used to grow protein crystals in capillaries and to compare the background X-ray scattering of the components, including thin-walled glass capillaries against Teflon, and various fluorocarbon oils against each other, is described. Using thaumatin as a test case at 1.8 angstrom resolution, this study demonstrates that high-resolution electron density maps and refined models can be obtained from in situ diffraction of crystals grown in microcapillaries

    Comparative evaluation of valethamate bromide and hyoscine butyl bromide on cervical dilatation in water buffaloes after detorsion

    After detorsion, incomplete cervical dilatation is major cause of dystocia. The present study aimed to investigate the comparative efficacy of valethamate bromide and hyoscine butyl bromide on cervical dilation after detorsion and their effect on haematological and biochemical parameters of torsion detorted water buffaloes. A total of 18 buffaloes were selected for study which were divided into three groups of six animals each. After detorsion by Sharma’s Modified Schaffer’s method, animals with grade I and grade II cervix were selected for the study. Group I (control group) buffaloes were administered with dexamethasone (40 mg, I/M), cloprostenol (500 μg, I/M) and mifex (450 mL, slow I/V) after successful detorsion. Along with these drugs, in group II and group III, valethamate bromide (100 mg, I/M) and hyoscine butyl bromide (160 mg, I/M) were administered, respectively. The cervical dilation rate (CDR) of each group was calculated and compared. Blood samples were collected three times: before detorsion, after treatment and at parturition. Group III (2.17±0.07 cm/h) had significantly higher CDR followed by group II (1.39±0.21 cm/h), and control group (0.47±0.19 cm/h). Serum calcium levels were also elevated after treatment.     Engineered reversal of drug resistance in cancer cells - metastases suppressor factors as change agents

    Building molecular correlates of drug resistance in cancer and exploiting them for therapeutic intervention remains a pressing clinical need. To identify factors that impact drug resistance herein we built a model that couples inherent cell-based response toward drugs with transcriptomes of resistant/sensitive cells. To test this model, we focused on a group of genes called Metastasis Suppressor Genes (MSGs) that influence aggressiveness and metastatic potential of cancers. Interestingly, modeling of 84 000 drug response transcriptome combinations predicted multiple MSGs to be associated with resistance of different cell types and drugs. As a case study, on inducing MSG levels in a drug resistant breast cancer line resistance to anticancer drugs caerulomycin, camptothecin and topotecan decreased by more than 50–60 %, in both culture conditions and also in tumors generated in mice, in contrast to control un-induced cells. To our knowledge, this is the first demonstration of engineered reversal of drug resistance in cancer cells based on a model that exploits inherent cellular response profiles

    Secondary metabolite induced tolerance to Fusarium oxysporum f.sp. cubense TR4 in banana cv. Grand Naine through in vitro bio-immunization: a prospective research translation from induction to field tolerance

    An innovative tissue culture mediated incorporation of metabolite-based biomolecule (Bio-immune) at in vitro stage itself in banana cv. Grand Naine was developed and validated for the production of Fusarium oxysporum f.sp. cubense TR4 tolerant plantlets. The novel bio-immune formulation developed by us, exhibited a significant antifungal potency against Foc TR4 with a high percent inhibition (100%) at a 2.5% concentration of bio-immune on the 5th, 7th, and 9th DAI. Bio-immune integrated during in vitro shoot proliferation stage in banana cv. Grand Naine recorded significant enhancement in the growth of roots and shoots. Bio-immune (0.5%) fortified media produced 12.67 shoots per clump whereas control registered only 9.67 shoots per clump. Similarly, maximum root numbers (7.67) were observed in bio-immune plants which were significantly higher over control (5.0). The bio-immunized banana transplants recorded a higher survival rate (97.57%) during acclimatization as compared to the control (94.53%). Furthermore, evaluation of the bio-immunized plants in pot experiments revealed that unimmunized plants treated with FocTR4 (TF) exhibited mortality between 60 and 90 days. On the 90th day after planting, a high mean disease severity index (DSI) of 3.45 was observed with unimmunized plantlets while the bio-immunized plants (TFBI) and ICAR-FUSICONT treated plants (TFTR) showed substantially reduced DSI (0.20 and 1.00) compared to FocTR4 treated control (TF). Significant increases in polyphenol oxidase (PPO), peroxidase (POD), β-1,3-glucanase, phenylalanine ammonia-lyase (PAL), chitinase activities, and enhanced phenol contents were recorded in bio-immunized plants compared to unimmunized plants. Field experiments at two different locations in Bihar, India revealed that bunch weight, no. of hands/bunch, and no. of fingers/hand of bio-immune treated plants were significantly higher compared to the control

    The stark malignancy of domestic violence which is also referred as intimate partner violence on the state of women hood can be determined by the prevalence of it in the present society. It bears to push women to the brink and fringes of deemed societal ladders.  The brunt of domestic violence lead to exploitation of women be it in the form of physical or be it mental and occurs chiefly in their matrimonial homes. The detrimental effect of this abuse and exploitation regrettably imprints itself not only on the suffering females but to the entire family including children which in turn causes monetary, health and societal issue and concerns.  What separates domestic violence from other crimes and abuses is that in Domestic violence there is familial interrelatedness and hence the victim is a known of the complainant. In contrast to the modern trends of raising the bar of rights for women the domestic violence is considered as a major road block and its waning as precursor for the society to bring female rights at par. However, on the other spectrum an alarming fact has been observed by the judiciary about the registration of sham cases against the husband or his family members or both for the purposes of harassment or extorting money. To carry off the said object the complaints are registered by overstating the facts, by addition of conjectures, overplaying the financial conditions of the husband or his family and misrepresenting relevant facts and materials. The supreme court opined in Rajesh Sharma v. State of U.P. that the bulk of the cases are filed impetuously and over trifling matters.   To comprehend the say of Indian judiciary on instances of false domestic violence and sham complaints registered by the complainant, secondary data-based research is conducted. Landmark Judgement from the apex court and different High Courts are also analyzed. Endeavor has been made to appreciate the liabilities and legal rights of both the parties in the cases of intimate partner violence which is perpetrated towards one another

We report a 4-year-old child with severe hypertriglyceridemia, pancreatitis, and diabetic ketoacidosis. Hypertriglyceridemia and pancreatitis with diabetic ketoacidosis can be successfully managed with insulin and hydration therapy in children.     Not AvailableThis study reports income enhancement of rural women by their capacity building in Malihabad tehsil of Uttar Pradesh. The study is based on 60 farm women selected from small, marginal and landless households who were provided training for production of improved quality of dried mango slices, commonly known as khatai. The intervention included use of mango peeler, solar dehydrator, preservative and marketing of produce through an NGO. This led to enhancement of income to the tune of 51.0 per cent in small, 50.6 per cent in marginal and 47.6 per cent in landless farm families. The use of mango peeler enhanced slice recovery by 4.5 per cent and reduced time in peeling mango by 26.5 per cent over the traditional method. The use of solar dehydrator enhanced temperature by 8–10 ºC over ambient temperature which helped in fast and uniform drying of slices. The mango slices produced through improved method contains lower were of superior quality with moisture-content (8.4%) and non-enzymatic browning (0.4) and high titrable acidity (21.3%) and total phenol (0.2 g/100g) as compared to the traditional produce.Not Availabl
